Anxiety and anxiety related disorders are increasing at a drastic rate in the past decade, with the NIMH reporting that 31.1% of U.S. adults will experience an anxiety disorder at some point in their lives. Anxiety disorders are currently one of the most predominant mental health conditions worldwide. Increased anxiety is associated with elevated attentional focus to threat also known as attentional bias to threat. Attention Bias Modification (ABM) is a type of computerized training, attempting to reduce attentional focus for threatening stimuli and has been found to successfully reduce symptoms of anxiety. Anxiety disorders are among one of the most debilitating and prevalent mental disorders. Maladaptive anxiety has been associated with enhanced attention bias to threat as well as heightened error-monitoring following an erroneous response. In an effort to reduce an anxious individual’s attention bias to threat, an attention training paradigm known as attention bias modification (ABM) was developed. Increased attentional bias to threat has been identified as a causal mechanism in the development of anxiety. As such, attention bias modification (ABM) was conceived as a treatment option where anxiety is alleviated through a computerized cognitive training regimen that reduces an individual’s attentional bias to threat. Neurotensin is a neuropeptide neurotransmitter that interacts with multiple neurotransmitter systems, including those regulating amygdalar function, via NTS1 and NTS2 receptors. Both receptors are expressed in the amygdala and agonists for NTS1 or NTS2 receptors have exhibited anxiolytic effects in animal models. Systemic adminstration of NTS1 receptor agonist PD149163 was recently shown to reduce footshock conditioned 22-kHz ultrasonic vocalizations in rats, suggesting that PD149163 produced an anxiolytic effect. This study explored the use of optogenetic tools to better understand treatments used for schizophrenia. The “positive” symptoms (e.g., hallucinations, paranoia, etc.) of schizophrenia may come from overexpression of dopamine in mesolimbic dopamine neurons. Positive symptoms can be produced in healthy volunteers treated with amphetamine, a psychostimulant drug and dopamine releaser. Conversely, antipsychotic drugs may reduce positive symptoms by blocking dopamine receptors. This study used optogenetics to explore how drugs that alter neurotransmission, might alter behaviors occurring from activation of mesolimbic dopamine (DA) neurons in the ventral tegmental area (VTA). The atypical antipsychotic drug clozapine remains one of most effective treatments for schizophrenia, given a lack of extrapyramidal side effects, improvements in negative symptoms, cognitive impairment, and in symptoms in treatment-resistant schizophrenia. The adverse effects of clozapine, including agranulocytosis, make finding a safe clozapine-like a drug a goal for drug developers. The drug dis- crimination paradigm is a model of interoceptive stimulus that has been used in an effort to screen experimental drugs for clozapine-like atypical antipsychotic effects. Major depressive disorder (MDD) is the most common mood disorder in the United States and European Union; however, the limitations of clinically available antidepressant drugs have led researchers to pursue novel pharmacological treatments. Clinical studies have reported that monotherapy with the atypical antipsychotic drug quetiapine produces a rapid reduction in depressive symptoms that are apparent following one week of quetiapine treatment, and it is possible that the active metabolite N-Desalkylquetiapine, which structurally resembles an antidepressant drug, produces antidepressant effects. Neuropharmacological evaluations of the neurotensin NTS1 receptor agonist PD149163 are suggestive of antidepressant efficacy, but the effects of a NTS …
