We present a comparison of techniques for estimating atmospheric emissions from fires using Australia's 2009 "Black Saturday" wildfires as a case study. Most of the fires started on Saturday the 7th of February 2009 (a date now known as "Black Saturday") and then spread rapidly, fanned by gale force winds, creating several firestorms and killing 173 people. The fires continued into early March, when rain and cooler conditions allowed the fires to be extinguished. The Kimberley region of northwest Australia contains one of the World's largest collections of rock art characterised by two distinct art forms; the fine featured anthropomorphic figures of the Gwion Gwion or Bradshaw paintings, and broad stroke Wandjina figures. Luminescence dating of mud wasp nests overlying Gwion Gwion paintings has confirmed an age of at least 17,000 yrs B.P. with the most recent dates for these paintings from around the mid-Holocene (5000 to 7000 yrs B.P.). Radiocarbon dating indicates that the Wandjina rock art then emerged around 3800 to 4000 yrs B.P. following a hiatus of at least 1200 yrs. …

Inhibition of GSL (glycosphingolipid) synthesis reduces Aβ (amyloid β-peptide) production in vitro. Previous studies indicate that GCS (glucosylceramide synthase) inhibitors modulate phosphorylation of ERK1/2 (extracellular-signal-regulated kinase 1/2) and that the ERK pathway may regulate some aspects of Aβ production. It is not clear whether there is a causative relationship linking GSL synthesis inhibition, ERK phosphorylation and Aβ production. In the present study, we treated CHO cells (Chinese-hamster ovary cells) and SH-SY5Y neuroblastoma cells, that both constitutively express human wild-type APP (amyloid precursor protein) and process this to produce Aβ, with GSL-modulating agents to explore this relationship. Background: Devil Facial Tumour Disease (DFTD) is a unique clonal cancer that threatens the world's largest carnivorous marsupial, the Tasmanian devil (Sarcophilus harrisii) with extinction. This transmissible cancer is passed between individual devils by cell implantation during social interactions. The tumour arose in a Schwann cell of a single devil over 15 years ago and since then has expanded clonally, without showing signs of replicative senescence; in stark contrast to a somatic cell that displays a finite capacity for replication, known as the "Hayflick limit". Generally, sigmoid curves are used to describe the growth of animals over their lifetime. However, because growth rates often differ over an animal's lifetime a single curve may not accurately capture the growth. Broken-stick models constrained to pass through a common point have been proposed to describe the different growth phases, but these are often unsatisfactory because essentially there are still two functions that describe the lifetime growth. Half a century of research into the physiology and biochemistry of sun–shade acclimation in diverse plants has provided reality checks for contemporary understanding of thylakoid membrane dynamics. This paper reviews recent insights into photosynthetic efficiency and photoprotection from studies of two xanthophyll cycles in old shade leaves from the inner canopy of the tropical trees Inga sapindoides and Persea americana (avocado). The chemical constituents and biological activities of Corydalis crispa (Fumariaceae) were investigated for the first time. The phytochemical study resulted in the isolation of nine known isoquinoline alkaloids: protopine (1), 13-oxoprotopine (2), 13-oxocryptopine (3), stylopine (4), coreximine (5), rheagenine (6), ochrobirine (7), sibiricine (8) and bicuculline (9), with complete NMR data for 2 and 3 provided here for the first time. Crude extracts exhibitedsignificant anti-inflammatory (p < 0.01) activity against TNF-alpha production in LPS activated THP-1 cells. The acetylcholinesterase inhibitory activity of compounds 2, 4 and 7 and the antiplasmodial activity of compound 5 against P. falciparum strains TM4/8.2 and K1CB1 (multidrug resistant strain) are reported here for the first time. Stylopine (4) did not show antimalarial activity against the K1CB1 strain in contrast to a previous report. This study generated a scientific basis for the use of this plant in Bhutanese traditional medicine, either individually or in combination with other medicinal ingredients to treat a broad range of disorders. This study also identified compound 5 as potential new antimalarial lead compound.

Time series of total column abundances of hydrogen chloride (HCl), chlorine nitrate (ClONO2), and hydrogen fluoride (HF) were determined from ground-based Fourier transform infrared (FTIR) spectra recorded at 17 sites belonging to the Network for the Detection of Atmospheric Composition Change (NDACC) and located between 80.05° N and 77.82° S. By providing such a near-global overview on ground-based measurements of the two major stratospheric chlorine reservoir species, HCl and ClONO2, the present study is able to confirm the decrease of the atmospheric inorganic chlorine abundance during the last few years. Solar hydrogen generation sustained for at least several hours is demonstrated using a dye-sensitised nickel(II) oxide photocathode and BiVO4 photoanode connected in a tandem configuration. H2 evolution rate is 120 nmol h−1 (>80% faradaic efficiency) without electrical bias or sacrificial oxidant/reductant, at pH = 7 using visible light (>420 nm).

The rotationally resolved infrared spectrum of the B+-D2 ion-neutral complex is recorded in the D-D stretch vibration region (2805–2875  cm−1) by detecting B+ photofragments. Analysis of the spectrum confirms a T-shaped equilibrium geometry for the B+-D2 complex with a vibrationally averaged intermolecular bond length of 2.247 Å, around 0.02 Å shorter than for the previously characterised B+-H2 complex [V. Dryza, B. L. J. Poad, and E. J. Bieske, J. Am. Chem. Soc. 130, 12986 (2008)10.1021/ja8018302]. The D-D stretch band centre occurs at 2839.76 ± 0.10 cm−1, representing a −153.8  cm−1 shift from the Q1(0) transition of the free D2 molecule. …

Field trips have long been central to geography, but have been subject to assessment of the role of the 'field' in teaching. At the same time, academics face barriers to running field trips. Distance education and enhanced educational access for non-metropolitan students represented such an obstacle at an Australian university. These obstacles were taken as an opportunity to draw on the regional nature of the students and staff to enhance teaching goals, run critically informed field trips by and manage academic workloads. Thirty two new stemofoline analogues were prepared from didehydrostemofoline for studies as AChE inhibitors. C-3 side-chain modified amino, carbamate, triazole and oxazole stemofoline derivatives were prepared. In general the amine derivatives were found to be stronger inhibitors of AChE than their alcohol analogues that we previously reported. Compounds 5 and 26, with small C-3 side chain substituents, were two of the most active inhibitors. Preliminary molecular docking studies suggested that these compounds may inhibit AChE by binding horizontally along the passage of the active-site gorge and block access to acetylcholine.

A Bayesian inversion technique to determine the location and strength of trace gas emissions from a point source in open air is presented. It was tested using atmospheric measurements of N2O and CO2 released at known rates from a source located within an array of eight evenly spaced sampling points on a 20 m radius circle. The analysis requires knowledge of concentration enhancement downwind of the source and the normalized, three-dimensional distribution (shape) of concentration in the dispersion plume. The Yellow River in China carries an extremely large sediment load. River channel-form and lateral shifting in a dynamic, partly meandering and partly braided reach of the lower Yellow River, have been significantly influenced by construction of Sanmenxia Dam in 1960, Liujiaxia Dam in 1968, Longyangxia Dam in 1985 and Xiaolangdi Dam in 1997. Using observations from Huayuankou Station, 128 km downstream of Xiaolangdi Dam, this study examines changes in the river before and after construction of the dams. We compare two conceptually different methods for determining methane column-averaged mixing ratios (XCH4) from Greenhouse Gases Observing Satellite (GOSAT) shortwave infrared (SWIR) measurements. These methods account differently for light scattering by aerosol and cirrus. The proxy method retrieves a CO2 column which, in conjunction with prior knowledge on CO2 acts as a proxy for scattering effects. The physics-based method accounts for scattering by retrieving three effective parameters of a scattering layer. We present a comparison of an atmospheric general circulation model (AGCM)-based chemistry-transport model (ACTM) simulation with total column measurements of CO2, CH4 and N2O from the Total Carbon Column Observing Network (TCCON). The model is able to capture observed trends, seasonal cycles and inter hemispheric gradients at most sampled locations for all three species. The model-observation agreements are best for CO2, because the simulation uses fossil fuel inventories and an inverse model estimate of non-fossil fuel fluxes. Tilted well-aligned carbon micro- and nano-hybrid rods were synthesized on Si at different substrate temperatures and incident angles of carbon source beam using the hot filament physical vapour deposition technique. The morphologic surfaces, chemical compositions and bond structures of the oblique carbon rod-like structures were investigated by scanning electron microscopy, field emission scanning electron microscopy, transmission electron diffraction and Raman scattering spectroscopy. The field emission behaviour of the fabricated samples was also measured.

Evidence of prehistoric dentistry has been limited to a few cases, the most ancient dating back to the Neolithic. Here we report a 6500-year-old human mandible from Slovenia whose left canine crown bears the traces of a filling with beeswax. The use of different analytical techniques, including synchrotron radiation computed micro-tomography (micro-CT), Accelerator Mass Spectrometry (AMS) radiocarbon dating, Infrared (IR) Spectroscopy and Scanning Electron Microscopy (SEM), has shown that the exposed area of dentine resulting from occlusal wear and the upper part of a vertical crack affecting enamel and dentin tissues were filled with beeswax shortly before or after the …

Hermit crabs are ever alert for more suitable shells to inhabit, but what this may mean for coastal shell middens has rarely been considered. Here, the impact of the most landward-based of hermit crab families, the tropical Coenobitidae, upon archaeological shell-bearing deposits is assessed using a case study: the Neolithic Ugaga site from Fiji. At Ugaga, hermit crabs were found to have removed the majority of shells from the midden and had deposited their old, worn shells in return. The potential of multi-elevated-temperature post-IR IRSL (MET-pIRIR) dating [Li, B., Li, S.H., 2011. Luminescence dating of K-feldspar from sediments: a protocol without anomalous fading correction. Quaternary Geochronology 6, 468-479] using polymineral fine grains (FG) (4-11 μm) is tested using loess samples from the Luochuan section in the Chinese Loess Plateau. Nine FG samples with ages within the last glacial-interglacial period are tested using the MET-pIRIR protocol. The MET-pIRIR results for FG are compared with both the coarse grain (63-90 μm) K-feldspar MET-pIRIR dating results and the coarse grain quartz OSL dating results. Spatial connectivity of areas of dry fuels is considered a significant influence on the incidence of large fires. Precipitation patterns can dynamically affect fuel connectivity through controls on the distribution of dry fuels. Spatio-temporal monitoring of precipitation-driven variations in dry fuel connectivity patterns could therefore offer the potential to monitor fire danger. In this paper we present an innovative graph theoretic-based approach to monitor fire danger using remotely sensed patterns of dry fuel connectivity. Captive breeding and release is a tool used by conservation biologists to re-establish populations of endangered or locally extinct species. Reintroduced animals that have been bred in captivity must learn to meet the challenges posed by free living, and to adjust to local environmental conditions, food and water sources. How well reintroduced animals might meet these challenges is uncertain as few longitudinal studies have investigated the physiology of reintroduced animals or the implications of this for successful establishment of new populations. Carbon monoxide (CO) is retrieved daily and globally from space-borne IASI radiance spectra using the Fast Optimal Retrievals on Layers for IASI (FORLI) software developed at the Universit´e Libre de Bruxelles (ULB). "Studies of long-term vegetation changes are critical for enhancing our understanding of successional dynamics in natural ecosystems. Bycomparing forest inventory data from the 1940s against field data from 2005, we document changes in stand structure over 60 years in forests co-dominated by Callitris glaucophylla J. Thompson & L. Johnson, Allocasuarina luehmannii (R. Baker) L. Johnson and Eucalyptus crebra F.Muell., in central Pilliga, New South Wales (NSW), Australia. Over the last decade, genome sequencing and other high-throughput techniques have provided tremendous insight into the molecular processes underlying cellular functions in a wide range of bacterial species. In recent genome-wide association studies, the extracellular chaperone protein, clusterin, has been identified as a newly-discovered risk factor in Alzheimer's disease. We have examined the interactions between human clusterin and the Alzheimer's disease-associated amyloid-β 1-40 peptide (Aβ 1-40), which is prone to aggregate into an ensemble of oligomeric intermediates implicated in both the proliferation of amyloid fibrils and in neuronal toxicity. Using highly sensitive single-molecule fluorescence methods, we have found that Aβ 1-40 forms a heterogeneous distribution of small oligomers (from dimers to 50-mers), all of which interact with clusterin to form long-lived, stable complexes. Mobilization of arsenic from freshwater and estuarine sediments during the transition from oxic to anoxic conditions was investigated using recently developed diffusive sampling techniques. Arsenic speciation and Fe(II) concentrations were measured at high resolution (1−3 mm) with in situ diffusive gradients in thin films (DGT) and diffusive equilibration in thin films (DET) techniques. Water column anoxia induced Fe(II) and As(III) fluxes from the sediment. A correlation between water column Fe(II) and As(III) concentrations was observed in both freshwater (rs = 0.896, p < 0.001) and estuarine (rs = 0.557, p < 0.001) mesocosms. Porewater sampling by DGT and DET techniques confirmed that arsenic mobilization was associated with the reductive dissolution of Fe(III) (hydr)oxides in the suboxic zone of the sediment; a relationship that was visible because of the ability to measure the coincident profiles of these species using combined DGT and DET samplers. The selective measurement of As(III) and total inorganic arsenic by separate DGT samplers indicated that As(III) was the primary species mobilized from the solid phase to the porewater. This measurement approach effectively ruled out substantial As(V) mobilization from the freshwater and estuarine sediments in this experiment. This study demonstrates the capabilities of the DGT and DET techniques for investigating arsenic speciation and mobilization over a range of sediment conditions.
