Background: The physicochemical characterisation of homoeopathic medicines remains a challenge. Raman spectroscopy is a promising tool for identifying structural modifications in the solvent matrix and specific vibrational ``fingerprints'' induced by the potentisation process. Objective: This study aimed to characterise three remedies (Nux vomica, Thuja occidentalis, and Nitricum acidum - 5C potencies) using Raman spectroscopy to identify specific vibrational markers and evaluate the technique's ability to differentiate between different preparations within a complex matrix. Ergonomics is the study of designing and arranging a workspace or product to optimize the “fit” between people and their work, ensuring safety, comfort, and efficiency. The scientific literature indicates that ergonomic perspectives on the workplace are connected to the anthropometrics of societies. This study primarily aims to create a model that integrates multiple predictive variables to estimate the target variable. Multiple linear regression analysis is used to identify how different anthropometric dimensions predict postural ergonomics during prolonged sitting. In liver-directed radiotherapy (RT), liver regions receiving higher doses typically undergo atrophy while contralateral/adjacent lower-dose regions may exhibit compensatory hypertrophy through regeneration of healthy tissue. Optimizing the RT plan to promote regional hypertrophy while minimizing the risk of developing atrophy has the potential to enhance post-RT liver function and long-term survivorship. However, current clinical practice largely relies on global liver dose-volume metrics during RT-planning, which may obscure favorable dose-response correlation and limit actionable guidance for clinicians. In these systems, fluorocarbon oil combined with fluorinated surfactants is the preferred medium for fluid encapsulation, offering exceptional stability and biocompatibility essential for sensitive biological and chemical processes. Purpose: Develop a practical, real-time permanent magnet electron energy spectrometer and evaluate it for beam tuning and quality assurance (QA) of therapeutic electron beams.
Methods: Aim 1: A 0.55 T permanent magnet was coupled to Sun Nuclear SRS MapCHECK® PC boards, providing two interlaced diode arrays. Detector response, Rmeas(zdiode) versus diode position zdiode, was extracted from raw diode array readings, correcting for diode sensitivity and other factors. This study aimed to evaluate the dosimetric impact of Hybrid Arc Palliative Radiation Therapy (HART) versus conventional anterior-posterior/posterior-anterior (APPA) beam arrangements for thoracic spine metastases. The primary objective was to assess differences in conformity, homogeneity, and organ-at-risk (OAR) sparing between the two techniques. A retrospective plan comparison was conducted using anonymized datasets from twenty patients previously treated with HART. Each case was replanned using a standard APPA technique, normalized to match the target coverage of the original HART plan within 0.1%. All plans were prescribed 20 Gy in 5 fractions. Astronauts aboard the International Space Station (ISS) are chronically exposed to a complex space radiation environment composed of galactic cosmic rays (GCR), solar particles, and Earth's trapped radiation. These high-energy particles interact with spacecraft shielding, producing secondary particles that can penetrate inside and deposit energy in sensitive tissues. Although animal studies have been useful in assessing space radiation risks, anatomical and physiological differences limit their applicability to human anatomy especially neuroanatomy. Breast cancer incidence continues to rise worldwide, particularly in low- and middle-income countries, where limitations in resources already constrain access to timely care. Radiotherapy is a cornerstone of breast cancer management, proven to significantly lower both recurrence and mortality. However, a growing shortage of radiation staff worldwide threatens the prompt delivery of these treatments.This thesis proposes an automated solution to address the limited accessibility of radiotherapy planning in breast cancer management by developing an end-to-end automated treatment planning model and evaluating its performance and limitations across diverse patient populations.

In this dissertation, methods are developed and described to allow for the rapid calculation of microdosimetric spectra (specifically, lineal energy) for protons. SuperTrack, a GPU-accelerated tool for calculation of microdosimetric spectra was developed and is capable of computing lineal energy spectra up to 5000x faster than using Geant4 directly. Proton lineal energy spectra generated by SuperTrack are indistinguishable from those generated by Geant4. With SuperTrack, large libraries of lineal energy spectra for monoenergetic protons spanning 0-300 MeV have been developed. Problems: A rediscovered radiotherapy technique, FLASH, involves increasing the dose rate to >40 Gy/s. For a given dose, FLASH is shown to decrease the normal tissue complication probability without affecting the tumor control probability. More preclinical studies are necessary to confirm and enable these desirable properties on humans. Current preclinical investigations have diverse constraints, requiring FLASH apparatuses with flexible parameters and accurate control. However, there are limited FLASH platforms available, the existing ones are costly. Our institution is not in possession of any such infrastructure. Sprint running accelerations require runners to apply surface forces that: support body weight by pushing downward, accelerate the body horizontally by pushing backward, and align the direction of the push with the body’s mass center to maintain balance and posture, which imposes an upper limit on the average forward acceleration force equal to the average gravitational force (1.0 G) acting on the runner. IROC’s mission is to help ensure consistent and comparable, high-quality radiotherapy across clinics that participate in national clinical trials. To obtain this mission, IROC’s phantom program provides a third-party end-to-end check of the clinical workflow of a patient receiving radiotherapy. The goal of the phantom audit is to compare the dose delivered to the dose planned by the treatment system ensuring dose delivery accuracy. The two main sources of primary space radiation, Galactic Cosmic Rays (GCRs) and Solar Particle Events (SPEs), encompass a spectrum of ions, ranging from protons (Z=1) to nickel ions (Z=28), with energies spanning from less than 1 keV to exceeding a few TeV. While SPEs represent sporadic events primarily comprised of a high flux of low- to intermediate-energy protons, GCRs are characterized by continuous low flux of diverse ions with higher energies, peaking in intensity near the solar minimum. Radiation treatment planning is a crucial and time-intensive process in radiation therapy. This planning involves carefully designing a treatment regimen tailored to a patient’s specific condition, including the type, location, and size of the tumor with reference to surrounding healthy tissues. For prostate cancer, this tumor may be either local, locally advanced with extracapsular involvement, or extend into the pelvic lymph node chain. External beam radiation therapy has been shown to be an effective treatment method for tumors and abnormalities of the spine and vertebral region. Treating the spine using a stereotactic body radiation therapy (SBRT) technique can reduce toxicity to the spinal cord. The 6 MV flattening filter free (6FFF) beam model is currently used to plan and calculate dose for SBRT treatment plans, and the treatment plans are delivered using a linear accelerator (LINAC).

Myeloid derived suppressor cells (MDSCs) are key regulators of immune responses and correlate with poor outcomes in hematologic malignancies. Here, we identify that MDSC mitochondrial fitness controls the efficacy of doxorubicin chemotherapy in a preclinical lymphoma model. Mechanistically, we show that triggering STAT3 signaling via β2-adrenergic receptor (β2-AR) activation leads to improved MDSC function through metabolic reprogram- ing, marked by sustained mitochondrial respiration and higher ATP generation which reduces AMPK signaling, altering energy metabolism. Furthermore, induced STAT3 signaling in MDSCs enhances glutamine consumption via the TCA cycle. Background: In the United States (U.S.), falls are the leading cause of injury among adults 65 and over, resulting in 36 million falls yearly (Moreland et al., 2020). According to the Centers for Disease Control and Prevention (CDC, 2023), one in four older adults experiences a fall each year. Falls are the world's second most prominent cause of accidental deaths (World Health Organization [WHO], 2021). Falls are the leading cause of both fatal and non-fatal injuries among older adults (Moreland et al., 2020).

Dysregulated eEF2K expression is implicated in the pathogenesis of many human cancers, including triple-negative breast cancer (TNBC), making it a plausible therapeutic target. However, specific eEF2K inhibitors with potent anti-cancer activity have not been available so far. Targeted protein degradation has emerged as a new strategy for drug discovery. The directivity function of a played musical instrument describes the angular dependence of its acoustic radiation and diffraction about the instrument, musician, and musician’s chair. Directivity influences sound in rehearsal, performance, and recording environments and signals in audio systems. Because high-resolution, spherically comprehensive measurements of played musical instruments have been unavailable in the past, the authors have undertaken research to produce and share such data for studies of musical instruments, simulations of acoustical environments, optimizations of microphone placements, and other applications. 18 million new cancer cases are diagnosed each year. Roughly half of these patients will be treated with radiation therapy, a complex technique that requires an interdisciplinary team of clinical staff and expensive equipment to be delivered safely. Cancer centers in Low- and Middle-Income Countries (LMIC) have an especially difficult time meeting the demands of radiation therapy as the complexity of treatment techniques increase, with only 37% of patients in these regions having access to the care they need. Background
Rectal cancer is a common type of cancer. There is an acute health disparity across the globe where a significant population of the world lack adequate access to radiotherapy treatments which is a part of the standard of care for rectal cancers. Safe radiotherapy treatments require specialized planning expertise and are time-consuming and labor-intensive to produce.

Effective connectivity, based on functional magnetic resonance imaging (fMRI) time series signals, is the quantification of how strongly brain activity in a certain source brain region contributes to brain activity in a target brain region, independent of the contributions of other source regions. Current methods to solve this problem have several limitations. They are either unable to model nonlinear relationships between source and target signals, unable to efficiently quantify time lags in source-target relationships, unable to identify time-varying relationships, or fail to account for variability in the hemodynamic response function that converts neuron activity to a measurable signal. The Imaging and Radiation Oncology Core (IROC) phantoms are used as an end-to-end test of an institution’s radiotherapy processes, and for clinical trial credentialing. Phantoms are treated like patients, and evaluation of the doses received by the thermoluminescent dosimeters (TLDs) inside the phantom, reflects the accuracy with which an institution can image, plan and irradiate a phantom or patient. Recent phantom results show that among the hundreds of various IROC phantoms irradiated annually, 8-17% of institutions fail this test. Although childhood cancer survivors have lengthy life expectancies, they run the risk of experiencing long-term health issues as a result of their treatment. The most frequent non-cancerous cause of morbidity and mortality for these survivors is cardiac disease. Radiation therapy (RT) has been linked in numerous cohort studies to a higher chance of developing a late cardiac disease in these survivors, and this risk rises with higher mean heart doses and increased RT exposure to larger cardiac volumes. Purpose: Treatment planning accuracy for spine stereotactic body radiation therapy (SBRT) varies depending on the dose calculation algorithm utilized in the treatment planning system (TPS). This project compared the end-to-end accuracy between spine SBRT plans calculated in a convolution-superposition based TPS (TPSCS) and Monte Carlo based TPS (TPSMC) with radiochromic film measurements. The hypothesis was that TPSMC would calculate the dose gradient in the critical region between the vertebral body and the spinal cord more accurately than TPSCS.
