Photodynamic therapy (PDT) is a minimally invasive treatment that uses light-activated photosensitizers to selectively destroy diseased tissue through the generation of reactive oxygen species. However, conventional in vitro models often fail to accurately reproduce the pharmacokinetics of photosensitizer accumulation and clearance observed in vivo. This study investigated protoporphyrin IX (PpIX) accumulation following 5-aminolevulinic acid (ALA) exposure under physiologically relevant conditions using two-dimensional (2D) monolayers and three-dimensional (3D) spheroids. By comparing continuous ALA exposure with transient exposure followed by ALA washout, this work provides a more biologically relevant framework for studying PpIX kinetics and optimizing photodynamic therapy.

The overarching motivation of this work is the development of seedless, non-intrusive, laser-based diagnostics for supersonic airflow in wind tunnels. To advance this goal, this dissertation fo-cuses on three-photon excitation in a research-grade argon beam within a tabletop vacuum system, which offers a controlled environment for testing and refining the approach. The chosen excitation scheme drives argon atoms to the 3d[5/2]₃ state using a pulsed Ti:Sapphire laser system, enabling time-of-flight (ToF) measurements on atoms that subsequently undergo a multi-step decay to the metastable 4s[3/2]₂ state. This dissertation presents novel algorithms to improve the mapping capabilities of a 360-degree underwater Pulsed Laser Line Scanner LiDAR (PLLS-360°). Due to its 360° field-of-view (FOV), the PLLS-360° is a compact full-waveform omnidirectional imager suitable for seafloor mapping, underwater asset inspection, object detection, ice-sheet mapping, and construction progress monitoring. The proposed methodology includes an improved waveform fitting technique for saturated waveform recovery, detection array response correction, radiometric corrections, and fusion of LiDAR and sonar bathymetric datasets. A natural inefficiency in conventional treatment of the plasma distribution arises from indiscriminate consideration of the contributions to field structure that are dynamically sourced from kinetic activity in disparate sectors of phase space. We introduce a range of techniques for model design to maximize representation efficiency without breaking key physical invariants indicative of physical reliability. An overview of varied adaptation to stable time-advance methods is given. Low-dimensional representation of a laser-driven plasma requires some creativity in differential structural modification, while direct efficiency is uncovered through the compression and rarefaction of a nonuniform momentum grid. High-power fiber lasers are steadily increasing in maximum power and rapidly approaching theoretical maximum power output from a single fiber aperture. The low thermal conductivity of silica fibers is an inherent limitation to the dissipation of heat from the fiber core, and the buildup of heat within the laser gain media is the principal driver of transverse mode instability within fiber systems. This limitation necessitates the exploration of alternate fiber host materials. Multiphysics Modeling Of Material Response To High-Intensity X-Ray And Laser Pulses: Heating, Ablation, And Plasma Expansion, Youssef Abouhussien

This dissertation presents a computational framework to investigate material response under high-intensity X-ray fluxes produced by an exo-atmospheric nuclear detonation and laser-material irradiations, with a focus on heating, ablation, and plasma expansion phenomena relevant to satellite vulnerability and high-energy-density environments. A hybrid Monte Carlo and Two-Temperature Model (MC-TTM) was developed to simulate X-ray and laser energy deposition and thermal relaxation in metals and semiconductors across a range of X-ray and laser pulse durations from femtoseconds to nanoseconds. Results demonstrate distinct thermal behavior between materials, with ablation thresholds and phase transitions captured in good agreement with experimental data.

The simultaneous generation of different forms of radiation from the same source is highly desirable but technically challenging. We present the generation of D-D fusion neutrons and >MeV-energy x-rays from extreme intensity (∼ 1019 W/cm2 ), high repetition-rate (1 kHz) laser pulses on a thin liquid target. Total neutron fluxes of ∼ 105 neutrons/s and x-ray exposure rates up to ∼ 200 R/hr at 74 cm are measured. Evidence of D-D fusion is verified with a measurement suite including three independent neutron detection systems: an EJ-309 organic scintillator, a 3He proportional counter, and a set of 36 …

Silicon photonic integrated circuits enable the capability of producing on-chip processing of photonic signals, including both emission and detection. Emission materials, however, have been limited for monolithic integration due to high processing temperatures, indirect bandgap materials, and material integration complications. The (Si)GeSn material system has recently attracted attention due to the bandgap transition from the Si and Ge, naturally indirect bandgap, to direct bandgap with sufficient Sn incorporation. This dissertation presents results of an experimental investigation of the time resolved fluorescence from a cold and dense ensemble of 87Rb atoms after the sample is illuminated by a short probe pulse. The goal of the experiment was to investigate cooperative subradiant behavior, characterized by a much longer decay time compared to the natural lifetime, induced in an off resonant probe regime. Such long lived states, if controllable, would provide a means to store quantum information. This research documents steps towards building a novel spectroscopic technique, namely, cavity-enhanced double-resonance (CEDR) spectroscopy, for investigating methane (CH4) and other molecules with significance in astrochemistry. These efforts focus on tackling the complexities of their ro-vibrational energy levels and the inefficiencies in analyzing vibrational spectra, particularly for molecules with high symmetry and strong intramolecular interactions. In the CEDR spectroscopy, the first photon, generated by a continuous-wave optical parametric oscillator (CW-OPO) locked to a Doppler-free saturation absorption line, excites the molecule from its ground level to a selected excited ro-vibrational (rotational-vibrational) level, e.g., that of the asymmetric CH-stretch mode. …

For the development of III-nitride-semiconductor-based monolithic micro-light-emitting diode (LED) displays, Eu,O-codoped GaN (GaN:Eu,O) is a promising material candidate for the red LEDs. The luminescence efficiency of Eu-related emission strongly depends on the local atomic structure of Eu ions. Our previous research has revealed that post-growth thermal annealing is an effective method for reconfiguring luminescent sites, leading to a significant increase in light output. We observed the preferential formation of a site with a peak at similar to 2.004 eV by the annealing process. Master Oscillator Power Amplifiers (MOPA) are laser systems that utilize a seed and pump amplification system to boost the output power of high-quality lower power seeding signals. MOPAs can generate high gain while avoiding many of the nonlinearities that negatively affect resonance-based lasers that are known to feature higher internal intensities. Additionally, MOPAs provide an easy alternative to the construction of novel laser technologies for higher output power as they can be easily combined with existing laser sources to amplify their output power.

This review paper describes the energy-upgraded Continuous Electron Beam Accelerator Facility (CEBAF) accelerator. This superconducting linac has achieved 12 GeV beam energy by adding 11 new high-performance cryomodules containing 88 superconducting cavities that have operated cw at an average accelerating gradient of 20 MV/m. After reviewing the attributes and performance of the previous 6 GeV CEBAF accelerator, we discuss the upgraded CEBAF accelerator system in detail with particular attention paid to the new beam acceleration systems. In addition to doubling the acceleration in each linac, the upgrade included improving the beam recirculation magnets, adding more helium cooling capacity to …

The thermal diffusion and acoustic properties of Nb impacts the thermal management of devices incorporating Nb thin films such as superconducting radiofrequency (SRF) cavities and superconducting high-speed electronic devices. The diffusion and acoustic properties of 200-800 nm thick Nb films deposited on Cu substrates were investigated using time-domain thermoreflectance (TDTR). The films were examined by X-ray diffraction, scanning electron microscopy, and atomic force microscopy. The grain size and thermal diffusivity increase with film thickness. The thermal diffusivity increased from 0.100± 0.002 cm2s-1 to 0.237± 0.002 cm2s-1 with the increase in film thickness from 200 …

Nonlinear optical studies have revealed that surfaces and interfaces possess unique optical properties due to the lack of inversion symmetry. These properties are significantly different from those of the bulk material. Second harmonic generation (SHG), a second-order nonlinear optical phenomenon, has emerged as a surface-specific technique to study these unique properties. The powerful selection rule allows generation of second harmonic light only in the bulk of noncentrosymmetric media and the regions with broken inversion symmetry, including surfaces and interfaces. In this experiment, an intensity autocorrelator is set up in order to measure the pulse duration of a passively mode-locked Titanium-Sapphire laser with a power of 267mW producing femtosecond pulses. Then this measurement, as well as a measurement of the laser’s spectrum, is used to test the Uncertainty Principle. Intensity autocorrelation is a well-established technique for measuring pulse duration, and is among the more intuitive techniques for this purpose, which is why it was selected for this experiment. Laser filamentation is a fascinating phenomenon that occurs when an intense laser beam travels through transparent materials, in particular air. At sufficiently high power (TW in the near IR, GW in the UV), instead of spreading out like a regular laser beam, something remarkable happens: the laser beam becomes tightly focused, creating a thin and intense column of light called a laser filament. The effect of radiation reaction is often negligible in inverse Compton scattering. However, in the nonlinear Compton regime, at high laser fields and high electron beam energies where electron recoil must be properly accounted for, there is experimental data which demonstrates the onset of radiation reaction * . We model the radiation reaction as a series of emissions from individual electrons with decreasing energy. This allows us to use the code we previously developed for simulating single-emission inverse Compton scattering events ** . Monitoring their population is paramount to understand the causes of their decline, as well as to guide and evaluate the efficiency of conservation policies. Monitoring populations of flying insects is generally done using physical traps, but this method requires long and expensive laboratory analysis where each insect must be identified by qualified personnel. This work includes experimental demonstrations of grating angle magnification accelerated optical beam scanners. Diffraction grating scanners governed by the grating equation can have scan speed advantages over the flat mirror bound by Snell's law of reflection. Scan speed enhancement of 750% was achieved with a 635nm laser and 1800 groove/mm diffraction grating configuration thanks to the grating angle magnification. A three-color diffraction grating scanner shows identical results at larger scan angles. Tunable acceleration speed is a feature enabled by the diffraction grating scanner to operate in a high-speed scanning region and/or high-resolution scanning region depending on the demands of the …

In this dissertation, I investigate the possibility of lasing and nonlinear phenomena in completely solid-state transverse Anderson localizing optical fibers (TALOFs). I examine three areas within this range of topics. The research in nonlinear phenomena focuses on four-wave mixing (FWM). FWM is of high interest in TALOFs due to the fact that guided localized modes of the fiber each have different propagation constants, and thus unique possible FWM pairs can be generated from the same input pump beam. I demonstrate the generation of FWM in the TALOF by pumping it with 532 nm light into a localized mode and observing …

In this work, we investigated the evolution in bunch length of beams through the CEBAF injector for low to high charge per bunch. Using the General Particle Tracer (GPT), we have simulated the beams through the beamline of the CEBAF injector and analyzed the beam to get the bunch lengths at the location of chopper. We performed these simulations with the existing injector using a 130 kV gun voltage. Finally, we describe measurements to validate these simulations. The Jefferson Lab KL experiment will run at the Continuous Electron Beam Accelerator Facility with a much lower bunch repetition rate (7.80 or 15.59 MHz) than nominally used (249.5 or 499 MHz). While the proposed average current of 2.5 - 5.0 µA is relatively low compared to the maximum CEBAF current of approximately 180 µA, the corresponding bunch charge is atypically high for CEBAF injector operation. In this work, we investigated the evolution and transmission of low-rep-rate, high-bunch-charge (0.32 to 0.64 pC) beams through the CEBAF injector. An electron beam with high bunch charge and high repetition rate is required for electron cooling of the ion beam to achieve the high luminosity required for the proposed electron-ion colliders. An improved design of the 300 kV DC high voltage photogun at Jefferson Lab was incorporated toward overcoming the beam loss and space charge current limitation experienced in the original design. To reach the bunch charge goal of ~ few nC within 75 ps bunches, the existing DC high voltage photogun electrodes and anode-cathode gap were modified to increase the longitudinal electric field (Ez) at the photocathode. Polarized electrons from GaAs photocathodes have been key to some of the highest-impact results of the Jefferson Lab science program over the past 30 years. During this time, various studies have given insight into improving the operational lifetime of these photocathodes in DC high-voltage photo-guns while using lasers with spatial Gaussian profiles of typically 0.5 mm to 1 mm FWHM, cathode voltages of 100 kV to 130 kV, and a wide range of beam currents up to multiple mA. Si photonics is a fast-developing technology that impacts many applications such as data centers, 5G, Lidar, and biological/chemical sensing. One of the merits of Si photonics is to integrate electronic and photonic components on a single chip to form a complex functional system that features compact, low-cost, high-performance, and reliability. Among all building blocks, the monolithic integration of lasers on Si encountered substantial challenges. Si and Ge, conventional epitaxial material on Si, are incompetent for light emission due to the indirect bandgap. This paper is meant to be an introduction to the nature of plasmas, lasers, laser-plasma interactions, and Laser Wakefield Acceleration (LWFA), with sources given where extra detail may be required.
