Single-Dilution Enzyme-Linked Immunosorbent Assay For Quantification Of Antigen-Specific Salmonid Antibody, Stewart W. Alcorn, Ronald J. Pascho

An enzyme-linked immunosorbent assay (ELISA) was developed on the basis of testing a single dilution of serum to quantify the level of antibody to the p57 protein of Renibacterium salmoninarum in sockeye salmon (Oncorhynchus nerka). The levels of antibody were interpolated from a standard curve constructed by relating the optical densities (OD) produced by several dilutions of a high-titer rainbow trout (O. mykiss) antiserum to the p57 protein. The ELISA OD values produced by as many as 36 test sera on each microplate were compared with the standard curve to calculate the antigen-specific antibody activity. Repeated …

We attempted to discern the contributions of physical habitat, water chemistry, nutrients, and contaminants from historic lead-zinc mining activities on the riffle-dwelling benthic fish community of the Spring River, a mid western warm water stream that originates in Missouri and flows into Kansas and Oklahoma. The Spring River has a fish community that includes the Neosho madtom Noturus placidus, a species federally listed as threatened. Although anthropogenic factors such as contaminants limited populations and densities of fishes, an integrated assessment of natural and anthropogenic factors was necessary to effectively estimate the influence of the latter. Investigating Ground And Surface Water In The Republican River Basin Of Nebraska Using Water Chemistry And Environmental Isotopes, Jennifer J. Schellpeper

Recent legal battles concerning water use initiated an investigation of the stream/aquifer relationships in the Republican River Basin located in southwest Nebraska. Ground and surface water was analyzed for tritium, deuterium, 18O, and major ions. Temperature, pH, conductivity and alkalinity were determined on-site. Samples were collected during 1997 and 1998 from irrigation wells throughout the entire basin. Localized interactions between groundwater and surface water were examined during twenty-four hour pump tests performed on the irrigation wells screened in the alluvium along the river valley and its tributaries. Changes in ideas and in technology can come about as slow incremental modifications, as well as by major paradigm shifts. In the case of the development of the ideas of geologic history and time, I will try to present these changes broadly and then look at how some of these have affected interpretation of Nebraska geology. Changes of view on three fronts were important in the development of geologic history and time concepts. First is the question of the nature of time. Is time cyclic or is it linear? The analysis of PCBs in fish tissues by immunoassay methods was evaluated using fish collected from a US monitoring program, the National Contaminant Biomonitoring Program of the US Department of Interior, Fish and Wildlife Service. Selected composite whole fish samples, which represented widely varying concentrations and sources of PCBs, were extracted and subjected to congener PCB analysis by gas chromatography (GC) and total PCB analysis using an ELISA (ePCBs) calibrated against technical Aroclor 1248. PCB congener patterns in these fishes were different from the patterns found in commercial Aroclors or their combinations as demonstrated by principal component analysis of normalized …

Loess and eolian sand cover vast areas of the western Great Plains of Nebraska, Kansas and Colorado (Fig. 1). In recent studies of Quaternary climate change, there has been a renewed interest in loess and eolian sand. Much of the attention now given to loess stems from new studies of long loess sequences that contain detailed records of Quaternary glacial-interglacial cycles, thought to be a terrestrial equivalent to the foraminiferal oxygen isotope record in deep-sea sediments (Fig. 2). The earth's climate is predicted to change because human activities are altering the chemical composition of the atmosphere through the buildup of greenhouse gases - primarily carbon dioxide, methane, nitrous oxide, and chlorofluorocarbons. The heat-trapping property of these greenhouse gases is undisputed. Although there is uncertainty about exactly how and when the earth's climate will respond to enhanced concentrations of greenhouse gases, observations indicate that detectable changes are under way. There most likely will be increases in temperature and changes in precipitation, soil moisture, and sea level, which could have adverse effects on many ecological systems, as well as on …

The presence of buried soils in Alaskan loess is controversial, and therefore criteria for identifying buried soils in these deposits need to be evaluated. In this paper, morphologic and chemical criteria for identifying buried soils are evaluated by studying modern soils developed mostly in Holocene loess under tundra, boreal forest, and transitional coastal-boreal forest vegetation in different parts of Alaska. Data from modern Alaskan soils that developed under vegetation similar to that of the present indicate that soil morphology, organic-matter concentrations, and P concentrations can be useful diagnostic tools for identifying buried soils. Photomicrographs 143A-143F record fabric variations in quartzite with increasing structural depth in a > 1 km thick, amphibolite-facies, normal-sense shear zone in the East Humboldt metamorphic core complex, Nevada (Figure 143.1). This shear zone and the overlying detachment system unroofed an infrastructure of high-grade, migmatitic gneiss during Oligocene to early Miocene extension (Dallmeyer and others, 1986; Wright and Snoke, 1993; McGrew and Snee, 1994). Thermobarometric constraints from near the base of the mylonitic zone record deformation conditions of 550°- 620°C and 300-400 MPa (Hurlow and others, 1991). The chemistry of the carbonate-free clay-size fraction of Owens Lake sediments supports the use of total organic carbon and magnetic susceptibility as indicators of stadial-interstadial oscillations, Owens Lake records of total organic carbon, magnetic susceptibility, and chemical composition of the carbonate-free, clay-size fraction indicate that Tioga glaciation began ~24,500 and ended by ~13,600 14C yr B.P. Many of the components of glacial rock flour (e.g., TiO2 , MnO, BaO) found in Owens Lake sediments achieved maximum values during the Tioga glaciation when valley glaciers reached their greatest extent. Egg sampling confirmed that Suwannee River Gulf sturgeon Acipenser ozyrinchus desotoi, a subspecies of Atlantic sturgeon A. o. oxyrinchus use the same spawning site at river kilometer (rkm) 215 from the mouth of the river each year. Forty-nine eggs were recorded in 1995, and 368 were recorded in 1996. Spawning began 4-7 d after the March new moon in both years and lasted 10-11 d; in 1996, a second 10-d spawning round began on the April new moon. Developmental synchrony among eggs recovered suggested serval discrete spawning events in both years. Total eggs deposited for three 1996 sampling days …

The northern part of the East Humboldt Range, Nevada, provides a rare opportunity to explore the petrogenetic environment of deep levels in the middle crust during both large-scale Mesozoic contraction and Cenozoic regional extension. On this segment of the field trip, we will explore evidence bearing on the character of the metamorphic and magmatic history of this terrane, and attempt to link these constraints to the rheology and tectonic evolution of the middle crust during the Mesozoic and Cenozoic.

The purpose of this geological excursion is to provide an overview of the multiphase developmental history of the Ruby Mountains and East Humboldt Range, northeastern Nevada. Although these mountain ranges are commonly cited as a classic example of a Cordilleran metamorphic core complex developed through large-magnitude, mid-Tertiary crustal extension, a preceding polyphase Mesozoic contractional history is also well preserved in the ranges. An early phase of this history involved Late Jurassic two-mica granitic magmatism, high-temperature but relatively low-pressure metamorphism, and polyphase deformation in the central Ruby Mountains. The climate of the North Atlantic region underwent a series of abrupt cold/warm oscillations when the ice sheets of the Northern Hemisphere retreated during the last glacial termination (17,700–11,500 years ago). Evidence for these oscillations, which are recorded in European terrestrial sediments as the Oldest Dryas/Bølling/ Older Dryas/Allerød/Younger Dryas vegetational sequence, has been found in Greenland ice cores. The geographical extent of many of these oscillations is not well known, but the last major cold event (the Younger Dryas) seems to have been global in extent. Oxygen-18 (18O) values of sediment from the Wilson Creek Formation, Mono Basin, California, indicate three scales of temporal variation (Dansgaard-Oeschger, Heinrich, and Milankovitch) in the hydrologic balance of Mono Lake between 35,400 and 12,900 14C yr B.P. During this interval, Mono Lake experienced four lowstands each lasting from 1000 to 2000 yr. The youngest low stand, which occurred between 15,500 and 14,000 14C yr B.P., was nearly synchronous with a desiccation of Owens Lake, California. Paleomagnetic secular variation (PSV) data indicate that three of four persistent low stands occurred at the same times as Heinrich events …

Alterations of normal histomorphology of fish tissues, behavior, and thermal tolerance are recognized by environmental toxicologists and fish biologists as powerful tools indicating diverse biochemical and physiological changes. Toxicity of atrazine (2-chloro-4-ethylamino-6-isopropylamino-1,3,5-triazine), terbufos (S-(((1,1-dimethyl-ethyl)thio)methyl)O,O-diethyl phosphorodithioate), and their mixture to red shiner (Cyprinella lutrensis) at 23$\sp\circ$C and 30$\sp\circ$C was investigated in 14-d bioassays after 14-d acclimation. During the bioassays, fish behavior, indications of toxicosis, and external anomalies were observed. After the bioassays, the critical thermal maximum (CTM) was determined. The Cork Street landfill in the City of Kalamazoo is unlined and constructed in glacial drift material. Although water quality in some of the landfill monitoring wells is poor, the degree of the landfill’s influence to groundwater, wetlands and Davis Creek located downgradient of the landfill is unknown. The objective of this study is to evaluate the degree of the landfill’s impact in this region through hydrologic and stable isotope analysis.
