Chemotherapeutic Drug Design: An Efficient Synthesis Of 4-Substituted Alpha-Methylene-Gamma-Lactones, Mark F. Mechelke, Katy Platt, Connor Pribula

Sesquiterpene lactones are plant-derived compounds that have been shown to possess significant activity against inflammation and cancer. Comparative studies of sesquiterpene lactone structure and tumor cytotoxicity indicate that the presence of an α-methylene-γ-lactone moiety is necessary for bioactivity. This observation has led to the hypothesis that simple compounds containing this pharmacophore may also exhibit similar anti-cancer properties. To test this theory, an efficient synthesis of 4-substituted α-methylene-γ-lactone has been developed. The model compound in this study, α-methylene-γ-dodecalactone, has been prepared from commercially available 1-decene in six steps. With the fast paced advancement of modern medicine, cancer treatments have improved greatly over the past few decades; however, the overall survival rate has not improved for head neck squamous cell carcinoma (HNSCC). Traditionally, the general affected population of HNSCC was male over 50-60 years of age, whom have had history of alcohol and tobacco use. Conversely, in the recent decades, HNSCC has exhibited significant rise in younger patients, largely due to the increase in human papillomavirus (HPV) infection among young adults.

Objective: Because partners are an important and unpaid resource in cancer care, understanding how destructive, controlling or interfering partner behaviors influence women's cancer care may be particularly relevant for health care providers seeking to provide cancer care and enhance recovery. Using a new measure of partner interfering behaviors in cancer care (PIB-C), we investigated whether women with a recent cancer diagnosis who additionally endorsed any PIB-C would report (a) more symptoms of depression and stress, and (b) lower Functional Assessment of Cancer Therapy (FACT-G) and lower Functional Assessment of Chronic Illness Therapy-Spiritual Well-being (FACIT-SP) scale scores indicating poorer quality of …

Epithelial ovarian cancer is one of the most common gynecological malignancies and the fifth most frequent cause of cancer death in women, affecting over 22,000 women annually. Nearly 15,500 affected women die from this disease annually, and chemoresistance from the commonly prescribed platinum-based drug, carboplatin, is a major contributor to this mortality rate. Previous studies have identified genes with CpG islands that are methylated and transcriptionally silenced in resistant epithelial ovarian cancer patients. Pyridazines are nitrogen-containing compounds that have been found to show a number of pharmaceutical applications. They exhibit antimicrobial, antifungal, and antibacterial properties to name a few. In particular: 3,4,6-triphenylpyridazine derivatives are known to possess anticancer properties. The purpose of this project is to synthesize tri-substituted pyridazine derivatives using microwave-assisted reactions, and to check its biological applications. Microwave synthesis will help shorten reaction times and produce high, efficient yields. Various catalysts, bases, reaction times, and temperatures will be explored to study the scope of the reactions.

Oxaliplatin is one of the three FDA-approved platinum anticancer drugs and considered a third generation drug, discovered after the first generation drug cisplatin and second generation drug carboplatin. It is known to react with proteins and DNA nucleotides in the body. Reaction with DNA occurs primarily at guanosine residues and secondarily at adenine residues for oxaliplatin and other platinum drugs. Dysregulation in signal transduction pathways can lead to a variety of complex disorders, including cancer. Computational approaches such as network analysis are important tools to understand system dynamics as well as to identify critical components that could be further explored as therapeutic targets. Here, we performed perturbation analysis of a large-scale signal transduction model in extracellular environments that stimulate cell death, growth, motility, and quiescence. Each of the model’s components was perturbed under both loss-of-function and gain-of-function mutations. Using 1,300 simulations under both types of perturbations across various extracellular conditions, we identified the most and least influential components based on …

This study investigates how food companies were affected by the International Agency for Research on Cancer’s report that linked processed meat and red meat consumption to increased risk of cancer. The results indicate that neither fast food companies nor meat producers were greatly affected by the announcement in the days immediately following the release.

Bone morphogenetic protein 2/4 (BMP2/4), members of the transforming growth factor β (Tgf-β) superfamily, play a dichotomous role in the progression of cancers through cell growth suppression and the enhancement of tumorigenesis in a variety of cancers. Cyclin dependent kinase 8 (CDK8), a protein kinase that regulates gene transcription, is involved in a variety of cancers including breast and pancreatic cancers as well as in mediating BMP2 signaling via its canonical Smad1 pathway. Our studies show that BMP4 induced EMT signals primarily though Smad1 and that Yap1, a transcriptional co-activator, is necessary for BMP4 induced EMT. Using a highly specific …

Cancer incidence is associated with exposures to multiple environmental chemicals, and geographic variation in cancer rates suggests the importance of accommodating spatially varying effects in the analysis of environmental chemical mixtures and disease risk. Traditional regression methods are challenged by the complex correlation patterns inherent among co-occurring chemicals, and the applicability of geographically weighted regression models is limited in the setting of environmental chemical risk analysis. In comparison to traditional methods, weighted quantile sum (WQS) regression performs well in the identification of important environmental exposures, but is limited by the assumption that effects are fixed over space. The purpose of this study was to test the feasibility of using an electronic tablet to supplement patient and caregiver education about PICC line maintenance and to compare the percentage of PICC line complications in the intervention group with national percentage rates for patients and caregivers. Newly diagnosed cancer patients who had PICC lines inserted participated in this study. They used an electronic tablet to learn the 10 steps for flushing their PICC lines correctly. They also met with the researcher via FaceTime to review the steps.

Current chemotherapeutics exhibit debilitating side effects as a result of their toxicity to healthy tissues. Reducing these side effects by developing chemotherapeutics with selectivity for cancer cells is an active area of research. Phototherapy is one promising modality for selective treatment, where drug molecules are “turned on” when irradiated with light, reducing damage to healthy tissues by spatially restricting the areas exposed to irradiation. A second approach to improve selectivity is to exploit the differences in cancerous versus healthy cells, such as increased metabolism and/or upregulation of cell surface receptors. A novel complex, [Cu(acetylethTSC)Cl]Cl · 0.25C2H5OH 1 (where acetylethTSC = (E)-N-ethyl-2-[1-(thiazol-2-yl)ethylidene]hydrazinecarbothioamide), was shown to have anti-proliferative activity against various colon and aggressive breast cancer cell lines. In vitro studies showed that complex 1 acted as a poison inhibitor of human topoisomerase IIᶐ which may account for the observed anti-cancer effects.
