In Australia, around 5.7 million hectares of agricultural land are currently affected by dryland salinity or at risk from shallow water tables and this figure is expected to increase over the next 50 years (LWRA, 2001). Most improved grassland species cannot tolerate the combined effects of salt and waterlogging and, therefore, the productivity of sown grasslands in salt-affected areas is low. However, there is potential to overcome the lack of suitably adapted fodder species by introducing new, salt and waterlogging-tolerant species and by diversifying the gene pool of proven species. White clover is an important forage legume for temperate regions, but very little is known about the genetic organisation of its breeding populations. The low amount of variability in the Indian collections of white clover for genetic improvement warrants the introduction of new germplasm and collecting local ecotypes for characterisation, utilisation and conservation. Several molecular techniques have been used for germplasm characterisation, variety identification, marker development and identification, molecular diagnostics, phylogenetic studies and diversity analysis. Fescues (Festuca spp.) are widely occurring temperate grasses with more than 450 species that represent a vast resource for genetic improvement of turfgrass and forage cultivars. Fescues are normally outcrossing species and exhibit many ploidy levels(x=7). Much of the work in classification of Festuca is predominantly based on morphological and cytological features. Difficulties in morphological characterization, which are largely subjected to environmental influences, have resulted in many synonymous species and uncertainties in phylogenetic relationships. DNA fingerprinting is considered a more stable and reliable technique to explore genetic diversity and relationships.

Mongolia wheatgrass (Agroypron mongolicum) is a cross-pollinated, long-lived, cool-season and drought-resistant perennial bunchgrass, which plays an important role in arid and semi-arid grasslands of Inner Mongolia. Collections of A. mongolicum from different areas of Inner Mongolia are valuable sources of useful genes for its breeding. The genetic diversity of 8 accessions of A. mongolicum were examined in this study. A dendrogram was constructed to obtain information on the relationship between cultivated and wild A. mongolicum genotypes, which is basic information to explore the possibility of its use in intra- and inter-specific breeding programs.

White clover (Trifolium repens L.) is a key important temperate pasture legume. Due to the obligate outbreeding nature of white clover, individual genotypes within cultivars are highly genetically heterogeneous. Genetic diversity has been assessed within and between 16 elite cultivars derived from Europe, North and South America, New Zealand and Australia.

Zoysiagrass consists of a number of interfertile species, some of which are important grasses for turfgrass and grazing pasture in Japan. Recently, we developed simple sequence repeats (SSRs) markers from Zoysia japonica “Asagake” genomic DNA by enriched genomic library method (Yamamoto et al., 2002). Here we identify genetic diversity in 38 ecotypes of zoysiagrass (Z. matrella and Z. tenuifolia) from a group of southwest islands of Japan based on morphological characteristics and SSR markers.

Little or no genotypic information is available for many forage grass populations. The degree of genetic similarity within and among populations greatly influences the choice of breeding strategies and germplasm for developing improved cultivars. Molecular markers have proven effective in classifying genetic diversity of a number of perennial grasses (e.g. Fu et al., 2004; Kubik et al., 2001). We present here an overview of our efforts to integrate molecular diversity data into our breeding program.

The genus Elymus is the largest genus in the tribe Triticeae with about 150 species distributed in most temperate regions of the world (Dewey, 1984). The genetic diversity of Elymus spp. from alpine regions is very important for improving resistance to adverse condition. The goals of this study were to investigate micro- satellite and enzyme polymorphism and population structure of different regions and Elymus spp. in China.

The common fungal endophytes (Neotyphodium species) of temperate pasture grasses are associated with improved tolerance to water and nutrient stress and resistance to insect pests, but are also the causal agents of animal toxicoses. Considerable variation exists among grass-endophyte associations for these beneficial and detrimental agronomic traits. The extent to which this variation may be attributed to the endophyte genotype, the host genotype or environmental interactions is currently unknown. Expressed sequence tag (EST) databases have been growing exponentially. The simple sequence repeat (SSR) has become one of the most useful molecular marker systems in plant breeding and is widely used in cultivar fingerprinting, genetic diversity assessment, molecular mapping and marker-assisted selection. Crown rust (Puccinia coronata f.sp lolii) is the most important fungal pathogen of perennial ryegrass (L.perenne L.). The physiological effects associated with infection include reduction of water soluble carbohydrate (WSC) reserves, causing decreased dry matter yield, digestibility and palatability for herbivores reared for meat, milk and wool production. Phenotypic variability of rust-infection in perennial ryegrass is likely to be due to environmental effects, as well as the interaction of defence and resistance genes in the grass and virulence genes in the pathogen. Lablab (Lablab purpureus L.) is an important annual multi-purpose legume used as a vegetable for human consumption, as forage for livestock, and as green manure and a cover crop to improve soil fertility. It has a high feed value with good digestibility and high crude protein content. The International Livestock Research Institute (ILRI) forage genebank holds a diverse set of 340 lablab accessions collected from different regions of the world. This work aimed to determine the genetic diversity of Colletotrichum gloeosporioides infecting Stylosanthes spp. in Brazil, China and India. A total of 132 isolate originating from S. seabrana, S. macrocephala, S. capitata, S. scabra, and S. guianensis were used. Four major genetic groups were identified from an analysis of genetic diversity using selection-neutral DNA markers. Group 1 contained 20 isolates and this may represent a genotype that migrated from the center of diversity in Brazil and Colombia to Australia, Thailand and India.
