Understanding the dynamic interplay between fire severity, topography, and tree mortality, is crucial for predicting future forest dynamics and enhancing resilience against climate change-induced wildfire regimes. This thesis develops a multi-sensor approach for automated estimation of tree mortality, then applies it to examine trends in tree mortality over a six-year period across a fire affected study site in the Trinity River basin in Northern California. The Random Forest model uses publicly available USGS 3D Elevation Program Lidar (3DEP) and NAIP imagery as inputs and is likely to be easily adaptable to other landscapes. Ungrading is a pedagogical approach in which no grades are given on any assignments. Instead, students are provided with many opportunities to submit work and gain feedback. The goal is to shift student focus from achieving a grade to growth as a learner and a person. As instructors, our ungrading approach utilized personalized learning plans, checkpoint reflections, and student-professor learning conferences to put agency in the hands of our students. We employed this method in upper-level biology and computer science courses and provide critical reflections here regarding our experiences and the connections between this approach and feminist STEM pedagogy tenets. …

No discussion on equity/inequity makes sense without bringing power into that discussion. As instructors we need to ask questions such as “who decides and controls what knowledge is”, “whose identities are empowered and whose are erased”, “who has access and opportunity and who doesn’t”. Traditional teaching in STEM, including mathematics, assumes knowledge is objective, transmittable, repeatable to everyone. When educators follow a traditional curriculum, just like their teachers before them, they do so thinking their methods ensure equality and objectivity. Wildfires have increased in frequency and severity over the past few decades due to the increased concertation of CO2 emissions from anthropogenic influence. Soil carbon (C) sequestration has been identified as a climate change mitigation strategy; however, the influx of large-scale wildfires has accelerated landscape processes such as erosion, reducing soil aggradation, and soil C and nitrogen (N) protection. Climate-appropriate agroforestry can provide low-input food security and ecosystem services for local Californian Mediterranean climates, while conserving natural resources (e.g., water, nitrogen, etc.). This project showcases a variety of agroforestry methods for five common plant communities of California and other analogous Mediterranean climates at the CAFES Experimental Farm on the campus of Cal Poly San Luis Obispo. Plant community species and their ethnobotanical uses were analyzed to mimic and incorporate aspects of native flora. All microscope imaging is largely affected by inherent resolution limitations because of out-of-focus light and diffraction effects. The traditional approach to restoring the image resolution is to use a deconvolution algorithm to “invert” the effect of convolving the volume with the point spread function. However, these algorithms fall short in several areas such as noise amplification and stopping criterion. This project examines the development of a smart boat which could serve as a possible marine research apparatus. The smart boat consists of a miniature vessel containing a low-cost microcontroller to live stream a camera feed, GPS telemetry, and compass data through its own WiFi access point. The smart boat also has the potential for autonomous navigation. My project captivated the interest of several members of California Polytechnic State University, San Luis Obispo’s (Cal Poly SLO) Marine Science Department faculty, who proposed a variety of fascinating and valuable smart boat applications.

This paper examines vineyard perceptions and adoption of climate change adaptation and water conservation measures in the Paso Robles American Viticultural Area (AVA). A survey was distributed to all 220 vineyards and vineyard management companies that operate in the AVA, with a 53.64% response rate. The objective of the survey was to determine vineyard manager and owner attitudes towards climate change and adaptation, as well as their perceptions of how these threats would impact their operation. Microorganisms are key players in the ecosystem functioning. In this thesis, we developed a framework to preprocess raw microbiome data, build a correlation network, and analyze co-occurrence patterns between microbes. We then applied this framework to a marine microbiome dataset. The dataset used in this study comes from a year-long time-series to characterize the microbial communities in our coastal waters off the Cal Poly Pier. In analyzing this dataset, we were able to observe and confirm previously discovered patterns of interactions and generate hypotheses about new patterns. Temperature variability in the nearshore coastal ocean influences various biological processes and can drive changes in biodiversity and habitat range. Despite recent progress, there are still significant gaps in the understanding of drivers of temperature variability in upwelling bays, particularly at higher frequencies. In this study, we analyzed a decade of nearshore temperature measurements both inside and outside a small coastal embayment located in central California [San Luis Obispo (SLO) bay], as well as temperature data from satellites, to characterize rapid warming events. Dissolved oxygen (DO) is an important biogeochemical factor that strongly influences nearshore coastal ecosystems. Low DO (hypoxic) events can cause physiological stressful environments for ecological and economically important species, potentially leading to mass mortalities. In order to better assess drivers of coastal hypoxia, we collected data from monthly cruises on the inner shelf and nearshore moorings inside and outside a small coastal embayment (San Luis Obispo Bay on the Central California Coast) across the full upwelling season (March to August). The endemic eelgrass (Zostera marina) community of Morro Bay Estuary, located on the central coast of California, has experienced an estimated decline of 95% in occupied area (reduction of 344 acres to 20 acres) from 2008 to 2017 for reasons that are not yet definitively clear. An estimated one-tenth of the planet’s land area experiences artificial light at night — and that rises to 23% if skyglow is included. Nanomaterials such as graphene oxide and carbon nanotubes, have demonstrated excellent properties for membrane desalination, including decrease of maintenance, increase of flux rate, simple solution casting, and impressive chemical inertness. Here, two projects are studied to investigate nanocarbon based membrane desalination. The first project is to prepare hybrid membranes with amyloid fibrils intercalated with graphene oxide sheets. The addition of protein amyloid fibrils expands the interlayer spacing between graphene oxide nanosheets and introduces additional functional groups in the diffusion pathways, resulting in increase of flux rate and rejection rate for the organic dyes. This thesis demonstrates efficacy of Octenyl Succinic Anhydride (OSA), as a biofilm sanitizer. Biofilms allow bacteria to adhere to solid surfaces with the use of excreted polymeric compounds. For example, surfaces found in food production or processing facilities such as the interior of a raw milk holding tank, are some of the most susceptible to biofilm contamination. When present, biofilms can cause a variety of negative effects, which include; reduction of product shelf life, corrosion, and outbreaks of foodborne illnesses. Variations in the abundance and composition of phytoplankton greatly impact ecosystem structure and function. Within the California Current System (CCS), phytoplankton community structure is tightly coupled to seasonal variability in wind-driven coastal upwelling, a process that drives changes in coastal water temperatures and nutrient concentrations. Based on approximately a decade (2008-2018) of weekly phytoplankton measurements, this study provides the first characterization of the seasonal and interannual variability of phytoplankton abundance and composition in San Luis Obispo (SLO) Bay, an understudied region within the CCS. Commercial carrot processors produce up to 175,000 tons of carrot waste annually. Carrot Mash (CM) is the term referring to the waste by-product of peeled baby carrot processing. Transportation of carrot processing waste is expensive due to its high- water content (approx. 83-95%). The analysis of the spatiotemporal variability of wind power remains limited during the planning stage of an offshore wind farm. This study provides a framework to investigate how offshore wind power varies along the Central California Coast over diurnal and seasonal time scales, which is critical for reliability and functionality of the grid system. We find that offshore wind power in this region peaks during evening hours across all seasons and maximizes in spring and summer. Estuaries are dynamic environments that are strongly affected by natural variability, as well as direct and indirect anthropogenic impacts. A better understanding of the drivers of carbon fluxes and biogeochemical variability in estuarine systems is needed, particularly with the increasing threat of ocean acidification. Morro Bay in Central California is a small nationally protected estuary, with seasonally low freshwater inputs. Since 2007, the bay has experienced a significant loss of native seagrass, Zostera marina, which is an important component of the marine ecosystem. The cost of transporting and rendering every dead animal is inhibitory to the general function of the agricultural operations and their thin budget. Microfluidic paper-based analytical devices (microPADs) are emerging as cost-effective and portable platforms for point-of-care assays. A fundamental limitation of microPAD fabrication is the imprecise nature of most methods for patterning paper. The present work demonstrates that paper patterned via wax printing can be miniaturized by treating it with periodate to produce higher-resolution, high-fidelity microPADs. The optimal miniaturization parameters were determined by immersing microPADs in various concentrations of aqueous sodium periodate (NaIO4) for varying lengths of time. Cell-free protein synthesis (CFPS) has emerged as an enabling biotechnology for research and biomanufacturing as it allows for the production of protein without the need for a living cell. Applications of CFPS include the construction of libraries for functional genomics and structural biology, the production of personalized medicine, and the expression of virus-like particles. The purpose of this research is to introduce a new data analysis method called Scale Invariant Geometric Data Analysis (SIGDA). SIGDA has been shown to be more informative than more common data analysis methods, such as Principal Component Analysis (PCA). SIGDA is used to visualize complex data sets in a way that accurately preserves data patterns and behavior. River restoration projects are being installed worldwide to rehabilitate degraded river habitat. Many of these projects focus on stream habitat improvement (SHI), and an estimated 60%of the 37,000 projects listed in the National River Restoration Science Synthesis Program focus on SHI for salmon and trout species. These projects frequently lack a sufficient monitoring program or account for the environmental costs associated with SHI. The present study used life cycle assessment (LCA) techniques and topographic effectiveness monitoring to quantify environmental costs on the basis of geomorphic change.
