The transmission of vector-borne diseases involves complex interactions between vectors and their host species. These complex host-parasite interactions can be difficult to study with traditional, field-based methods. My dissertation aims to use a population genomics approach to elucidate transmission pathways of plague among prairie dog colonies. Plague is a flea-borne, zoonotic disease caused by the bacterium Yersinia pestis. It is infamous for causing the Black Death (1347-1353), one of the most devastating pandemics in human history. Gait analysis presents the challenge of detecting a periodic waveform in the presence of time varying frequency, amplitude, DC offset, and waveform shape, with acquisition gaps from partial occlusions. The combination of all of these components presents a formidable challenge. The Extended Kalman Filter for this system model has six states, which makes it weakly identifiable within the standard Extended Kalman Filter network. Gamma-aminobutyric acid type A receptors (GABAAR) are transmembrane pentameric ligand-gated chloride ion channels that respond to GABA, the major inhibitory neurotransmitter in the central nervous systems (CNS). The benzodiazepines (BZDs) bind at the extracellular interface of the α+γ2-subunits of GABAAR. The binding of ligands at different subunits of GABAA receptors specifically at α1-6β2/3 γ2 ion channels, can affect a wide variety of brain functions. The early Cambrian represents an important transition in the evolution of life, perhaps most vividly exemplified by reef ecosystems as they changed from microbial-supported to metazoan-supported framework reefs. Microbial reefs were initially composed of Renalcis- and Epiphyton-group calcifying microbes. Subsequent reefs began to incorporate archaeocyathan sponges within this framework. This represents a shift in the source of carbonate production, which can be quantified using thin section point counts. In archaeocyathan reefs from the western USA, carbonate contribution from metazoan framework builders increased from zero to 29.7%. Similar reefs from Mongolia increased from zero to 5.0%. Calcitroic acid (CTA) was isolated and characterized more than four decades ago.5 Radiolabeled calcitriol (1,25-dihydroxyvitamin D3) was used at that time to enable the identification of radioactive CTA formed in vivo, which was subsequently extracted and characterized by derivatization.7 CTA was found to be predominantly formed in the liver and secreted into the gut through the bile duct via enterohepatic circulation, leading to fairly high concentrations of this metabolite of vitamin D in the intestine.8 However, assuming it was only a catabolic product of calcitriol, it was ignored thereafter.

This thesis investigates the use of science by media to shape discussions about policy in society. Specifically, it investigates how one influential trade publication, Ethanol Producer Magazine, used science in its construction of pro-ethanol production stories. This study is a multi-method analysis of Ethanol Producer Magazine’s feature articles. To determine how sources and writers use science to frame ethanol production during policy changes in the industry, 36 feature stories from four issues in 2009 and 2010 of Ethanol Producer Magazine were analyzed. Cascades frogs (Rana cascadae) in the southern Cascades Range of California have been declining over the last 30 years, primarily due to the fungal pathogen, Batrachochytrium dendrobatidis (Bd). In the Lassen Region of the southern Cascades, at least six of the eleven remaining localities face extirpation within 50 years. These small and isolated populations are prone to negative genetic effects including reduced diversity and increased inbreeding which could potentially exacerbate declines. The field of molecular recognition focuses on the selective and reversible binding of small guest molecules to larger host molecules. This dissertation describes synthesis of small molecules as binding guests for enzymatic substrates as well as molecules as host chemosensors to detect and identify metal ions. Two approaches to new antibiotic drugs have been explored, and an array of sensors for the quantitation of aqueous metals is being commercialized.

Within the last decade, X-ray Free Electron Lasers (XFELs) have emerged across the world. These XFELs produce X-ray pulses with a duration on the order of femtoseconds, each of which contains 1012 photons. Before the XFEL, the brightest X-ray sources were 3rd generation synchrotrons. While these facilities are still very important for many experiments, XFELs allow for time-resolved experiments with femtosecond time resolution and mixing experiments that are not possible at the synchrotron. Enzymatic processes have promising prospects for medicine which use proteins as drug targets and enhance our understanding of these important biomolecules.

Verdicchio is a white grape varietal notably grown in the Marche wine region of east-central Italy within two Denominazione Origine Controllata (DOCs); Castelli di Jesi and Matelica, located less than 50 km apart. Jesi lies in the Apennine foothills near the coast of the Adriatic Sea and produces a smooth wine noted with apple-pear undertones and crisp, clean acidity. Conversely, Matelica is located in a synclinal valley of a fold-and-thrust belt and produces stronger wine with penetrating floral aromas and bold acidity. A Hemimysis Driven Novel Ecosystem At A Modified Boulder Breakwall, Eric John Geisthardt

The US Army Corps of Engineers (USACE) is mandated to maintain and repair aging breakwall structures in all commercial ports on the Great Lakes. In May of 2014, the construction of Milwaukee Harbor USACE “green” breakwall (GBW) reconciliation created complex rocky aquatic habitat by depositing cobble-sized stone as a veneer over standard 6-10 ton boulders, thus creating “control” (boulder) and “treatment” (cobble) habitats. The breakwall is home to a prolific population of Hemimysis anomala, the introduced Ponto-Caspian mysid, which is significantly more abundant on cobble versus boulders (p<0.05, using a novel trap for Hemimysis). Fish and forage communities were sampled in 2015 and 2016 using a combination of experimental and micromesh gill nets, night scuba diving surveys, and a novel Hemimysis trap. This nearshore lithophilic mysid appears to provide a significant new seasonal food resource in the Milwaukee Harbor for pelagic prey fishes during inshore spawning migrations and upwelling events. Alewife (Alosa pseudoharengus) and rainbow smelt (Osmerus mordax) fed heavily on Hemimysis with some individuals consuming hundreds of mysids. Night scuba diving surveys and gill netting confirmed that rainbow smelt preferred to forage on the cobble section (p<0.05), and also consumed more Hemimysis there than they did at the control breakwall site (p<0.05). Hemimysis were also the primary food item consumed by nearshore game fishes such as YOY yellow perch (Perca flavescens), YOY largemouth bass (Micropterus salmoides), and juvenile rock bass (Ambloplites rupestris) caught at the breakwall. This study provides the first documented evidence that where abundant in the Laurentian Great Lakes, Hemimysis do have the ability to significantly impact local food webs and drive the feeding ecology of both pelagic transient and nearshore resident fishes.

Interaction With Nitric Oxide Of The Nitrosomonas Europaea Tetraheme Protein Cytochrome C554, And Two Of Its Variants, In Increasingly Reducing Environments, Jennifer M. Mcgarry

A re-investigation of the interaction with NO of the small tetraheme protein cytochrome c554 (C554) from Nitrosomonas europaea has shown that the 5-coordinate heme II of the 2-electron or 4-electron reduced protein will nitrosylate reversibly. The nitrosylation process was found to be first order in C554, first-order in NO, and second-order overall. The rate constant for NO binding to the heme was determined to be 3000 ± 140 M-1s-1, while the rate constant for dissociation was 0.034 ± 0.009 s-1; the degree of protein reduction does not appear to significantly influence the nitrosylation rate. The following dissertation focuses upon the organization of Pleistocene / Holocene period lithic technology in Wisconsin circa 10,000 – 10,500 years before present. Lithic debitage and flaked stone tools from the Plainview/Agate Basin components of the Heyrman I site (47DR381), the Dalles site (47IA374), and the Kelly North Tract site at Carcajou Point (47JE02) comprise the data set. These Wisconsin sites are located within a post glacial Great Lakes dune environment, an inland drainage/riverine environment, and an inland wetland/lacustrine environment. Survivability of cancer strongly depends on the stage of cancer. In most previous works, machine learning survivability prediction models for a particular cancer, were trained and evaluated together on all stages of the cancer. In this work, we trained and evaluated survivability prediction models for five major cancers, together on all stages and separately for every stage. We named these models joint and stage-specific models respectively. The problem of determining the structure of a biomolecule, when all the evidence from experiment consists of individual diffraction patterns from random particle orientations, is the central theoretical problem with an XFEL. One of the methods proposed is a calculation over all measured diffraction patterns of the average angular correlations between pairs of points on the diffraction patterns. The point of use biosand filter (BSF) is used globally as a drinking water treatment solution. In this research, point of use BSFs were inoculated with active biosand from the Linnwood Drinking Water Treatment plant slow sand filter beds (Milwaukee, Wisconsin) and with sands collected from point of use filters operating in the Dominican Republic. These filters were maintained with varying source waters (surface water, groundwater, or tap water to simulate chlorination encountered in the field). For example, there is now software for recording and tracking one's exercise activity or blood pressure. Even though there are already many of these services, the mobile health field still presents many opportunities for new research.
