Engineered nanomaterials (ENMs) have become ubiquitous in consumer products and industrial applications, and consequently the environment. Much of the environmentally released ENMs are expected to enter terrestrial ecosystems via land application of nano-enriched biosolids to agricultural fields. Among the organisms most likely to encounter nano-enriched biosolids are the key soil bacteria known as plant growth promoting rhizobacteria (PGPR). I reviewed what is known concerning the toxicological effects of ENMs to PGPR and observed the need for high-throughput methods to evaluate lethal and sublethal toxic responses of aerobic microbes. We developed a new technology to study protein-protein interaction in mammalian cells. This technology is based high resolution of Nucleic Magnetic Resonance (NMR) spectroscopy. Using this technology we studied interaction between the receptor for advanced glycation endproducts (RAGE). RAGE- is a multiligand receptor of immunoglobulin receptor family that is activated by a multitude of ligands. Activation of RAGE results in signal transduction that leads to the inflammatory response implicated in the complications of Diabetes. Land use and land cover (LULC) of Table Rock Lake (TRL) region has changed over the last half century after the construction of Table Rock Dam in 1959. This study uses one meter spatial resolution imagery to classify and detect the change of LULC of three typical waterside TRL regions. The main objectives are to provide an efficient and reliable classification workflow for regional level NAIP aerial imagery and identify the dynamic patterns for study areas. Recreational aviation on public lands may negatively impact wildlife. However, land-use decisions need to balance user need with wildlife impact. We know very little about 1) how back country airstrip use affects local wildlife, or 2) attitudes and perceptions of recreational pilots toward possible management actions. For my Master’s research, I investigated how aircraft activity influenced physiological measures of stress in white-tailed deer, while also modeling how psychometrics such as wildlife attitudes and place attachment predict the willingness of recreational pilots engage in impact-mitigating behaviors. Synthesis Of Satellite Microwave Observations For Monitoring Global Land-Atmosphere Co2 Exchange, Lucas Alan Jones

This dissertation describes the estimation, error quantification, and incorporation of land surface information from microwave satellite remote sensing for modeling global ecosystem land-atmosphere net CO2 exchange. Retrieval algorithms were developed for estimating soil moisture, surface water, surface temperature, and vegetation phenology from microwave imagery timeseries. Soil moisture retrievals were merged with model-based soil moisture estimates and incorporated into a light-use efficiency model for vegetation productivity coupled to a soil decomposition model. Results, including state and uncertainty estimates, were evaluated with a global eddy covariance flux tower network and other independent global model- and remote-sensing based products.

Colorimetric methods combined with color-changing chemical probes are widely used as simple yet effective tools for identifying and quantifying a wide variety of molecules in solution. For nucleic acids (DNA and RNA), perhaps the most commonly used colorimetric probe is potassium permanganate, which can be used to identify single-stranded pyrimidines (thymine and cytosine) in polymers. Unfortunately, permanganate is not an effective probe for identifying purines (adenine and guanine), especially in the presence of the more reactive pyrimidines. Therefore, robust methods for discriminating between the purines remain elusive, thereby creating a barrier toward developing more complex colorimetric applications. In this dissertation, …

The impact of increased sediment exposure on coastal marine ecosystems is one of the most important contemporary environmental issues. Sediment exposure is defined as the concentrations of sediment particles suspended in the water column and the amount of sediment depositing on the seabed. In addition to natural events, such as land erosion, rainfall, and tidal currents, anthropogenic activities such as land use, road building, logging, mining, port maintenance and dredging, contribute to the discharge of a great amount of sediment in the water column. How genetic variation is distributed across space (genetic structure) and what factors influence the spatial genetic structuring is one of the primary questions in population genetics. The interaction between species biology (e.g. life-history traits) and physical processes operating in the seascape over time, including palaeo-historical events (e.g. sea level fluctuations) and contemporary processes (e.g. ocean currents), have been predicted to influence the extent of gene flow and the spatial genetic structuring in marine organisms. The Red List of marine bony fishes of the Eastern Central Atlantic (ECA) is a review of the conservation status of all native marine bony fishes in ECA according to the global Categories and Criteria of the IUCN Red List of Threatened Species. It identifies those species that are threatened with extinction at the global level and occur within the ECA region. Marine protected areas are considered important tools for protecting marine biodiversity, and animal tracking is a key way to determine if boundaries are effectively placed for protection of key marine species, including seabirds. We tracked chick-rearing brown noddies (Anous stolidus) from the Dry Tortugas National Park in Florida USA in 2016 using 1.8 g Nanofix GPS tags (n = 10), making this the first time this species has ever been tracked. Thrombin is the key protease that regulates hemostasis; the delicate balance between procoagulation and anticoagulation of blood. In clotting disorders, like deep vein thrombosis or pulmonary embolism, procoagulation is up-regulated, but propagation of clotting can be inhibited with drugs targeting the proteases involved, like thrombin. Such drugs however, have serious side effects (e.g., excessive bleeding) and some require monitoring during the course of treatment. The reason for these side effects is the mechanism by which the drugs’ act. The two major mechanisms are direct orthosteric and indirect allosteric inhibition, which will completely abolish the protease’s activity. Herein we sought an …

There is limited information on forage sorghum and corn silage yield response to full and deficit irrigation in Kansas. The objective of this study was to generate information on forage sorghum (brown mid-rib hybrids (BMR and non-BMR)) and corn silage yield response to different levels of irrigation as influenced by irrigation capacity in southwest Kansas. Preliminary results indicate the effect of irrigation capacity on forage yield was significant (P = 0.0009) in 2014 but not 2015, probably due to high growing season rainfall received in 2015. Corn silage produced significantly (p<0.05) higher biomass at all irrigation capacities compared to forage sorghum hybrids in 2015. BMR forage sorghum produced significantly lower biomass compared to non-BMR hybrid in both 2014 and 2015 (P<0.05). The highest amounts of forage produced for corn silage, BMR, and non-BMR forage sorghum were 24.6, 17.4, and 21.1 tons/a adjusted to 65%, moisture respectively. Water productivity ranged from 1.0 to 1.4 dry matter tons/a/in. More research is needed under normal and dry years to quantify forage sorghum and corn silage yield and forage quality response to full and deficit irrigation.

Mobile Drip Irrigation (MDI) involves attaching driplines to center pivot drops. MDI has potential to eliminate water losses due to spray droplet evaporation, water evaporation from the canopy, and wind drift. MDI also may reduce soil water evaporation due to limited surface wetting. A study was conducted with the following objectives: 1) compare soil water evaporation under MDI and in-canopy spray nozzles; 2) evaluate soil water redistribution under MDI at 60 inch dripline lateral spacing; 3) compare corn grain yield, water productivity, and irrigation water use efficiency; and 4) compare end-of-season profile soil water under MDI and in-canopy spray at …
