It is well understood that lactating dairy cows require metabolizable protein (MP) for milk yield and milk protein synthesis. Diets with conventional feed sources like corn and soybean meal (SBM) may not sufficiently meet all MP and amino acid (AA) requirements so supplementation with more rumen undegradable protein (RUP) rich feeds or rumen-protected AA can be fed to support animal performance. The growth trajectory of biofuels has resulted in dramatic increases in the production of feeds like heat treated soybean meal (HTSBM). Snakes in human care often lack diversity in the diet. Rodents account for the majority of meals, while wild snakes are consuming a varied diet composed of multiple different whole prey species. By feeding only one species of prey, the chance of nutrient deficiencies and other health concerns increases. Improved methane (CH4) mitigation strategies are needed to achieve the greenhouse gas emission goals of the dairy industry. The first experiment was conducted to evaluate the effects of feeding black cumin (Nigella sativa) seed oil and acetate (BCA) on CH4 production and whole-animal energy utilization. Twelve dry Jersey cows were used in a 2-period crossover design study with 28d periods. The BCA treatment was compared to a control diet with corn oil. Pigs are the only mammalian livestock species encoding functional proteins for the second form of gonadotropin-releasing hormone (GnRH-II) and its receptor (GnRHR-II). In the first study, GnRHR-II localized to developing germ cells and the connecting piece of ejaculated spermatozoa. Concurrently, GnRH-II was most abundantly produced in the testis compared with other reproductive tract organs and was present within seminal plasma. Moreover, treatment of extended semen with GnRHR-II analogues altered sperm motility, morphology, and kinematics. For remaining studies, we utilized a swine line with ubiquitous knockdown of GnRHR-II (GnRHR-II KD) developed in our laboratory. The second study demonstrated that circulating GnRH-II …

This study compared pregnancy rates of beef heifers artificially inseminated with multi-sire semen to single-sire semen. It was hypothesized pregnancy rates resulting from multi-sire semen would be increased compared to single-sire semen. Heifers were artificially inseminated with semen from one of three single-sires or semen from a combination of the same three sires. Pregnancy rates did not significantly differ. Paternity testing suggests sire parentage can be unequal when semen is mixed from multiple sires. In summary, producers can expect similar pregnancy rates using single-sire and multi-sire semen, but progeny may have unequal sire representation.

Populations of zoo-managed African elephant (Loxodonta Africana) exhibit substantial individual- and population-level variation in reproductive success, yet research has largely focused on females, leaving male fertility poorly understood. Subfertility in bulls, characterized by low libido and inconsistent semen quality, poses significant challenges to genetic management and long-term population sustainability. This study a) systematically evaluates semen quality, b) identifies physiological, behavioral, social, and environmental determinants of fertility, and c) advances ex situ conservation through the expansion of the African elephant sperm bank. Variation is thought to occur in the use of dietary energy for production in dairy cattle. First, variance can be observed when new methane mitigation strategies are fed resulting in increased H2 output potentially resulting in downstream effects on estimates heat production via the Brouwer (1965) equation. Therefore, in chapter 3 a review was created to systematically describe the mathematics and theory behind the Brouwer equation, and rederive a heat production equation to integrate H2 production. Results indicate heat production using the classical Brouwer equation (1965) relative to the rederived equation resulted in an overestimation of heat production …

Project 1 focused on eight calves in a Nebraska herd (composite Simmental, Red Angus, Gelbvieh) that displayed exercise intolerance during forced activity. Available sire pedigrees contained a paternal ancestor within 2-4 generations in all affected calves. Pedigrees of the calves’ dams were unavailable, however, the cows were ranch-raised and retained from prior breeding seasons, where bulls used for breeding occasionally had a common ancestor. Therefore, it was hypothesized that a de novo autosomal recessive variant was causative of exercise intolerance in these calves. Impact Of Strategic Postweaning Supplementation On March- And May-Born Yearling Range Heifer And Calf Growth And Performance In The Nebraska Sandhills, Josie Nicole Crouch

Three experiments were conducted at the Gudmundsen Sandhills laboratory near Whitman, Nebraska. Experiment 1, a retrospective regression analysis, evaluated the impact of heifer percentage of mature body weight (BW) at breeding on heifer performance, subsequent pregnancy rates, and calf production. Overall pregnancy rates were greater (P < 0.01) for heifers at 60 to 70% of mature BW than 50 to 55%, with no differences in pregnancy rates (P ≥ 0.15) after 2-years of age. Heifers at 50 to 60% of mature BW had a greater (P = 0.05) proportion give birth in the first 21-d. A higher percentage of mature BW at breeding resulted in greater (P < 0.01) BW at birth and weaning for offspring.

Myocarditis is one of the major causes of heart failure in children and young adults and can lead to dilated cardiomyopathy. Lymphocytic myocarditis could result from autoreactive CD4+ and CD8+ T cells, but defining antigen specificity in disease pathogenesis is challenging. To address this issue, we generated T cell receptor (TCR) transgenic (Tg) C57BL/6J mice specific to cardiac myosin heavy chain (Myhc)-α 334–352 and found that Myhc-α-specific TCRs were expressed in both CD4+ and CD8+ T cells. Immune checkpoint inhibitors (ICIs) are an effective therapy for various cancers; however, they can induce immune-related adverse events (irAEs) as a side effect. Myocarditis is an uncommon, but fatal, irAE caused after ICI treatments. Currently, the mechanism of ICI-associated myocarditis is unclear. Here, we show the development of myocarditis in A/J mice induced by anti-PD-1 monoclonal antibody (mAb) administration alone without tumor cell inoculation, immunization, or viral infection. Mice with myocarditis have increased cardiac infiltration, elevated cardiac troponin levels, and arrhythmia. Anti-PD-1 mAb treatment also causes irAEs in other organs. Hematology and serum chemistry reference intervals have been previously established for the endangered Hawaiian monk seal (Neomonachus schauinslandi) as an imperative measure for health assessments. Monitoring the health of the wild population depends upon reference intervals that are context specific; hence we developed reference intervals from fresh samples, as opposed to frozen, from wild monk seals. This study builds on the number of parameters from previous efforts by using samples collected between 2004 and 2015 from wild monk seals. This Peer Review of Teaching Project portfolio focuses on the Animal Physiology II course which is required for first year veterinary medicine students. Weekly quizzes assess baseline knowledge and had been administered individually and in groups. I hypothesized the discontinuation of group quizzes would increase student effort when preparing for quizzes. Unit exams involve scenario-based questions and require students to apply information. I hypothesized the implementation of group exams would help under-performing students improve their ability to apply information they had learned. Background: We recently reported the identification of Bacillus sp. NRRL B-14911 that induces heart autoimmunity by generating cardiac-reactive T cells through molecular mimicry. This marine bacterium was originally isolated from the Gulf of Mexico, but no associations with human diseases were reported. Therefore, to characterize its biological and medical significance, we sought to determine and analyze the complete genome sequence of Bacillus sp. NRRL B-14911.

Results: Based on the phylogenetic analysis of 16S ribosomal RNA (rRNA) genes, sequence analysis of the 16S-23S rDNA intergenic transcribed spacers, phenotypic microarray, and matrix-assisted laser desorption ionization time-offlight mass spectrometry, we propose that this …

Myocarditis is an inflammation of the myocardium, but only ~10% of those affected show clinical manifestations of the disease. To study the immune events of myocardial injuries, various mouse models of myocarditis have been widely used. This study involved experimental autoimmune myocarditis (EAM) induced with cardiac myosin heavy chain (Myhc)-α 334-352 in A/J mice; the affected animals develop lymphocytic myocarditis but with no apparent clinical signs. Intense selective pressures applied over short evolutionary time have resulted in homogeneity within, but substantial variation among, horse breeds. Utilizing this population structure, 744 individuals from 33 breeds, and a 54,000 SNP genotyping array, breed-specific targets of selection were identified using an FST-based statistic calculated in 500-kb windows across the genome. A 5.5-Mb region of ECA18, in which the myostatin (MSTN) gene was centered, contained the highest signature of selection in both the Paint and Quarter Horse. Development of multiple sclerosis (MS) is more prevalent in females than in males, but the underlying mechanisms are not clear. Microbial infections have been suspected as triggers of MS and it is not known whether gender differences in reactivity to environmental antigens contribute to the disease pathogenesis. We demonstrated that ACA 83–95, a mimicry epitope from Acanthamoeba castellanii for proteolipid protein (PLP) 139–151, induces clinical signs of encephalomyelitis in both male and female SJL mice. The birth of a wild ass was completely described and photographed in the wild for the first time. It happened in the daytime, in the open desert, and in the vicinity of other wild asses. The mother isolated her foal afterwards for an entire day. After birthing she drove away her male yearling, which did not join her any longer from this moment on.
