The Effects Of Bovine Viral Diarrhea Virus And Bovine Herpesvirus Type 1 On Monocyte-Derived Dendritic Cells, Jacob Sobraske

Bovine viral diarrhea virus (BVDV) and bovine herpesvirus type 1 (BHV-1) are important pathogens that affect cattle. In this in vitro study, the main goal was determining the direct effects of BVDV and BHV-1 infections of monocyte-derived dendritic cells (MDDC) on cytokine mRNA expression. For the BVDV infected MDDC experiments, Holstein Friesian and Brown Swiss calves were included with a breed comparison with cytokine expression. Surface marker expression was also measured in BHV-1-infected MDDCs. Bovine viral diarrhea virus (BVDV) is one of the highly prevalent and economically important diseases of cattle industry worldwide. The two major consequences of this disease are persistent infection and immunosuppression. Several studies have been done to determine the underline mechanisms of BVDV-induced immunosuppression targeting antigen presenting cells, adaptive immune system cells and cytokine gene expression. However, very little research has been done to determine the effect of BVDV on neutrophils. Sera samples were collected from 21 free-ranging, captured female elk (Cervus elaphus nelsoni) in 1995- 96, and tissue and sera samples were collected from 415 hunter-harvested elk from 1995 to 2006 and tested for selected diseases. Titers for Anaplasma marginale were detected in 81 of 436 (19%) elk. Occurrence of antibodies to anaplasmosis increased from 4 to 40 elk from 2002 to 2006. Titers for bovine viral diarrhea virus (BVDV) were detected in 18 of 346 (5%) samples. Titers for Leptospira interrogans serovars were detected in 21 of 289 (7%) of samples from 1995 to 2004. The susceptibility of wild ruminants, especially cervids, to bovine viral diarrhea virus (BVDV) has remained an enigma. Two white-tailed deer (Odocoileus virginianus) were submitted to the Animal Disease Research and Diagnostic Laboratory (ADRDL) in the fall of 2003 by the South Dakota Game Fish and Parks for chronic wasting disease (CWD) testing. Both animals were CWD negative. The animals were necropsied and histopathology, viral antigen detection, and virus isolation were performed. A noncytopathic (NCP) BVDV was isolated from the lungs and several other tissues of both animals. Although commonly associated with infection in cattle, bovine viral diarrhea viruses (BVDV) also replicate in many domestic and wildlife species, including cervids. Bovine viral diarrhea viruses have been isolated from a number of cervids, including mule deer (Odocoileus hemionus), European roe deer (Capreolus capreolus), red deer (Cervus elaphus), white-tailed deer (Odocoileus virginianus), and mouse deer (Tragulus javanicus), but little information is available regarding clinical presentation and progression of infection in these species.
