Several quantitative diagnostic techniques are available to estimate gastrointestinal parasite counts in the feces of ruminants. Comparing egg and oocyst magnitudes in naturally infected samples has been a recommended approach to rank fecal techniques. In this study, we compared the Mini-FLOTAC (sensitivity of 5 eggs per gram (EPG)/oocysts per gram (OPG)) and different averaged replicates of the modified McMaster techniques (sensitivity of 33.33 EPG/OPG) in 387 fecal samples from 10 herds of naturally infected North American bison in the Central Great Plains region of the USA. Porcine reproductive and respiratory syndrome virus (PRRSV) is an RNA virus which belongs to the family Arteriviridae. Since its emergence in the late 1980’s and early 1990’s, PRRSV has had a major economic effect on the swine industry. The most notable effects of this disease are respiratory distress in piglets and late-term pregnancy failures in sows. BVDV is a major pathogen of ruminants that causes immune dysfunction and lead to secondary bacterial infection. The underlying mechanism of this dysfunction is not well understood. Based on previous results, we believe that macrophages play a central role in BVDV-associated immune dysfunction. Previously, we have shown that the infected monocyte-derived macrophage (MDM) supernatant of the high virulent strains of BVDV significantly induced lymphocyte apoptosis. In the current study, we performed experiments to identify the apoptotic factors in the infected MDM supernatants in vitro. Swine Acute Diarrhea Syndrome Coronavirus (SADS-CoV) is a member of the Coronaviridae family. The virus is associated with severe small intestine inflammation and diarrhea in suckling piglets. In 2017, SADS-CoV was first detected and identified as the causative agent of a devasting swine disease outbreak in southern China. Routine monitoring and early detection of the source of infection is therefore integral to the prevention and control of a SADS-CoV outbreak in the United States. Senecavirus A (SVA) has been associated with several outbreaks of vesicular disease (VD) since 2014 in major swine-producing countries around the world. The virus causes VD that is clinically indistinguishable from foot-and-mouth disease (FMD) in pigs. Currently there are no commercial vaccines available for SVA. Here, we developed a live attenuated SVA strain by mutating a wild type SVA strain SD15-26 (wt SVA). The live attenuated virus, rSVA mSacII, replicated less efficiently in cell cultures in vitro and expressed lower amounts of VP1 and VP2 capsid proteins than wt SVA. Swine influenza A viruses pose a serious concern for global health and worldwide swine industry considering their evasion from vaccine-induced immune responses. Formulation of effective influenza vaccines can be complicated by both antigenic shift to a different subtype of hemagglutinin (HAs) and antigenic drift within a particular HA subtype. To address these concerns, we employed a systematic approach to determine the antigenic determinants of swine influenza A viruses by focusing on H1 and H3 subtypes. Bovine viral diarrhea virus (BVDV) and bovine herpesvirus type 1 (BHV-1) are important pathogens that affect cattle. In this in vitro study, the main goal was determining the direct effects of BVDV and BHV-1 infections of monocyte-derived dendritic cells (MDDC) on cytokine mRNA expression. For the BVDV infected MDDC experiments, Holstein Friesian and Brown Swiss calves were included with a breed comparison with cytokine expression. Surface marker expression was also measured in BHV-1-infected MDDCs. The purpose of this project was to define monoclonal antibodies against viral proteins from Epizootic Hemorrhagic Disease Virus (EHDV) to be used in quality control of vaccines produced by an industrial partner, and development of tools to identify EHDV. EHDV is an Orbivirus, which also includes Bluetongue Virus and African Horse Sickness, which are all transmitted through arthropod vectors. EHDV causes significant morbidity and mortality in white-tailed deer, but has recently been found to infect cattle. Recent, severe outbreaks of porcine epidemic diarrhea virus (PEDV) in Asia and North America highlight the need for well-validated diagnostic tests for the identification of PEDV infected animals and evaluation of their immune status to this virus. PEDV was first detected in the U.S. in May 2013 and spread rapidly across the country. A novel reassortant influenza A virus (H5N2) was first detected in British Columbia, Canada, in December 2014. The virus rapidly spread along the waterfowl migration flyways in the United States, causing multiple HPAI outbreaks in poultry. Here, we present the complete genome sequence of HPAIV-H5N2 from a commercial chicken flock in Iowa.

Background: Porcine epidemic diarrhea virus (PEDV), a highly pathogenic and transmissible virus in swine, was first detected in the U.S. in May, 2013, and has caused tremendous losses to the swine industry. Due to the difficulty in isolating and growing this virus in cell culture, few vaccine studies using cell culture propagated PEDV have been performed on U.S. strains in pigs. Therefore, the objective of this study was to evaluate the humoral immune response to the selected inactivated PEDV vaccine candidate in a dose-titration manner.

Background: Since its initial detection in May 2013, porcine epidemic diarrhea virus (PEDV) has spread rapidly throughout the US swine industry. Recently, contaminated feed was confirmed as a vehicle for PEDV infection of naïve piglets. This research provides in vivo data supporting the ability of a liquid antimicrobial product to reduce this risk.
Results: Sal CURB® (Kemin Industries, Des Moines, IA, USA) is a FDA-approved liquid antimicrobial used to control Salmonella contamination in poultry and swine diets. Background: Since its initial detection in May 2013, porcine epidemic diarrhea virus (PEDV) has spread rapidly throughout the US swine industry. Initially, contaminated feed was proposed as a risk factor for PEDV; however, data were not available to support this theory. Here we provide proof of concept of this risk by describing a novel means for recovering PEDV-contaminated complete feed material from commercial swine sites and conducting an in vivo experiment to prove its infectivity.

Since the first reported cases of ducks infected with a previously unknown flavivirus in eastern China in April 2010, the virus, provisionally designated Duck Tembusu Virus (DTMUV), has spread widely in domestic ducks in China and caused significant economic losses to poultry industry. In this study, we examined in detail structural, antigenic, and evolutionary properties of envelope (E) proteins of six DTMUV isolates spanning 2010–2012, each being isolated from individual farms with different geographical locations where disease outbreaks were documented. Of the Orthomyxoviridae family of viruses, only influenza A viruses are thought to exist as multiple subtypes and has nonhuman maintenance hosts. In April 2011, nasal swabs were collected for virus isolation from pigs exhibiting influenza-like illness. Subsequent electron microscopic, biochemical, and genetic studies identified an orthomyxovirus with seven RNA segments exhibiting approximately 50% overall amino acid identity to human influenza C virus. Based on its genetic organizational similarities to influenza C viruses this virus has been provisionally designated C/Oklahoma/1334/2011 (C/OK). Bovine diarrhea is economically one of the most important problems in cattle industry. Enterotoxigenic Escherichia coli (ETEC), particularly strains expressing K99 (F5) fimbriae and heat-stable type I (STa) enterotoxin is the predominant bacterial cause of diarrhea in calves; whereas bovine viral diarrhea virus (BVDV) causes diarrhea and other illness to cattle at all ages. A vaccine that prevents against both K99 fimbrial ETEC and BVDV could benefit greatly to cattle producers worldwide. The susceptibility of wild ruminants, especially cervids, to bovine viral diarrhea virus (BVDV) has remained an enigma. Two white-tailed deer (Odocoileus virginianus) were submitted to the Animal Disease Research and Diagnostic Laboratory (ADRDL) in the fall of 2003 by the South Dakota Game Fish and Parks for chronic wasting disease (CWD) testing. Both animals were CWD negative. The animals were necropsied and histopathology, viral antigen detection, and virus isolation were performed. A noncytopathic (NCP) BVDV was isolated from the lungs and several other tissues of both animals. Formalin-fixed ear notches from both animals were positive for BVDV antigen by immunohistochemistry. …

Although commonly associated with infection in cattle, bovine viral diarrhea viruses (BVDV) also replicate in many domestic and wildlife species, including cervids. Bovine viral diarrhea viruses have been isolated from a number of cervids, including mule deer (Odocoileus hemionus), European roe deer (Capreolus capreolus), red deer (Cervus elaphus), white-tailed deer (Odocoileus virginianus), and mouse deer (Tragulus javanicus), but little information is available regarding clinical presentation and progression of infection in these species. Bovine viral diarrhea virus (BVDV) is a common virus among cattle, and there are many different strains of the virus. BVDV can cause mucosal disease in infected cattle. There are lab strains of BVDV used for testing, but there are also wild-type viruses that may develop within a herd or population. These wild-type strains can be dangerous due to cross contamination between herds. This project compares serum samples from two different herds on the basis of the antibody response to a wild-type virus isolated from a persistent infected animal in one of the herds.
