While consequences of equine obesity are well defined, the extent to which persistent hyperleptinemia and accompanying leptin (Lep) resistance affect the fetal-maternal environment warrants further exploration. This study compared endocrine, fetal, placental, and uterine parameters during early pregnancy (through day 70) in mares with elevated leptin versus normal leptin (NL) concentrations. Fourteen mares were classified as either NL (n = 6; Lep 0.9 ± 0.3 ng/mL), moderate leptin (ML; n = 4; Lep 3.0 ± 0.3 ng/mL), or high leptin (HL; n = 4; Lep 5.8 ± 0.7 ng/mL) based on annual plasma leptin. Improved methane (CH4) mitigation strategies are needed to achieve the greenhouse gas emission goals of the dairy industry. The first experiment was conducted to evaluate the effects of feeding black cumin (Nigella sativa) seed oil and acetate (BCA) on CH4 production and whole-animal energy utilization. Twelve dry Jersey cows were used in a 2-period crossover design study with 28d periods. The BCA treatment was compared to a control diet with corn oil. Pigs are the only mammalian livestock species encoding functional proteins for the second form of gonadotropin-releasing hormone (GnRH-II) and its receptor (GnRHR-II). In the first study, GnRHR-II localized to developing germ cells and the connecting piece of ejaculated spermatozoa. Concurrently, GnRH-II was most abundantly produced in the testis compared with other reproductive tract organs and was present within seminal plasma. Moreover, treatment of extended semen with GnRHR-II analogues altered sperm motility, morphology, and kinematics. For remaining studies, we utilized a swine line with ubiquitous knockdown of GnRHR-II (GnRHR-II KD) developed in our laboratory. The second study demonstrated that circulating GnRH-II …

Colostrum composition, including lactose concentration in relation to metabolomic profiles on kids’ growth performance of Black Bengal goats, has not yet been elucidated. Macronutrients, including lactose and metabolomic profiles, were determined in 37 colostrum samples using MilkoScan and 1H-nuclear magnetic resonance technique, respectively. Kids’ growth performance, including birth weight, average daily gain at 0–4 weeks (ADG0-4W), 4–8 weeks (ADG4-8W), and wean weight were determined. A total of 51 metabolites were detected in the colostrum. From the general linear model for univariate analysis, kids born from multiple litter sizes showed lower ADG during the first month postpartum (P = 0.004). …

This study compared pregnancy rates of beef heifers artificially inseminated with multi-sire semen to single-sire semen. It was hypothesized pregnancy rates resulting from multi-sire semen would be increased compared to single-sire semen. Heifers were artificially inseminated with semen from one of three single-sires or semen from a combination of the same three sires. Pregnancy rates did not significantly differ. Paternity testing suggests sire parentage can be unequal when semen is mixed from multiple sires. In summary, producers can expect similar pregnancy rates using single-sire and multi-sire semen, but progeny may have unequal sire representation.

Populations of zoo-managed African elephant (Loxodonta Africana) exhibit substantial individual- and population-level variation in reproductive success, yet research has largely focused on females, leaving male fertility poorly understood. Subfertility in bulls, characterized by low libido and inconsistent semen quality, poses significant challenges to genetic management and long-term population sustainability. This study a) systematically evaluates semen quality, b) identifies physiological, behavioral, social, and environmental determinants of fertility, and c) advances ex situ conservation through the expansion of the African elephant sperm bank. Assisted reproduction technology and the use of specialized laboratories have increased in the horse breeding industry over the last decade. This expansion has highlighted the inability to routinely cryopreserve oocytes and preserve female genetics. Oocyte vitrification, a cryopreservation process, continues to be characterized as an experimental procedure in horses. The goal of this dissertation was to evaluate the viability, developmental competence, and nuclear structures of cryopreserved equine oocytes to illuminate potential areas of improvement in the vitrification process. Project 1 focused on eight calves in a Nebraska herd (composite Simmental, Red Angus, Gelbvieh) that displayed exercise intolerance during forced activity. Available sire pedigrees contained a paternal ancestor within 2-4 generations in all affected calves. Pedigrees of the calves’ dams were unavailable, however, the cows were ranch-raised and retained from prior breeding seasons, where bulls used for breeding occasionally had a common ancestor. Therefore, it was hypothesized that a de novo autosomal recessive variant was causative of exercise intolerance in these calves. An excretory/secretory (E/S) product, common to many helminths, is neutrophil inhibitory factor (NIF) which negatively affects neutrophil migration and activity. Due to differences in neutrophil accumulation between parasite resistant St. Croix STC and parasite susceptible Suffolk (SUF) sheep, we hypothesized that Hc-NIF may inhibit migration of SUF and not STC neutrophils. Previous studies have demonstrated that Hc produces NIF but is not commercially available. To test our hypothesis, neutrophils were cultured from STC and SUF sheep in the presence of Ancylostoma caninum-derived NIF (Ac-NIF) and measure chemotaxis to Interleukin-8 (IL-8), HcLA or H. contortus third-stage excretory/secretory (E/S) products. Pets have historically been viewed as family members, children, property, or economic resources. However, research surrounding animal maltreatment has expressed this issue as an individually-based problem, rather than a community-based phenomenon. Correlations have been found between animal cruelty, antisocial behaviors, and future interpersonal violence, whether this correlation be a predictive relationship, or a resultant relationship. Past research has also found correlations between animal treatment practices and the rural/urban differences of this behavior. Hematology and serum chemistry reference intervals have been previously established for the endangered Hawaiian monk seal (Neomonachus schauinslandi) as an imperative measure for health assessments. Monitoring the health of the wild population depends upon reference intervals that are context specific; hence we developed reference intervals from fresh samples, as opposed to frozen, from wild monk seals. This study builds on the number of parameters from previous efforts by using samples collected between 2004 and 2015 from wild monk seals. Equine arteritis virus (EAV) has a global impact on the equine industry as the causative agent of equine viral arteritis (EVA), a respiratory, systemic, and reproductive disease of equids. A distinctive feature of EAV infection is that it establishes long-term persistent infection in 10 to 70% of infected stallions (carriers). In these stallions, EAV is detectable only in the reproductive tract, and viral persistence occurs despite the presence of high serum neutralizing antibody titers. The mare experiences a transient innate immune response to breeding, the resolution of which is crucial for optimal fertility. The majority of mares are able to modulate this inflammation in a timely fashion, but a subpopulation exists which fail to do so and are considered susceptible to persistent breeding-induced endometritis (PBIE). Seminal plasma has been shown to modulate aspects of this inflammation. Recently, two seminal plasma proteins have garnered interest for their immune modulating properties: cysteine-rich secretory protein-3 (CRISP-3) and lactoferrin. Degenerative joint disease, or osteoarthritis, is a major cause of lameness and morbidity in horses, humans, and dogs. There are no truly satisfactory cures for this widespread problem and current treatments all have limitations or unwanted side effects.
New cell-based strategies to repair joint surface lesions have generated a high level of interest, but have yet to achieve the full restoration of articular cartilage structure and function. Currently used therapy cells include autologous chondrocytes and adult mesenchymal cells such as bone marrow derived cells and adipose derived cells. Artificial insemination (AI) is commonly used for breeding of horses. Selecting high quality semen is critical to AI success but is difficult to achieve. In this study, we investigate the potential of generating antibodies against stallion sperm to identify subpopulations of post-thaw sperm based on surface markers. Female Balb/C mice are immunized with semen from a 24-year-old fertile stallion followed by booster immunizations each week for at least six weeks. Monoclonal antibodies are generated by fusion of myeloma cells to mouse spleen cells. Eight monoclonal antibodies resulting from these fusions are subdivided into four immunostaining patterns. The birth of a wild ass was completely described and photographed in the wild for the first time. It happened in the daytime, in the open desert, and in the vicinity of other wild asses. The mother isolated her foal afterwards for an entire day. After birthing she drove away her male yearling, which did not join her any longer from this moment on.
