The aim of this study was to identify the presence and prevalence of bovine tuberculosis in the eastern Anatolian border province of Van and its villages and to collect the preliminary data required for programs for the prevention and control of this zoonotic disease. The tuberculin test was performed on 210 cattle. Nasal swab and milk samples were obtained from the animals tested. Three cattle that had a positive tuberculin test also tested positive for the DNA target, a 580-bp fragment of IS6110 specific for members of the Mycobacterium tuberculosis complex. A total of 1500 serum samples were collected from cattle, buffaloes, and humans. The Rose Bengal plate test (RBPT) and enzyme-linked immunosorbent assay (ELISA) detected Brucella antibodies in 56 (10.18%) and 44 (8%) of the cattle, respectively. In samples collected from buffaloes 9.38% and 6.92% were positive, whereas 14% and 11% of the samples from humans were found to be positive by RBPT and ELISA, respectively. Among village workers a higher prevalence of brucellosis was recorded in females than in males. Abattoir-associated personnel also had a higher incidence of brucellosis. The aim of the present study was to evaluate the efficiency of different doses of epidermal growth factor (EGF) on maturation and fertilisation of bovine oocytes in vitro. A total of 1209 high quality oocytes (follicles 2-8 mm in diameter, with unexpanded cumulus cells) were obtained by aspiration from ovaries collected at a slaughterhouse. The oocytes were then assigned into 6 groups: 1, 10, 100, and 1000 ng/ml EGF groups, as well as positive (EGF replaced with hCG) and negative (TCM 199 only, no supplement) control groups. In this study, a total of 30 Escherichia coli isolates obtained from milk samples of dairy cows suffering from subclinical mastitis in Konya, Turkey were typed according to plasmids and protein patterns. Agarose gel electrophoresis and sodium doedecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) methods were used to identify plasmids and whole-cell protein profiles. Of these two methods, SDS-PAGE typing proved to be more discriminate for typing the isolates.

Meningitis Associated With Brucella Abortus In An Aborted Bovine Foetus, Cengi̇z Çeti̇n, İ. Taci̇ Cangül, Mehmet Akan, Ebru Özel, Ahmet Akkoç

In this study, the brain and other organs from an aborted bovine foetus were examined histopathologically and microbiologically. In addition, a serum sample from the aborting cow was examined serologically. A granulomatous inflammation, characterised by the infiltration of lymphocytes, plasma cells with some neutrophils and macrophages was observed in the meninges of the brain, liver and kidney. Brucella abortus biovar 1 was isolated in pure culture from the brain and other organs. The serum sample was positive for brucellosis. These findings indicate that Brucella abortus is a pathogen capable of inducing meningitis in bovine foetuses.

First Identification Of Neospora Caninum In Thailand, Than Kyaw, Prachin Virakul, Manop Muangyai, Wijit Banlunara

An immunohistochemical examination of aborted bovine tissues, collected from two dairy farms that had a high abortion rate (7%), detected the organism Neospora caninum (NC). Five out of 12 aborting cows (41.7%) were seropositive to NC using a competitive ELISA test. Neospora-tachyzoites were detected in the placenta of a seropositive aborting cow. Neither NC parasites nor cysts were detected in other fetal tissues. The identification of the Neospora-parasite and a high percentage of Neospora seropositive aborting cows, suggested NC as the possible cause of abortion. This is the first identification of the NC parasite in Thailand.

Cyclodextrins are used extensively in food, cosmetic, drug and chemical industries to increase aqueous solubility and stability, and reduce or eliminate the unpleasant taste and smell of many products. Experiments were conducted to examine the effect of a pre-haemolytic dose of methyl b-cyclodextrin (MBCD) on the osmotic fragility of erythrocytes collected from sheep, cattle and humans. Heparinised blood collected from these three species were analysed within 3 h of collection. Erythrocyte suspensions were mixed with MBCD to give concentrations of 2 mM, 4 mM or 6 mM cyclodextrin. This study was conducted to determine leptospirosis from aborted bovine fetuses. Tissue sections of the liver, lung, spleen and kidney were stained with avidin-biotin peroxidase complex, and antigen localization was examined. These examinations showed positive staining in eight (24.24%) out of 33 fetuses for the presence of leptospira antigens. In these eight cases, leptospira antigens were found in three lung (37.5%), two liver (25%), four kidney (50%) and four spleen (50%) samples. Microscopic studies demonstrated that leptospiral antigens were located in the interalveolar and interlobular septum of the lung; in the cytoplasm of hepatocytes and macrophages in the portal regions …

Transgenic Bovine Nuclear Transfer Embryos From Adult Somatic Cell Lines*, Sezen Arat, S. Jacek Rzucidlo, Steven L. Stice

This study was performed to examine whether adult fibroblast cells can be manipulated genetically and whether clonal lines derived from those cells can support embryonic development. A primary adult cell line was established from the ear of an aged cow. Adult cells with a plasmid containing the enhancer green fluorescence protein (EGFP) gene, and non-transfected cells were used for cloning. Green fluorescence expression was observed in 35/49 (71.4%) adult clones. The developmental rates of embryos were significantly lower for cell lines expressing EGFP (11.4%) than for non-transfected cells (20.1%, P < 0.05). However, there was no decrease in nuclear transfer (NT) developmental rates (21.5%) when donor nuclei from EGFP transfected cell lines not expressing EGFP but retaining neomycin-resistance gene expression were used as donor nuclei. The NT embryos from transfected and non-transfected cell lines had similar morphology and cell numbers. The results indicated that adult cells can complete clonal propagation including transfection and selection and can be used to produce transgenic NT embryos.

This study was carried out in order to serologically and haematologically determine the prevalance of Bovine Leukaemia Virus infection in cattle raised in Kars and environs.For this purpose a total of 443 blood samples and 383 milk samples belonging to the various breeds (Zavot, Holstein, Brown-Swiss and Native), and sexes (383 female, 60 male)were examined serologically (Agar Gel Immunodiffusion (AGID)and Enzyme Linked Immunosorbent Assay (ELISA))and haematologically (lymphocyt counts, total leucocyt counts and haematocrit value). Primer oocytes (n=157) collected from ovaries of slaughtered cows were matured at 39°C with 95-100% humidity and under a gas mixture of 5% O 2 , 5% CO 2 and 90% N 2 for 22 (n=52), 24 (n=52) and 26 (n=53) hours. Modified Parker's Medium (MPM) supplemented with FSH and 20% ECS was used as maturation medium. Neuraminidase-treated bovine serum amyloid P component (SAP) has chromatographic profile from DEAE-HPLC column of shorter retention time and less peak number than intact molecule does. SDS-PAGE analysis reveals a decrease in size of bovine SAP subunits after neuraminidase treatment, compared with an untreated molecule. The interaction between semen quality of Holstein Friesian, Jersey, Crossbred and Nili-Ravi buffaloes and the environment was studied. Semen was collected for one year at the Semen Production Unit, Surizai District, Peshawar from 10 Holstein Friesian, 4 Jersey and 4 Crossbred bulls and 3 buffaloes. Semen was collected early in the morning, twice weekly, using an artificial vagina. Environmental data regarding the monthly average temperature, humidity and daylight, was recorded. The correlation co-effi-cient between the semen volume, motility and sperm concentration and the three environmental parameters was calculated. Bovine CRP cDNA of 2.2 kb was isolated from bovine liver cDNA library by using the probe containing sequence homologous in human CRP and rabbit CRP. Restriction map of this cDNA showed only one site for each following restriction enzyme EcoRI, HindIII, PstI, Sacl and Xbal. DNA sequence analysis revealed that bovine CRP cDNA has 57 bp coded for signal peptide of 19 amino acid and 618 bp for mature protein of 206 amino acid residues. Three monoclonal antibodies (MAbs) against bovine C-reactive protein (CRP) were produced by fusing murine myeloma cell line P3X63Ag8U1 and splenic lympocytes from two BALB/c mice immunized with 100 ug of purified CRP, followed by two booster injections at 10-day intervals. The hybridomas producing antibodies specific for bovine CRP were obtained with effciencies of 1.3 % and 0.2 % respectively. All three MAbs were IgM class with k- light chains. The MAbs obtained from ascitic fluid showed definite antibody activity at 1,000 to 10,000 and the antibody concentrations were 0.9 to 1.7 mg/ml.
