West Nile virus (WNV) was first isolated in 1937 from a febrile Ugandan woman. WNV now has a worldwide distribution from Australia and India in the east to Europe and the United States in the west. The first human cases of WNV in the United States were detected in New York in 1999. The North American isolates were also found to be highly neuroinvasive and neurovirulent and in many cases leading to mortality or permanent CNS sequelae among humans. Ascending placentitis is a significant cause of abortions, stillbirths, and perinatal loss in horses. A technique for laparoscopic-guided catheterization of the allantoic space was developed and utilized in an experimental model of streptococcal infective pre-term delivery in pony mares. Mares received either 1 x 107 CFU live S. zooepidemicus (n=3), 5.1 x 108 CFU live S. zooepidemicus (n=1), 1 x 107 heat-killed S. zooepidemicus (n=3), 1 mL sterile PBS (n=3). Sham control mares did not receive a transcervical inoculation (n=3). One mare not instrumented with an allantoic catheter received 5.1 x 108 CFU live S. zooepidemicus. Heme oxygenase-1 (HO-1) catalyzes the initial and rate-limiting reaction in heme catabolism (yielding equimolar quantities of iron, carbon monoxide, and biliverdin), which is further metabolized to bilirubin by biliverdin reductase. HO-1, an important enzyme with antioxidant and cytoprotective properties, functions against oxidative stress generated in response to heme and to various stress stimuli, such as zinc. The cytoprotective function of HO-1 is dependent on both the activities of the reaction products and on the inducibility of the heme oxygenase gene (hmox-1), which is regulated primarily at the level of gene transcription by two distal enhancers, E1 and E2. HSV-1 acquires its final envelope by budding into cytoplasmic vesicles thought to be derived from Trans-Golgi Network (TGN) membranes. This process is facilitated by interactions among the carboxyl termini of viral glycoproteins and tegument proteins. To investigate the relative importance of different viral glycoproteins in cytoplasmic virion morphogenesis, a set of recombinant viruses were constructed silencing expression of the glycoprotein E (ΔgE), the carboxyl terminus of glycoprotein D (gDΔcp), and the membrane protein UL20 (ΔUL20). In addition, recombinant viruses were constructed having the ΔgE+gDΔcp, and the ΔgE+ΔgM (glycoprotein M) deletions. A conditional silencing system using anti-gB siRNAs was devised to investigate the structure and function of the KSHV gB. Transient co-transfection of plasmids constitutively expressing gB and anti-gB siRNAs in 293 cells substantially inhibited gB mRNA levels and protein production. Similarly, transient expression of siRNAs into the basal cavity-based lymphoma cells (BCBL-1) caused substantial reduction of gB transcription and protein synthesis. TaqMan real-time PCR and infectivity assays showed that gB was essential for virion egress and infectivity. Transfection of a codon-optimized gB not recognized by the anti-gB siRNAs, efficiently rescued virion egress and infectivity in BCBL-1 cells in the presence …

Kaposi’s sarcoma-associated herpesvirus (KSHV) replication and transcription activator (RTA) is necessary and sufficient for the switch from KSHV latency to lytic replication. RTA activates promoters by several mechanisms. RTA can bind to sequences in viral promoters and activate transcription. In addition, RTA interacts with the cellular recombination signal sequence-binding protein-J kappa (RBP- Jκ), a transcriptional repressor, converts the repressor into an activator and activates viral promoters via RBP- Jκ. Bovine herpesvirus type 1 (BHV-1) is an important pathogen that can initiate bovine respiratory disease complex. Like other members of the subfamily Alphaherpesvirinae, BHV-1 establishes latency in sensory neurons. The latency-related (LR) gene expresses a family of alternatively spliced transcripts in infected sensory neurons that have the potential to encode several LR proteins. An LR mutant virus that contains three stop codons near the 5’ terminus of the first open reading frame in the LR gene does not express two LR proteins or reactivate from latency. Ingestion of endophyte infected (E+) fescue by pregnant mares can cause significant reproductive problems. Plateau® and Cimarron® herbicides suppress fescue while leaving desired forages unharmed. To determine if these herbicides are harmful to pregnant mares, they were allowed to graze pastures treated with Plateau®, Cimarron®, or vehicle carrier. Pregnancies were monitored via ultrasonography, blood chemistry, and hematology. Of the components measured only creatinine differed among treatments over time (P=0.0003) and that increase was only significant in one of four studies.

Computational models have been successfully applied to a wide variety of research areas including infectious disease epidemiology. Especially for questions that are difficult to examine in other ways, computational models have been used to extend the range of epidemiological issues that can be addressed, advance theoretical understanding of disease processes and help identify specific intervention strategies. We explore each of these contributions to epidemiology research through discussion and examples. We also describe in detail models for raccoon rabies and methicillin-resis-tant Staphylococcus aureus, drawn from our own research, to further illustrate the role of computation in epidemiological modeling.

Citizens and animals in states without a veterinary school are at a disadvantage if the presence of a veterinary school significantly improves public health and access to medical services in those states that have a veterinary school.

Sheep scrapie is the prototypical transmissible spongiform encephalopathy (prion disease), which has a fundamental pathogenesis involving conversion of normal cellular prion protein (PrPC [C superscript stands for cellular]) to disease-associated prion protein (PrPSc [Sc superscript stands for sheep scrapie]). Sheep microglial cell cultures, derived from a prnp 136VV/171QQ near-term fetal brain, were developed to study sheep scrapie in the natural host and to investigate potential cofactors in the prion conversion process. Bovine herpesvirus 1 (BHV-1), a member of the alpha-herpesvirinae subfamily, causes significant losses to the cattle industry. BHV-1 establishes latency in trigeminal ganglionic sensory neurons, but periodically reactivates from latency. Previous studies suggested that infection with BHV-1 induced novel morphological changes in rabbit skin cells (RS) versus bovine kidney cells (MDBK). Consequently, we hypothesized that viral infection led to a novel form of cell death in RS cells compared to MDBK cells. Infection of cattle by bovine herpesvirus type 1 (BHV-1) can lead to upper respiratory tract disorders, conjunctivitis, genital disorders and immune suppression. BHV-1-induced immune suppression initiates bovine respiratory disease complex (BRDC), which costs the US cattle industry approximately 3 billion dollars annually. BHV-1 encodes at least three proteins that can inhibit specific arms of the immune system: (i) bICPO inhibits interferon-dependent transcription, (ii) the UL41.5 protein inhibits CD8+ T-cell recognition of infected cells by preventing trafficking of viral peptides to the surface of the cells and (iii) glycoprotein G is a chemokine-binding protein that prevents homing of lymphocytes to …

Background: In order to promote consumer-oriented informed medical decision-making regarding screening mammography, we created a decision model to predict the age dependence of the cancer detection rate, the recall rate and the secondary performance measures (positive predictive values, total intervention rate, and positive biopsy fraction) for a baseline mammogram.
Methods: We constructed a decision tree to model the possible outcomes of a baseline screening mammogram in women ages 35 to 65. We compared the single baseline screening mammogram decision with the no screening alternative. Equine bone marrow-derived mesenchymal stromal cells (MSCs) and adipose tissue-derived stromal cells (ASCs) were compared for frequency within their respective tissues, cell doubling characteristics and differentiation multipotential in culture based on histochemical staining and compositional analysis. Equine MSCs and ASCs from young adult horses were harvested and isolated from sternal bone marrow and supragluteal subcutaneous adipose tissue, respectively, and grown up to passage 10 (P10) to determine cell doubling characteristics. In this research smoking and canning techniques were applied to cultured mussels (Mytilus galloprovincialis, L.) from the Çanakkkale Strait in Eceabat, Turkey. Mussels that were smoked by liquid and traditional methods were canned in different sauces. The chemical composition and amino acid composition of the canned smoked mussels were determined by the results of laboratory analyses. As a result of smoking and canning mussels, a food with high nutritional value was obtained. A Study Of Somatic Cell Counts In The Milk Of Holstein-Friesian Cows Managed In Mediterranean Climatic Conditions, Atakan Koç

Somatic cell counts (SCC) in the milk of Holstein-Friesian (HF) cows were determined by direct microscopic SCC technique. For a two-year period, milk samples were collected monthly from buckets during the morning and evening milkings at 4 different dairy farms. In total, 1,464 SCC readings from 88 HF cows were analyzed by using repeated measures. Herd, lactation month, parity, milking time, and herd interaction effects on SCC were statistically significant (P < 0.05). The average SCC in milk per herd was between 296,483 and 688,811 cells/ml. SCC in milk increased as parity increased. The average SCC in milk from evening milking was about 83,165 cells/ml higher than from morning milking. Improving milking management, reducing stress, providing extra care during the first month of lactation, and milking at a uniform interval will help to decrease SCC in milk and the prevalence of mastitis.

The quantity and quality of Japanese quail (Coturnix japonica) semen collected individually after stimulation by a teaser female was assessed. From among 25 mature males, 10 were selected for experimental purposes on the basis of reaction intensity to stimulation, size of the proctodeal foam gland, and ejaculation speed. Males were kept individually in cages (32 x 44 x 24 cm) and 5 females that served for male stimulation were kept in a group cage (60 x 45 x 48 cm). The aims of this study were to isolate Pseudomonas aeruginosa from dogs with otitis externa (OE), to determine the susceptibility of isolated strains to antibiotics, and to evaluate the incidence of the infection in the Aydın region. For this purpose, ear swab samples were obtained from 92 dogs with symptoms of OE infection and the samples were examined bacteriologically. Bacteria were isolated from 88 (96%) of 92 samples but no isolation was performed on 4 (4%) dogs. In total 93 microorganisms were identified. This study was conducted to investigate the effects of mannan oligosaccharides (MOSs), which are commonly used as alternatives to antibiotics, on the growth performance and some blood parameters in broiler chickens. The study included 120 Ross 308 male broiler chicks (1-day-old at the beginning of the study). There were 4 treatment groups, each consisting of 3 replicates. This study aimed to determine the shelf life, as well as chemical and microbiological quality of gilthead seabream (Sparus aurata L., 1758) prepared by 2 different methods of smoking (hot smoking and cold smoking). The effects of hot and cold smoking on the chemical composition and microbial load of gilthead seabream, as well as organoleptic analysis of the smoked product were investigated. Significant (P < 0.05) differences were found in the chemical composition of fresh and smoked seabream. The panelists liked the hot smoked fish more than the cold smoked fish, according to sensory analysis results. Changes in pH, thiobarbituric acid (TBA), and total volatile basic nitrogen (TVB-N) values were significant (P < 0.05) during storage at 4 °C. Microbiological analysis results demonstrated that the smoking techniques reduced the microbial content of the fish, whereas microbial content increased during storage. The smoking methods tested had a small effect on the level of vitamin D_3 in gilthead seabream.

A total of 554 cattle from the Cappadocia region were divided into 5 different groups according to vaccination and were examined for Theileria annulata by microscope and indirect fluorescence antibody test (IFAT). The sampled cattle were checked for tick infestation. The prevalence of T. annulata was 60.5% by microscopic examination and 67.5% by IFAT. According to the microscopic examination, the prevalence of T. annulata was significantly (P < 0.001) higher in the unvaccinated group. The seropositivity according to IFAT was significantly (P < 0.001) lower in the unvaccinated group. The morbidity was significantly (P < 0.001) higher in the unvaccinated, semi-grazed, pure breed, and >2 years age groups. The mortality was significantly higher in the unvaccinated (P < 0.001), semi-grazed (P < 0.05), pure breed (P < 0.05), and female (P < 0.001) groups. A total of 1585 tick specimens belonging to the genera Hyalomma, Rhipicephalus, Dermacentor, Boophilus, and Ornithodorus were detected in 21.48% of examined cattle. The developing forms of T. annulata were determined in the H. anatolicum anatolicum ticks microscopically. Out of 554 cattle, 156 (27.61%) were diagnosed with acute tropical theileriosis. Out of these 156 cattle, 86 (56.21%) died due to the disease.

In this case report, metastasis of a transmissible venereal tumor (TVT) to the uterus and ovaries is presented. A 7-year-old female boxer dog with hemorrhagic vulvar discharge and a vaginal mass was brought to Ankara University Faculty of Veterinary Medicine, Clinics of Gynecology and Obstetrics. TVT was diagnosed by clinical findings and cytologic examination. Multifocal nodules in the uterus and enlargements of the uterine wall were detected by ultrasonographic examination. Ovariohysterectomy was performed because of uterine pathology. Histopathologic examination of the uterus and ovarium showed that the multifocal masses in the uterus and ovaries originated from vaginal TVT.

Jambo grass (Sorghum bicolour x Sorghum sudanefe) harvested 50 days after sowing was ensiled in laboratory silos using 2 additives, cane molasses and ground corn grains, each at the rate of 2%, 4%, and 6% levels for 30, 35, and 40 days at room temperature (28 °C). Data regarding incubation days were pooled. The pH and lactic acid concentration in jambo grass silage were not affected by the additive type, level, or fermentation period. Dry matter (DM) content of jambo silage was significantly (P < 0.05) affected by additive type; however, additive levels and fermentation periods did not affect its DM contents. Crude protein content of jambo grass was not affected by ensiling time, additive type, or level. The true protein contents of jambo grass silage were significantly (P < 0.05) higher with molasses than with ground corn. Neutral detergent fiber (NDF) and hemicellulose content of jambo grass were significantly (P < 0.05) affected by additives and their levels, but fermentation periods did not influence the NDF contents of jambo grass silage. Cellulose, acid detergent fiber, and acid detergent lignin contents of jambo silage were not affected by additive type, level, or fermentation period. Therefore, jambo grass ensiled with 2% molasses for 30 days was selected for in situ digestion kinetics in bulls. Dry matter and NDF degradabilities of jambo grass were significantly higher than those of its silage at 48 h of ruminal incubation. Ruminal lag time, and rate and extent of DM and NDF degradation were similar for jambo grass and its silage. The results of this study implied that jambo grass ensiled with 2% molasses for 30 days has similar nutritive value to that of its forage in bulls.
