The Evaluation Of Selected 13 Polymorphic Microsatellite Markers For The Parentage Test Of Thai Holstein Cattle, Lerdchai Chintapitaksakul, Rapiphan Uavechanichkul
The Thai Journal of Veterinary Medicine
Nineteen families of 44 Thai Holstein cattle were evaluated for microsatellite characteristics and the parentage test. A total of 13 microsatellite markers were selected from the genome database and the genomic DNAs were isolated and used as templates for amplification. Multiplex PCR of these thirteen markers in a single reaction showed that only ten markers; b1, b2, b3, b4, b6, b7, b10, b11, b12 and b13 could amplify the fragments ranging from 5 to 12. The experiment was conducted to investigate the effects of renin-angiotensin blockade on renal function, renal norepinephrine (NE) contents and renal oxidative stress in Cyclosporine A (CsA) induced renal impairment in rats. Rats were assigned into three groups; group 1 (control group), receiving vehicle (propylene glycol) 1 ml/kg./day, subcutaneously (s/c). for 28 days; group 2 (CsA group), receiving CsA 15 mg/kg/day, s/c for 28 days; group 3 (CsA and losartan (LST) group), receiving CsA 15 mg/kg./day, s/c with the administration of LST 10 mg/k.g./day orally for 28 days. Thioredoxin (TRx) is known to control redox homeostasis in cells. In recent years, a specific TRx binding protein called thioredoxin binding protein-2 (TBP-2) was found in other cell types and it appeared to negatively regulate TRx bioavailability and thereby control TRx biological function. In view of the sensitivity of lens transparency to redox status, proper regulation of TRx bioavailability is of the utmost importance. This study was conducted to examine the presence and function of TBP-2 in human lens epithelial cells (HLE B3). BACKGROUND: Identification of biomarkers among thousands of genes arrayed for disease classification has been the subject of considerable research in recent years. These studies have focused on disease classification, comparing experimental groups of effected to normal patients. Related experiments can be done to identify tissue-restricted biomarkers, genes with a high level of expression in one tissue compared to other tissue types in the body.
RESULTS: In this study, cartilage was compared with ten other body tissues using a two color array experimental design. Thirty-seven probe sets were identified as cartilage biomarkers. Of these, 13 (35%) have existing annotation associated with …

The susceptibility of wild ruminants, especially cervids, to bovine viral diarrhea virus (BVDV) has remained an enigma. Two white-tailed deer (Odocoileus virginianus) were submitted to the Animal Disease Research and Diagnostic Laboratory (ADRDL) in the fall of 2003 by the South Dakota Game Fish and Parks for chronic wasting disease (CWD) testing. Both animals were CWD negative. The animals were necropsied and histopathology, viral antigen detection, and virus isolation were performed. A noncytopathic (NCP) BVDV was isolated from the lungs and several other tissues of both animals. Formalin-fixed ear notches from both animals were positive for BVDV antigen by immunohistochemistry. …

In swine, the most common and severe enterotoxigenic Escherichia coli (ETEC) infections are caused by strains that express K88 (F4)+ fimbriae, heat-labile enterotoxin (LT), heat-stable enterotoxin b (STb), and enteroaggregative E. coli heat-stable toxin 1. Previous studies based on a design that involved enterotoxin genes cloned into a nontoxigenic fimbriated strain have suggested that LT but not STb plays an important role in dehydrating diarrheal disease in piglets study, we compared these two toxins in terms of importance for piglets >1 week old with a study design that involved construction of isogenic single- and double-deletion mutants and inoculation of …

In July 2005, a Minnesota beef herd tested positive for Mycobacterium bovis (M. bovis) and was officially declared infected with bovine tuberculosis (TB). This was the first infected herd identified since Minnesota’s Accredited Free (AF) status was obtained in 1976. Subsequent testing identified infection in white-tailed deer and several cattle herds adjacent to this index herd. In February 2008, Minnesota declared its 11th infected cattle herd, resulting in the downgrade of the entire State’s TB status to Modified Accredited (MA).
In an effort to minimize the impact of MA status to the State and producers, Minnesota initiated the …

The applications of molecular detection using nested PCR and the routine microscopic examination of ehrlichiosis were applied to 43 clinically affected dogs residing in Bangkok and Samutraprakarn. This study, performed in 2005-2006, has determined that 79.5% (31/39) of the buffy coat smears were having the ehrlichial morulae plus initial bodies found in monocytes and lymphocytes. The nested PCR was used to confirm the occurrence of ehrlichiosis after tested by morulae identification. Both tests demonstrated that neither Ehrlichia chaffeensis nor E. ewingii, the zoonotic agents of human, were found in this area. However, the prevalence of E. canis alone detected by …

An immunohistochemical study describes the localization of IgM containing cells involved in the primary immune organs of tilapia and the fate of E. tarda antigen distributed in the tissues during a course of intraperitoneal challenge. The E. tarda antigen was diffusely detected in various organs except the brain and spinal cord. The positive signals were identified within various cell types, including phagocytic cells, serosal mesothelia, myocardia, intestinal and gill epithelia and glomerular capillary endothelia. No extracellular deposition of bacterial antigen was observed. The bacterial antigen can persist up to 1 month in the granuloma and melano-macrophage center, confirming the intracellular …

The amount of new information of the last seven years regarding Lawsonia intracellularis sensitivity to antimicrobial drugs, prevention/control based on treatment and vaccination, diagnosis and, finally, genome is amazing. Nevertheless, not much is known about the epidemiology of the disease, mainly regarding the source of infection, resistance of the bacteria in the environment and possible biological vectors that could spread the infection among farms. It was learned that as few as 1,000 to 10,000 bacteria are enough to induce infection and fecal shedding two to three weeks after inoculating susceptible pigs. Tube Realignment For Patellar Luxation Repair In Dogs, Chalika Wangdee, Marissak Kalpravidh

Grade II patellar luxations in dogs were repaired by trochlear wedge recession in 5 stifles (group 1) and by proximal tube realignment in 5 stifles (group 2). A combination of the two techniques was used to correct grade II luxation in 10 stifles (group 3) and grade III luxation in 5 stifles (group 4). The position of the patella, weight bearing, and animal gait were postoperatively examined for 12 weeks. Satisfactory results were observed in 1(20%), 3(60%), and 9(90%) stifles with grade II luxation in groups 1, 2, and 3, respectively. Good recovery from grade III luxation was observed in …

The present study was conducted to synchronize estrus and fixed time insemination in dairy cattle during the hot and humid months (July to November) in Thailand. Thirty-four cows and heifers were treated by using norgestomet+PGF2α+equine chorionic gonadotropin (eCG) program. Norgestomet was implanted subcutaneously at the back of the ear for 10 days. PGF2α was administered IM on day 8 and one injection of eCG was given on the day of implant removal. Fixed time artificial insemination(AI) were performed twice, at about 54-60 and 70-74 h, after the eCG injection. Blood samples were collected at 22 days after AI for progesterone …

Recombinant mycobacterial strains which overproduce essential biosynthetic enzymes of pathogenic mycobateria are provided. These strains overproduce enzymes involved in the synthesis and incorporation of D-alanine into mycobacterial peptidoglycan, the backbone of the mycobacterial cell wall. These overproducing strains may be used as reference strains in in vitro screening methods to identify antimycobacterial agents.

The Caribbean spiny lobster Panulirus argus, an important fisheries species, is host to Panulirus argus virus 1 (PaV1), a lethal, unclassified virus-the first found in any species of lobster-prevalent in juvenile lobsters. We describe a series of laboratory experiments aimed at assessing the likely modes of disease transmission, determining the survival of lobsters relative to each transmission pathway and identifying potential alternate hosts. Given evidence for lower prevalence of PaV1 in large lobsters, the effect of lobster size on susceptibility was also examined. Porcine reproductive and respiratory syndrome virus (PRRSV) is an economically significant agent for which there currently are no effective treatments. Development of antiviral agents for PRRSV as well as many other viruses has been limited by toxicity of known antiviral compounds. In contrast, antibiotics for non-virus microbial infections have been widely useful, in part because of their acceptable toxicity in animals. We report here the discovery that the quinolone containing compound Plasmocin™, as well as the quinolones nalidixic acid and ciprofl oxacin, have potent anti-PRRSV activity in vitro. The experiment was conducted to investigate the effects of supplementing a commercially available probiotic (an Active Elements®; AE) containing Lactobacillus plantarum and Saccharomyces cerevisiae, an antibiotic; Chlortetracycline (CTC), and the probiotic plus antibiotic, via the drinking water on broiler production performance, mortality rate and the antibiotic residue in meat. Four hundred one-day-old male Cobb broiler chicks were randomly allocated to 4 treatments; a non-treated control, a 0.05% CTC, a 1:500 AE, and a combination of CTC and AE.
