This study evaluated feeding wet or dry traditional distillers grains with wet and dry fractionated distillers (Solbran) fed at 40% of diet DM compared to a corn control diet on enteric methane emissions and performance. Compared to the control, there was no difference in carbon dioxide production, but there was an increase in methane production in the steers fed fractionated distillers compared to conventional distillers. Feeding steers traditional or fractionated distillers grains increased consumption, increased gain, but improved conversions only for steers fed WDGS, while steers fed the other distillers grains had worse or the same feed …

Populations of zoo-managed African elephant (Loxodonta Africana) exhibit substantial individual- and population-level variation in reproductive success, yet research has largely focused on females, leaving male fertility poorly understood. Subfertility in bulls, characterized by low libido and inconsistent semen quality, poses significant challenges to genetic management and long-term population sustainability. This study a) systematically evaluates semen quality, b) identifies physiological, behavioral, social, and environmental determinants of fertility, and c) advances ex situ conservation through the expansion of the African elephant sperm bank. Variation is thought to occur in the use of dietary energy for production in dairy cattle. First, variance can be observed when new methane mitigation strategies are fed resulting in increased H2 output potentially resulting in downstream effects on estimates heat production via the Brouwer (1965) equation. Therefore, in chapter 3 a review was created to systematically describe the mathematics and theory behind the Brouwer equation, and rederive a heat production equation to integrate H2 production. Results indicate heat production using the classical Brouwer equation (1965) relative to the rederived equation resulted in an overestimation of heat production …

Government agencies and landowners use a variety of lethal methods to reduce invasive wild pig populations and resulting agricultural damages. To provide decision-makers with information regarding Alabama farmers’ experiences with wild pigs and their preferences for management actions, we surveyed Alabama Farmers Federation members in 2020. We found that wild pigs were commonly present on livestock, timber, and crop production acreage, but wild pig damages and costs were much more common among crop producers. Respondents' wildlife acceptance capacity for wild pigs was low and all lethal control methods were considered aceptable by most reposndents, particularly those performed by landowners. While …

A classifier tool was created to identify major serogroups of Moraxella bovis based on spectral profiles collected from bacterial cells through the use of Fourier transformed infrared spectroscopy. M. bovis strains were selected from a group of previously sequenced and genotyped isolates, which provided the pilin sequence type for each isolate. The obtained spectroscopic profiles were analyzed through artificial neural networks to create a classifier tool that could identify the PilA type for unknown strains. With the ever-increasing demand for animal protein and the continual need for the assurance of proper animal welfare, investigations of preventative strategies for diseases is greatly needed. Specifically in the cattle industry, infectious bovine keratoconjunctivitis (IBK), commonly known as pinkeye, causes a large economic loss to the producer. However, effective mitigation strategies are lacking. Partly, this is due to the lack of studies investigating the ocular microbiome. Simparica TRIO®, a canine endectocide manufactured by Zoetis, contains sarolaner, moxidectin, and pyrantel pamoate salt in an orally administered chewable tablet. These compounds are all toxic to both cats and dogs in sufficient amounts. As Simparica TRIO® rises in popularity among pet owners, Felis catus (felines, cats) in multi-pet households (households with both cats and dogs) are at increased risk of overdosage and toxicosis from accidental ingestion of Simparica TRIO® due to their smaller weight.

Project 1 focused on eight calves in a Nebraska herd (composite Simmental, Red Angus, Gelbvieh) that displayed exercise intolerance during forced activity. Available sire pedigrees contained a paternal ancestor within 2-4 generations in all affected calves. Pedigrees of the calves’ dams were unavailable, however, the cows were ranch-raised and retained from prior breeding seasons, where bulls used for breeding occasionally had a common ancestor. Therefore, it was hypothesized that a de novo autosomal recessive variant was causative of exercise intolerance in these calves. A genome-wide association analysis followed by whole-genome sequencing led to the identification of a variant in the …

Impact Of Strategic Postweaning Supplementation On March- And May-Born Yearling Range Heifer And Calf Growth And Performance In The Nebraska Sandhills, Josie Nicole Crouch

Three experiments were conducted at the Gudmundsen Sandhills laboratory near Whitman, Nebraska. Experiment 1, a retrospective regression analysis, evaluated the impact of heifer percentage of mature body weight (BW) at breeding on heifer performance, subsequent pregnancy rates, and calf production. Overall pregnancy rates were greater (P < 0.01) for heifers at 60 to 70% of mature BW than 50 to 55%, with no differences in pregnancy rates (P ≥ 0.15) after 2-years of age. Heifers at 50 to 60% of mature BW had a greater (P = 0.05) proportion give birth in the first 21-d. A higher percentage of mature BW at breeding resulted in greater (P < 0.01) BW at birth and weaning for offspring.

Pregnancy to artificial insemination and late gestation fetal programming of the next calf crop are central to genetic development, reproductive longevity, calf crop productivity, and efficiency of cow/calf ranching. The purposes of this research are to find methods to improve pregnancy rate to artificial insemination, the growth and reproductive health of pregnant heifers, and the productivity of their offspring. In the first of four studies we found that extending the period of progesterone administration in an estrus synchronization protocol did not significantly hasten estrus response or increase pregnancy rate to artificial insemination, but timing of estrus within the melengestrol acetate …

Introduction: Control of Campylobacter from farm to fork is challenging due to the frequent emergence of antimicrobial-resistant isolates. Furthermore, poultry production systems are known reservoirs of Campylobacter. The twinarginine translocation (Tat) pathway is a crucial bacterial secretion system that allows Campylobacter to colonize the host intestinal tract by using formate as the main source of energy. However, Tat pathway is also a major contributing factor for resistance to copper sulfate (CuSO4).

In this study, we utilized an in vitro model consisting of human malignant melanoma as well as non-tumorigenic immortalized keratinocyte cells with the aim of characterizing the therapeutic effectiveness of the clinical epigenetic drug Tazemetostat alone or in combination with various isothiocyanates. In doing so, we assessed markers of cell viability, apoptotic induction, and expression levels of key proteins capable of mediating the therapeutic response. Our data indicated, for the first time, that Tazemetostat caused a significant decrease in viability levels of malignant melanoma cells in a dose- and time-dependent manner via the induction of apoptosis, while non-malignant keratinocytes were …

To assess the susceptibility of elk (Cervus canadensis) and mule deer (Odocoileus hemionus) to SARS-CoV-2, we performed experimental infections in both species. Elk did not shed infectious virus but mounted low-level serologic responses. Mule deer shed and transmitted virus and mounted pronounced serologic responses and thus could play a role in SARS-CoV-2 epidemiology.

Myocarditis is one of the major causes of heart failure in children and young adults and can lead to dilated cardiomyopathy. Lymphocytic myocarditis could result from autoreactive CD4+ and CD8+ T cells, but defining antigen specificity in disease pathogenesis is challenging. To address this issue, we generated T cell receptor (TCR) transgenic (Tg) C57BL/6J mice specific to cardiac myosin heavy chain (Myhc)-α 334–352 and found that Myhc-α-specific TCRs were expressed in both CD4+ and CD8+ T cells. Proteolytic activation of the hemagglutinin (HA) glycoprotein by host cellular proteases is pivotal for influenza A virus (IAV) infectivity. Highly pathogenic avian influenza viruses possess the multibasic cleavage site of the HA which is cleaved by ubiquitous proteases, such as furin; in contrast, the monobasic HA motif is recognized and activated by trypsin-like proteases, such as the transmembrane serine protease 2 (TMPRSS2). Phenethyl isothiocyanate (PEITC) is a secondary metabolic product yielded upon the hydrolysis of gluconasturtiin and it is highly accumulated in the flowers of watercress. The aim of the current study was to assess the role of a naturally derived PEITC-enriched extract in the induction of oxidative stress and to evaluate its anti-melanoma potency through the regulation of its metabolism with the concurrent production of the N-acetyl cysteine conjugated by-product. In October 2020, a new population of invasive brown treesnakes (Boiga irregularis) was discovered on the 33-ha Cocos Island, 2.5 km off the south coast of Guam, United States. Cocos Island is a unique conservation resource, providing refuge for many lizards and birds, including endangered species, which were extirpated from mainland Guam by invasive predators including brown treesnakes. North American populations of Black Vultures Coragyps atratus have increased and expanded their distribution in the southern and eastern USA. In conjunction with these patterns has been a rise in human–vulture conflicts. To improve our understanding of space use patterns and better inform management, we evaluated the movements of Black Vultures (n = 23) in the southeastern USA using a long-term GPS tracking database. Our specific objectives were to: (1) quantify home-range sizes in relation to season and geographical study location and (2) examine within-home-range resource selection to identify landscape and anthropogenic factors influencing roost and diurnal space use. …

North American horned lizards of the genus Phrynosoma are largely myrmecophagous and squirt systemic blood from circumocular sinuses during encounters with certain mammalian predators. The predators react with distinct revulsion. Using bioassay-guided fractionation of blood plasma, we endeavored to identify the source and characteristics of the active chemical compound(s) responsible for this negative oral response. Our results of coyote bioassays and a series of novel mouse bioassay experiments were largely concordant. The active compound(s) was likely plasma-borne with a molecular weight (mw) of between 800 and 1,600. To identify the source of the active compound(s), we noted bioassay responses …

Feral swine (Sus scrofa) were targeted for and successfully eradicated from Saint Vincent Island (SVI), a National Wildlife Refuge (NWR) along the coast of Florida’s panhandle to protect its habitats and uncharacteristically high diversity of wildlife species for barrier islands in the region, including federal and state-listed threatened and endangered species. The eradication effort was initiated in early 2015 and concluded in 2019. A total of 438 feral swine were removed from the Island, 417 by federal control experts and 21 by recreational hunters. Temperature is a primary factor influencing organismal development, and the fluctuating daily and seasonal thermal regimes of temperate climates may challenge the ability of viviparous reptiles to optimize body temperatures during gestation. Globalization has led to the frequent movement of species out of their native habitat. Some of these species become highly invasive and capable of profoundly altering invaded ecosystems. Feral swine (Sus scrofa × domesticus) are recognized as being among the most destructive invasive species, with populations established on all continents except Antarctica. Migratory birds exhibit seasonal geographic range (hereafter, range) dynamics during the annual cycle. Few studies have examined how migratory birds select their habitats for range occupancy at the species level and space use at the individual level simultaneously. We hypothesized that environmental variables directly related to fitness components would affect the range occupancy probabilities of migrants, whereas environment variables related to movements and flights would affect the space use intensities of migrants. Density estimation for unmarked animals is particularly challenging, yet density estimates are often necessary for effective wildlife management. Raccoons (Procyon lotor) are the primary terrestrial wildlife reservoir for Lyssavirus rabies within the United States. The raccoon rabies variant (RRVV) is actively managed at landscape scales using oral rabies vaccination (ORV) within the eastern United States. This study was conducted to determine the accuracy of using 3D imaging technology as a method to predict shrunk body weight (BW) of growing yearling beef heifers. Red Angus x SImmental heifers (n = 69, BW = 726 ± 62 lbs; 12 months of age) were utilized for data collection. A time-of-flight depth camera (Azure Kinect, Microsoft) was used to collect depth videos as heifers walked out of the chute. An experiment was conducted to evaluate the effect of varying inclusion of modified distillers grains plus solubles on a weekly basis with two inclusions of grass hay on the performance of finishing steers. no interaction was observed between modified distillers grains plus solubles inclusion (0%, 25% constant, or 25% varying from 15-35%) and hay inclusion (6% or 12%). When evaluating the effect of hay inclusion on performance, cattle fed 6% grass hay had less dry matter intake than those fed 12% grass hay, and there was a tendency for gains to be greater for cattle fed 6% grass hay.
