Swine Acute Diarrhea Syndrome Coronavirus (SADS-CoV) is a member of the Coronaviridae family. The virus is associated with severe small intestine inflammation and diarrhea in suckling piglets. In 2017, SADS-CoV was first detected and identified as the causative agent of a devasting swine disease outbreak in southern China. Routine monitoring and early detection of the source of infection is therefore integral to the prevention and control of a SADS-CoV outbreak in the United States. However, the United States does not currently have any diagnostic or surveillance tests to identify this emerging disease. To address these industry needs, we developed monoclonal …

Senecavirus A (SVA) is a positive sense single-strand RNA virus of the genus Senecavirus, family Picornaviridae. Senecavirus A is an emerging picornavirus of swine, which causes vesicular disease (VD) in pigs that is clinically indistinguishable from footand- mouth disease and other high consequence VDs of swine. The cultivability and the potential role of rumen bacteria in the field of livestock production, host health and immunity are important to study. Porcine reproductive and respiratory syndrome virus (PRRSV) is the costliest swine disease in North America. Vaccines and management alone have not been effective in controlling this disease. Genetic selection for resilience may be a complimentary approach for controlling PRRSV. Senecavirus A (SVA) has been associated with several outbreaks of vesicular disease (VD) since 2014 in major swine-producing countries around the world. The virus causes VD that is clinically indistinguishable from foot-and-mouth disease (FMD) in pigs. Currently there are no commercial vaccines available for SVA. Here, we developed a live attenuated SVA strain by mutating a wild type SVA strain SD15-26 (wt SVA). The live attenuated virus, rSVA mSacII, replicated less efficiently in cell cultures in vitro and expressed lower amounts of VP1 and VP2 capsid proteins than wt SVA. Chronic wounds are longstanding dermal wounds that do not heal in a normal amount of time and can commonly last for over a year. These wounds plague humans and farm animals alike. Wounds are characterized by chronic inflammation and are often painful, difficult to heal, have a high recurrence rate, and are potentially debilitating. Pig weaning process results in reduced growth performance and gut health of weaned pigs. Antibiotics can be added in diets for weaned pigs to improve growth performance and gut health, but their use is being discouraged because they can lead to development of antimicrobial resistant microorganisms. Swine influenza A viruses pose a serious concern for global health and worldwide swine industry considering their evasion from vaccine-induced immune responses. Formulation of effective influenza vaccines can be complicated by both antigenic shift to a different subtype of hemagglutinin (HAs) and antigenic drift within a particular HA subtype. To address these concerns, we employed a systematic approach to determine the antigenic determinants of swine influenza A viruses by focusing on H1 and H3 subtypes. Bovine viral diarrhea virus (BVDV) and bovine herpesvirus type 1 (BHV-1) are important pathogens that affect cattle. In this in vitro study, the main goal was determining the direct effects of BVDV and BHV-1 infections of monocyte-derived dendritic cells (MDDC) on cytokine mRNA expression. For the BVDV infected MDDC experiments, Holstein Friesian and Brown Swiss calves were included with a breed comparison with cytokine expression. Surface marker expression was also measured in BHV-1-infected MDDCs. Effects of enzymatically hydrolyzed yeast (EHY) and supplementation frequency (FREQ) on immune parameters among beef cows and calves was evaluated. Eighty multiparous (parity = 4.2 ± 0.3) cows were fed a common brome hay-based diet (CP = 8.0 ± 0.17%). Cows were blocked by expected calving date and stratified by body condition score (BCS) before random assignment of treatment. Beginning 88 ± 5 d prior to parturition, cows were provided 1 kg daily or 3 kg every 3 d of a soybean hull-based supplement (CP = 34.0, % of DM) that contained 0 or 3 g/kg EHY. BVDV is an important pathogen of cattle that affects both the dairy and beef industry causing severe economic losses. The main problem of BVDV is the immune suppression that is associated with lymphoid depletion shortly post infection. The mechanism of this immune suppression is not well understood. In this in vitro study, we sought to determine the indirect effect of BVDV infection in macrophages as a key player in the pathogenesis of BVD especially, the apoptotic effect induced on lymphocytes. Porcine reproductive and respiratory syndrome virus (PRRSV) is an economically devastating virus that causes a persistent infection and spreads quickly. The virus causes abortions in sows and respiratory diseases in young piglets. Current PRRSV vaccines appear to provide limited cross-protection. A virus-like particles (VLPs) vaccine is made up of key PRRSV structural proteins, which could be cross protective among the different strains. We tested PRRSV VLPs as a vaccine candidate with a 2’, 3’-cGAMP VacciGradeTM adjuvant against pigs infected with PRRSV after intranasal immunization to evaluate the immune response and protective efficacy by the VLPs and adjuvant. The purpose of this project was to define monoclonal antibodies against viral proteins from Epizootic Hemorrhagic Disease Virus (EHDV) to be used in quality control of vaccines produced by an industrial partner, and development of tools to identify EHDV. EHDV is an Orbivirus, which also includes Bluetongue Virus and African Horse Sickness, which are all transmitted through arthropod vectors. However, there have been only a few reports of SVA associated with disease in last two decades. In late 2014 early 2015, a sudden increase in the number of cases of Senecavirus A associated with vesicular disease was observed in Brazil, the US and China. Development And Characterization Of A Recombinant Orf Virus Vector Expressing The Spike Protein Of Porcine Epidemic Diarrhea Virus, Kyle Hain

Orf virus (ORFV), the type member of the genus Parapoxvirus of the family Poxviridae, causes orf or cutaneous pustular dermatitis in sheep and goats. ORFV is a ubiquitous virus capable of re-infecting its hosts multiple times over time. ORFV causes a non-systemic, self-limiting disease which is usually restricted to the skin surrounding the virus entry sites. ORFV has evolved several immunomodulatory proteins (IMPs) that evade and/or modulate host immune responses to infection and contribute to virus virulence and disease pathogenesis.
