Comparative Potency Testing Of Diphtheria Toxoid In Guinea Pigs And Mice For In-House Diphtheria-Tetanus-Pertussis And Diphtheria-Tetanus Vaccines, Wanlop Leeden, Supanee Ummalukacholk, Orawin Mangkhaladul, Eakachai Prompetchara

The Thai Journal of Pharmaceutical Sciences

Background: Diphtheria remains endemic in parts of Thailand and continues to threaten unvaccinated populations. To strengthen vaccine self-sufficiency, the Government Pharmaceutical Organization has developed in-house diphtheria–tetanus–pertussis (DTP) and diphtheria– tetanus (dT) vaccines. Potency testing of diphtheria toxoid is traditionally performed in guinea pigs; however, limitations in animal availability, cost, and logistics have prompted evaluation of alternative models.
Objectives: To compare diphtheria toxoid potency estimates of in-houseproduced DTP and dT vaccines using guinea pigs and mice, followed by a Vero cell-based toxin neutralization assay, and to assess the suitability of mice as an alternative model for potency testing.

The Thai Journal of Pharmaceutical Sciences

Background: Aripiprazole (ARI), a biopharmaceutics classification system (BCS) class IV drug, exhibits poor water solubility and low permeability, resulting in limited bioavailability. Addressing these challenges requires innovative approaches to enhance solubility. This study focuses on the development and evaluation of solid dispersions (SDs) of ARI using “hot-melt extrusion,” (HME) a scalable and solvent-free method known for its ability to create stable amorphous structures. Soluplus® (SP) and Kollidon® 30 were employed as hydrophilic polymeric carriers to improve the solubility and stability of the amorphous drug form. Results: SDs were prepared by taking drugs and carriers (SP and Kollidon …

The Thai Journal of Pharmaceutical Sciences

Background: Platelet aggregation plays a crucial role in hemostasis; however, excessive platelet activation can lead to thrombus formation, contributing to cardiovascular diseases. While aspirin (acetylsalicylic acid [ASA]) is commonly used as an antiplatelet agent, its longterm use is associated with gastrointestinal side effects. Angelica dahurica (AD), a traditional herbal remedy, has shown in vitro antiplatelet activity and cyclooxygenase enzyme inhibition. However, its in vivo effects on platelet aggregation and gastric safety remain unclear.

Objectives: This study aimed to investigate whether Angelica dahurica root extract (ADE) could inhibit platelet aggregation and prevent aspirin-induced gastric lesions.

Materials and Methods: AD was extracted …

Concurrent Use Of Skeletal Muscle Relaxants And Nsaids In Low Back Pain Management: A Critical Review Of Current Evidence And Future Directions, Rupesh A. Saindane, Nilima A. Thombre

The Thai Journal of Pharmaceutical Sciences

Low back pain (LBP) represents a leading cause of non-fatal disability globally. Its high prevalence and disabling nature highlight the urgent need for effective management strategies. The exact cause of LBP still remains unclear, and thus, its treatment remains symptomatic. LBP is commonly treated with non-steroidal anti-inflammatory drugs (NSAIDs) and skeletal muscle relaxants (SMRs). These agents are frequently used in combination to target both inflammation and muscle spasms. However, the clinical guidelines vary widely on the recommendation for the use of such combination therapy. The present review evaluates evidence from randomized clinical trials and systematic reviews on the concurrent use …

A Review Of Overlapping Severe Cutaneous Adverse Reaction, Ploylarp Lertvipapath, Roongrat Piyananjaruhsri, Diana Karimee

The Thai Journal of Pharmaceutical Sciences

Severe cutaneous adverse reactions (SCARs) are life-threatening drug hypersensitivity reactions, including acute generalized exanthematous pustulosis (AGEP), drug reaction with eosinophilia and systemic symptoms (DRESS), Stevens–Johnson syndrome (SJS), and toxic epidermal necrolysis (TEN). SCARs occur in approximately 0.1% of hospitalizations and have mortality rates ranging from 5% for AGEP to 5–35% for SJS/TEN. AGEP typically presents with acute erythematous areas, sterile pustules, fever, and neutrophilia within 48–72 h of drug exposure. In contrast, DRESS manifests as fever, facial edema, morbilliform rash, and exfoliative dermatitis, often accompanied by hematologic abnormalities and multi-organ involvement, including hepatitis, pneumonitis, and myocarditis. Moreover, SJS and TEN …
