Roseofungin, a polyene macrolide antibiotic produced by Streptomyces roseoflavus, was evaluated for antiviral activity against Influenza A virus using complementary in vitro, in ovo, and preliminary in vivo toxicity models, supported by molecular docking and molecular dynamics simulations. The compound exhibited antiviral activity against multiple influenza A virus subtypes, including an oseltamivir-resistant H1N1 strain, with EC50 values ranging from 2.0 to 2.5 μg/mL and selectivity indices of 20–25. Functional assays demonstrated that roseofungin interfered with early stages of the viral life cycle by reducing viral adsorption and partially inhibiting neuraminidase activity. Efficacy of Ferula L. Species Extracts from Tajikistan Against Influenza Viruses One of the challenges facing both the domestic and international pharmaceutical industries is the limited selection and suboptimal efficacy of available antiviral drugs. The high mutational variability of influenza viruses contributes to rapid acquisition of drug resistance, which diminishes or nullifies the effectiveness of existing therapies, thereby necessitating the exploration for novel therapeutics. In Tajikistan, the Allium genus is notably prolific, with over 80 species documented, and new species continually being identified in the mountainous regions. The Gorno- Badakhshan Autonomous Region (GBAO) of Tajikistan, within the Pamir Mountains, is significant for its 31 distinct Allium species, including endemics like A. afghanicum Wendelbo and A. schugnanicum Vved. The distribution of these species is often geographically restricted, and their biological activities, especially their antimicrobial properties, remain largely unexplored, presenting significant opportunities for future phytochemical and pharmacological research. Objective: To evaluate the spectrum of antibacterial activity of plants growing in the central part of the Republic of Tajikistan.
Methods: The antibacterial properties of 18 plant species from the 14 families were studied. The extract samples on dry filter discs were prepared according to the method developed at Rutgers University. Screens-to-Nature (STN) and disc diffusion (DD) methods were used to assess antimicrobial properties of the extracts. Juzen-taiho-to (JTT) is an immune-boosting herbal formulation with an ideal balance of safety and efficacy. For example, JTT is clinically used in Japan to stimulate the immunological functions of cancer patients undergoing chemotherapy and radiation. Although the clinical effects of JTT are recognized, the active compounds and mechanism of action are unknown. The studies conducted previously in our laboratory associated several phytosterols and plant glycolipids with the potent immunostimulatory activity of JTT: namely, β-sitosteryl β-D-glucoside (BSSG), glucocerebroside (GluCer), digalactosyldiacylglycerol (DGDG) and monogalactosyldiacylglycerol (MGDG).
