Characterizing The Cutaneous Microbiome Of Eurycea Lucifuga As A Potential Defense Against Chytridiomycosis, Madeline Key Dec 2022

Characterizing The Cutaneous Microbiome Of Eurycea Lucifuga As A Potential Defense Against Chytridiomycosis, Madeline Key

Senior Honors Theses

Chytridiomycosis is an emerging infectious disease that is significantly reducing global amphibian populations. The disease is caused by Batrachochytrium dendrobatidis (Bd), a fungus that lethally modifies amphibian skin. Recent research has suggested that the cutaneous microbiome of individual amphibians may play a role in susceptibility to the pathogen. In this study, twelve cave salamanders (Eurycea lucifuga) were collected. Cutaneous bacteria from each salamander were isolated and identified using Sanger Sequencing. Additionally, a Bd-challenge assay was performed to determine each isolate’s antifungal activity. Results indicated many microbial isolates possessed inhibitory capabilities against Bd, which may …

A Paraventricular Thalamo-Accumbal Circuit Necessary For Behavioral Inhibition Is Dysfunctional During Opioid Use And Dependence, Kelsey M. Vollmer Sep 2022

A Paraventricular Thalamo-Accumbal Circuit Necessary For Behavioral Inhibition Is Dysfunctional During Opioid Use And Dependence, Kelsey M. Vollmer

MUSC Theses and Dissertations

Suppression of dangerous or inappropriate reward-motivated behaviors is critical for survival, whereas therapeutic or recreational opioid use can unleash detrimental behaviors and opioid use disorder (OUD). Nevertheless, the neuronal systems that suppress maladaptive motivated behaviors remain unclear, and whether opioids disengage those systems is unknown. Here, we investigate the posterior paraventricular thalamus (pPVT) as a behavioral mediator due to its strong connectivity with brain structures important for reward processing, such as the nucleus accumbens (NAc). We and others have found pPVT-NAc neuronal activity is inhibited during reward seeking, but activated when such behaviors would be inappropriate, such as during stress …

Integration Of Mouse Ovary Morphogenesis With Developmental Dynamics Of The Oviduct, Ovarian Ligaments, And Rete Ovarii, Jennifer Mckey, Dilara N Anbarci, Corey Bunce, Alejandra E Ontiveros, Richard R Behringer, Blanche Capel Sep 2022

Morphogenetic events during the development of the fetal ovary are crucial to the establishment of female fertility. However, the effects of structural rearrangements of the ovary and surrounding reproductive tissues on ovary morphogenesis remain largely uncharacterized. Using tissue clearing and lightsheet microscopy, we found that ovary folding correlated with regionalization into cortex and medulla. Relocation of the oviduct to the ventral aspect of the ovary led to ovary encapsulation, and mutual attachment of the ovary and oviduct to the cranial suspensory ligament likely triggered ovary folding. During this process, the rete ovarii (RO) elaborated into a convoluted tubular structure extending …

How Environmental Change Will Impact Mosquito-Borne Diseases, Arsal Khan May 2022

How Environmental Change Will Impact Mosquito-Borne Diseases, Arsal Khan

Master's Projects and Capstones

Mosquitos, the most lethal species throughout human history, are the most prevalent source of vector-borne diseases and therefore a major global health burden. Mosquito-borne disease incidence is expected to shift with environmental change. These changes can be predicted using species distribution models. With the wide variety of methods used for models, consensus for improving accuracy and comparability is needed. A comparative analysis of three recent modeling approaches revealed that integrating modeling techniques compensates for trade-offs associated with a singular approach. An area that represents a critical gap in our ability to predict mosquito behavior in response to changing climate factors, …

Extracellular accumulation of amyloid-beta (Aβ) plaques is one of the major pathological hallmarks of Alzheimer's disease (AD), and is the target of the only FDA-approved disease-modifying treatment for AD. Accordingly, the use of transgenic mouse models that overexpress the amyloid precursor protein and thereby accumulate cerebral Aβ plaques are widely used to model human AD in mice. Therefore, immunoassays, including enzyme-linked immunosorbent assay (ELISA) and immunostaining, commonly measure the Aβ load in brain tissues derived from AD transgenic mice. Though the methods for Aβ detection and quantification have been well established and documented, the impact of the size of the …

Cannabinoids And Retinal Fibrotic Disorders., Lucy June Sloan May 2022

Cannabinoids And Retinal Fibrotic Disorders., Lucy June Sloan

Electronic Theses and Dissertations

Retinal fibrosis is detrimental to vision. Retinal pigment epithelial (RPE) cells contribute to several retinal fibrotic diseases. Upon exposure to TGF-β, a key fibrotic cytokine, RPE cells trans-differentiate to myofibroblasts marked by the integration of α-SMA fibers into F-actin stress fibers, which confer strong contractility. Myofibroblasts produce and contract the collagen-rich fibrotic scar and disrupt retinal architecture. In this study, we investigated the in vitro effects of the putative endocannabinoid compound N-oleoyl dopamine (OLDA) on TGF-β2 induced porcine RPE cell contraction and α-SMA expression. Using an in vitro collagen matrix contraction assay, we found that OLDA inhibited TGF-β2 induced contraction …

Dental Morphology In A Juvenile Mammal From The Late Cretaceous Of South America: Tooth Eruption And Development., Kayla E Newton May 2022

Dental Morphology In A Juvenile Mammal From The Late Cretaceous Of South America: Tooth Eruption And Development., Kayla E Newton

Electronic Theses and Dissertations

Meridiolestids are a diverse and distinct clade of Cretaceous–Miocene South American dryolestoid mammals ranging in size from small shrew-sized insectivores to large dog- sized omnivores. Cronopio dentiacutus from the Cenomanian La Buitrera locality, Rio Negro Province, Argentina (Candeleros Fm.) is the oldest and best known of them, represented by skull material and associated jaws. A juvenile skull as attributed to Cronopio dentiacutus, based on tooth size and morphology, was collected from the type locality. The specimen includes the skull and both articulated lower jaws. The fossil is poorly ossified and partially flattened; CT scanning reveals most of the dentition …

Strong Inferences About Pain In Invertebrates Require Stronger Evidence, Edgar T. Walters Jan 2022

Strong Inferences About Pain In Invertebrates Require Stronger Evidence, Edgar T. Walters

Animal Sentience

Evidence for sentience in animals distantly related to humans is often sought in observations of behavioral and neural responses to noxious stimuli that would be painful in humans. Most proposed criteria for painful sentience in “lower” animals such as decapod crustaceans have no necessary links to the affective (“suffering”) component of pain. The best evidence for painful affect in animals is learned aversion to stimuli associated with noxious experience, and conditioned preference for contexts associated with relief from aversive consequences of noxious experience, as expressed in voluntary behavior. Such evidence is currently lacking for any invertebrate except octopus.

Deoxyribose Phosphate Aldolase Is Required For Development Of Hematopoietic Stem Cells In The Zebrafish Embryo, Anubhav Thapaliya, Sanyam Patel, Teerth Y. Patel, Sivam J. Bhatt, Erich W. Damm Jan 2022

Deoxyribose Phosphate Aldolase Is Required For Development Of Hematopoietic Stem Cells In The Zebrafish Embryo, Anubhav Thapaliya, Sanyam Patel, Teerth Y. Patel, Sivam J. Bhatt, Erich W. Damm

Undergraduate Research Posters

Hematopoietic stem cells (HSCs) are an integral part of modern biomedical research because they are precursors to all blood cell lineages in vertebrates. Further understanding of hematopoietic development will allow for improvements in the success of therapeutic bone marrow transplantation; which is a crucial treatment in management of both malignant and non-malignant blood disorders. Although HSC transplantation is curative for many blood disorders, finding compatible donors is difficult due to a need for immunologic compatibility. Consequently, recent research efforts have been directed towards generating HSCs for the patient, from the patient themselves through the directed differentiation of induced pluripotent stem …

No Difference In Corticosterone Concentrations Between Missouri Three-Toed Box Turtles Living In An Urban And A Rural Site, Bennett A. Lamczyk, Jamie L. Palmer, Corinne P. Kozlowski, Stephen Blake, Sharon L. Deem Jan 2022

Deem

Human–Wildlife Interactions

Baseline health data for species of conservation concern are important for understanding threats to the long-term viability of populations. One indication of health is physiological stress among individuals. Corticosterone (CORT) is frequently used to quantify stress in free-living reptile populations, as high values may be associated with reduced fitness. Herein, we describe and validate methods for quantifying blood CORT levels in three-toed box turtles (Terrapene mexicana triunguis). We subsequently use this information to evaluate stress levels in 2 populations of free-living three-toed box turtles in Missouri, USA. To our knowledge, this is the first quantification of CORT levels …
