The best understood functional activity of the pineal gland is its diurnal production of melatonin in response to environmental lighting cues. Several enzymes of the melatonin pathway respond to daily photoperiod changes, for example hydroxyindole-O-methyltransferase (HIOMT) and serotonin N-acetyltransferase (SNAT). Increased levels of the glycolytic enzyme neuron-specific enolase (NSE) are thought to reflect increased physiological demands placed on neurons and neuroendocrine tissues. Homodimer non-neuronal enolase isoenzyme (NNE) is immunolocalized to cells, and the hybrid enolase (consisting of subunits from NSE and NNE) has been seen in cerebellar stellate and basket cells. Several monoclonal antibody (MAb) types derived from mice have been developed specifically for the use of targeting human tumors. The specificity of these murine MAb’s for their respective tumor antigens is very high making them potentially good immunotherapeutic agents for cancer treatment and diagnosis. A limiting factor in using these murine MAb’s in patients is the development of human anti-mouse antibody (HAMA). The presence of HAMA may reduce the effectiveness of MAb for tumor targeting. If patient sensitization could be quickly detected following the initial infusion of MAb, then future infusion could be altered or terminated to prevent further sensitization. …

The effect of autonomic nervous system on cholesterol transport is of clinical interest because the relationship between lipoprotein metabolism and atherogenesis is expressed in coronary artery disease.
Generally speaking, beta blockers increase serum VLDL level but decrease both HDL and HDL2 cholesterol levels without affecting the LDL in humans. We are unaware of reports concerning the effects of metoprolol on lipoprotein metabolism nor comparison of the effects of metoprolol and propranolol in the unhandled rat model. The root canal system, which includes the periapical region of the tooth, may become infected by several types of microorganisms which are normally associated with these tissues. A bacteria-specific antibody response may occur at the local level, where various classes of immunoglobulins have been found and reported in the literature. The purpose of this study was to quantitatively measure locally produced immunoglobulin' levels in periapical lesion abscess materials and to determine the specific reactivity of these antibodies to several microorganisms which are normal flora, as well as being previously associated with the development and pathogenesis of endodontic infections. The highly selective permeability of cerebral blood vessels, termed the blood-brain barrier, is due to endothelial tight junctions (zonulae occludentes). Hyperosmotic solutions reversibly open the blood-brain barrier. The purpose of this study is to determine the immediate effect of hyperosmotic arabinose perfusion upon the cerebrovascular endothelium, the morphology of the cerebral cortex, and the cortical extracellular space.
Twelve adult male Sprague-Dawley rats were separated into three groups of four animals each: control, intermediate, and extreme. This thesis is based on a semiquantitative analysis of four nuclear relay centers [nucleus tractus solitarius (NTS), parabrachial nucleus (PBN), ventroposteromedial nucleus (VPM), gustatory neocortex (GNC)] in the chorda tympani nerve pathway. Metabolic changes were examined by the 2-deoxyglucose method in which one chorda tympani nerve was chemically stimulated and its contralateral side lesioned. Coronal sections of four taste centers were prepared for autoradiography and Nissl staining. Densities of the autoradiograms were determined by means of a video based image processor. Continuous administration of estrogens for 7-9 months, both steroidal and nonsteroidal, to male BALB/c mice, leads to the formation of testicular Leydig cell tumors. Three days following the subcutaneous implantation of a pellet of estrogen in cholesterol, there is a peak in the incorporation of 3H-tymidine into the DNA of the interstitial cells. These effects are hypothesized to be mediated by the estrogen receptor system in the Leydig cell. Common experimental techniques for the measurement of hormone binding, such as dextran coated charcoal treatment, proved to be impossible to employ in this system, therefore a procedure was developed using …

The existence of extrathyroidal monoiodotyrosine (MIT) has been debated because of uncertain results from measuring serum MIT. The reason is that the isotopic and chromatographic assays used have not been sensitive enough. This investigation undertook to establish the existence of MIT in serum utilizing a radioimmunoassay (RIA), to study the physiology of serum MIT, and to discover the pharmacologic effects of exogenous MIT.

Eight healthy male lacto-ovo-vegetarian subjects averaging 28.0 ± 3.6 years of age (22 to 53 years) were fed a diet rich in α-linolenic acid (18:3 ω 3) for four weeks. Arterial blood pressure and body weight were recorded each week. Plasma 6-keto-prostaglandin F1α (6-keto- PGF1α ), thromboxane B2 (TXB2), total cholesterol, triglyceride, and total phospholipid fatty acid composition were analyzed at 0, 2 and 4 weeks.
Systolic blood pressure increased significantly between weeks 0 and 1 (106.5 ± 2.7 mmHg to 114.0 ± 3.7 mmHg; p < 0.003), but decreased to 107.3 t 2.6 mrHg (p < 0.02) at week 2. There was an insignificant increase in both systolic and diastolic blood pressures after four weeks. There was no significant change in either total cholesterol or triglyceride levels during the four weeks.

The zinc status of twelve lacto-ovo-vegetariah pregnant women and seventeen non-vegetarian pregnant women was evaluated. Zinc intakes were calculated from three-day dietary records and twenty-four hour dietary histories. Plasma zinc levels were drawn during the last trimester of pregnancy and approximately ten weeks postpartum. Gestational age at birth, birthweight, and APGAR scores of the infants born were obtained from the medical charts. Dietary zinc intakes for the vegetarians (10.5 ± 4.0 mg) did not differ significantly from the intakes of the non-vegetarians (10.9 ± 3.0 mg). This model describes the orthoiodohippurate (OIH) distribution and clearance in the renovascular system. The model is described by five compartments: the blood, left and right kidneys, bladder and the red blood cell compartment. Data for these compartments, except the RBC compartment, are collected with scintillation detectors monitoring OIH tagged with 1-131 as the radiopharmaceutical passes through each compartment. Time/ activity curves are plotted for the data and used as inputs to the model parameter estimation routine. The compartments are described by a set of first order ordinary differential equations solved using the Adam's methods. For parameter estimation, an iterative predictor-corrector …

The urinary excretion of 3-Methylhistidine was used as an index of muscle catabolism in four groups of obese persons receiving a Protein Sparing Modified Fast (PSMF). The composition of the supplements were as follows: Group I: 75 g protein, 25 g complex carbohydrate (starch) per day; Group II: 25 g protein, 75 g complex carbohydrate (starch) per day; Group III: 75 g protein, 25 g simple carbohydrate (glucose) per day; Group IV: 25 g protein, 75 g simple carbohydrate (glucose) per day. Stress of handling, partial or complete sialoadenectomy, and the administration of parotid hormone (PH), sodium fluoride (NaF), or guanidino-succinic acid (GSA) were examined for effects on fasting plasma glucose and insulin or effects on intravenous glucose tolerance in male Sprague-Dawley rats. Basal glucose and insulin were substantially elevated as a result of the stress of translocation in large animals, but were not altered in small animals. A rise in plasma insulin occured in control animals 20 minutes following anesthesia and the removal of a blood sample by cardiac puncture but not in animals that had been handled twice daily for …

A computer model of the left ventricular (LV) epicardial surface was developed from Jang’s truncated ellipsoidal model, which has a minor to major axis ratio of 6:10. The model was applied in calculating the area of a segment of the myocardium, jeopardized by an impending occlusion of its associated artery. The endangered segment of the LV muscle was also visualized in different projections by rotations in three space and graphical displays simulating the three dimensions.

These studies explore the distribution of blood flow among small samples of placental tissue. Labeled microspheres (14 ± 1 pm) were Injected Into the left ventricle of six un-anesthetized ewes and Into the jugular vein of the fetus In utero. A total of 3,576 samples weighing 31. +11. (S.D.) mg were taken from seventeen cotyledons and counted for flow labels. Maternal and fetal flow within the cotyledons were both normally distributed with standard deviations of 44%. Plasma samples were obtained from seven hypertriglyceridemic (>150 mg/dl) chronic renal failure patients before and after regular hemodialysis. The effect of the plasma on the lipolytic activity of bovine milk lipoprotein lipase was investigated. The lipoprotein lipase activity, as measured by release of C14-labeled free fatty acid from triglyceride, was decreased 8-67% when incubated with uremic plasma as compared to the control plasma (44 mg/dl triglyceride). The inhibition noted prior to hemodialysis did not appear to be alleviated by the dialysis but subsequent treatment with an isolated ultrafiltration technique did appear to remove an inhibitory substance as …

A study was undertaken to determine the concentration at which a sealer reaches a point of nontoxicity. An in vitro model utilizing HeLa cells and human fibroblasts was used to observe the effect of various concentrations of eight freshly mixed and eight set root canal sealers. Evaluation of toxicity was determined by cell survival which was measured by incorporation of 3H-thymidine into the nucleic acid components of each cell type used. Each dilution tested was allowed direct contact with the cells for 24 hours before toxicity evaluation. The results indicate that all sealers at concentrations presently used are cytotoxic …

Despite extensive clinical and experimental investigations of venous thrombosis, relatively little is known about the early phases of healing of the venous thrombus. Soon after its discovery heparin has become the most popular agent for the prevention and treatment of venous thrombosis, but there are a number of unanswered questions regarding the most satisfactory treatment for venous thrombosis.

Speculation: In the fetal lamb during prolonged intrauterine hypoxia, total and regional cerebral blood flows increase to the same extent without evidence of preferential shunting to critical brainstem or subcortical areas. Neuropathologic studies have indicated relative sparing of these areas during similar animal experimental or human neonatal conditions. This suggests that the pattern of hypoxic ischemic insult to the neonatal central nervous system associated with asphyxia may differ from that produced by hypoxia alone. Cell mediated immune (CMI) cytotoxic reactivity of Balb/c mice against H238 cells, a Herpes simplex virus (HSV) Type 2 - induced sarcoma, was measured by the 125IUdR release assay. The Balb/c mouse response to the growing HSV-induced tumor, treated with radiation and , was measured by survival rate, tumor growth and immune cytotoxicity of spleen and peritoneal exudate cells (PEC) as determined by 125IUdR release assay. Subcutaneous (s.c.) inoculation of 1 x 106 H238 cells (high dose) produced progressive tumor growth while s.c. inoculation of 1 x 104 H238 cells (low dose) produced no tumors. A …
