To ensure culturally-sensitive assessments, the researcher designed a Pediatric Breathing Survey (PBS) to measure quality of life for African-American children. The purpose of this study was to test whether the PBS is a reliable and valid instrument. Comparisons of group differences on race, income and parent's level of education based on parent responses to the questionnaires were conducted. More specifically, differences in Health Related Quality of Life (HRQOL) between Caucasian and African-American children were examined. The researcher-designed instrument was compared to Juniper's Pediatric Asthma Quality of Life Questionnaire. The sample consisted of parents of 84 school-aged children diagnosed with asthma. …

The Behavior Model of Utilization played a key role in explaining health services use for pediatric members with asthma in one urban, regional managed care company. The critical role the asthma management program played in promoting the health and well-being of enrolled members was elucidated. Information gleaned from this empirical study can be used to encourage appropriate resource utilization, formulate pediatric asthma health policy, and deliver cost-effective, quality health care. Asthma is the most common chronic illness of childhood. Pediatric asthma hospitalization rates and costs of care have risen in recent years. Readmissions, an adverse outcome often used to monitor quality of care, may account for the increase in asthma hospitalization rates. This research examined selected hospital and patient risk factors related to asthma readmissions, using Donabedian's framework for structure, process, and outcomes to assess the quality of care for children with asthma. Outcome measures included readmission for asthma within one year of initial hospitalization, readmission within 30 days, and multiple readmissions within one year. Human T-cell leukemia virus type 1 (HTL V-1) is dependent upon the enzymatic activity of its protease for maturation. Maturation of the protease is facilitated by cleavage of specific amino acid residues, followed by dimerization. The effects of the amino acid sequence located N-terminally to the cleavage site on the ability of the protease to become active were the focus of the current study. These amino acid sequences were contributed by the plasmid vector into which the protease gene was inserted.

The most prevalent life-threatening disease worldwide, secondary to coronary artery thrombosis is Acute Myocardial Infarction (AMI). Increases in three clinical markers have been explored as an early means of AMI detection: Cardiac myoglobin, and the two specific cardiac isoforms of Creatine Kinase, CK-MM and CK-MB. However, myoglobin levels have shown to give a more accurate correlation (R = 0.89) than does Creatine Kinase evaluation. Therefore, we have developed a two-site Enzyme-Linked Immunosorbent Assay (ELISA) using monoclonal antibodies against myoglobin isolated from human heart tissue. Currently, AIDS is the leading cause of death amongst young people in the United States. To that effect, Panavirocide was conceived by Medicine and Applied Science·s, Inc. Recently, it has been reported that human type I iodothyronine deiodinase, an enzyme important in the conversion of T4 to T3 , is a selenoenzyme. Several studies have reported alterations in plasma selenium level in hypo- and hyperthyroid patients.
Using polarized Zeeman-effect atomic absorption spectroscopy, we measured selenium, zinc, copper and manganese concentrations in the plasma and/or red blood cells in children with or without thyroid disease being seen at the outpatient clinic at Children's Hospital of the King's Daughters. Gout is a chronic metabolic disorder caused by deposition of uric acid on the joint. It is categorized into two types: primary and secondary. Primary gout is uric acid overproduction, caused by excessive synthesis of the purine precursors. Secondary gout occurs also as the result of overproduction or decreased renal excretion of uric acid resulting from other disease processes or medication.

The human immunodeficiency virus (HIV) reverse transcriptase (RT) (EC 2.7.7.49) is the central replication enzyme for HIV. In general, the kinetic mechanism for this and all other polymerases involves the ordered binding of two substrates: a primer-template (PT) followed by a deoxyribonucleoside triphosphate (dNTP). Previous investigations prompted this research when it was discovered that the substrate dNTP, in absence of PT, could protect a recombinant c-terminal mutant HIV-1 RT from inhibition by pyridoxal-5'-monophosphate (PLP), an active-site dNTP inhibitor. The purpose of this study was to explore the concept of coping with a cystic fibrosis (CF) child and to define coping strategies from the perspective of the mother. The research design was based upon a phenomenological approach with analysis of transcribed, semi-structured interviews with mothers of CF children. A purposive sample of eight mothers with CF children were interviewed to explore the maternal CF experience. Each interview was tape recorded and followed up with a phone call to allow the mothers to add additional information, if so desired. Motivation of clients with Peripheral Vascular Disease (PVD) to adhere to Recommended risk-reducing behaviors (RRB’s) was explored. The sample (n =30) was comprised of clients in a vascular physician group’s practice with a medical diagnosis of PVD. A nonexperimental, descriptive design was utilized to investigate how much variation in adherence to RRB’s by PVD clients could be accounted for by the client’s perceived effectiveness of a RRB and the client’s perceived difficulty in adhering to a RRB. Drugs such as adrenoceptor antagonists and certain centrally acting agents are known to lower blood pressure and thus serve as clinically useful antihypertensive agents. Norepinephrine 4, a natural ∝-adrenoceptor agonist, is the major agent associated with increased blood pressure in the periphery. Based on a careful structure-activity comparison, a series of substituted oxazolidines, 20, 21, 22 and 23, have been designed as possible antihypertensive candidates. Specifically, 22 and 23 have been synthesized to serve as potential norepinephrine antagonists.

Trace element levels in blood from diabetic children were investigated and compared to control children. Plasma, erythrocyte, mononuclear and polymorphonuclear cells were analyzed for magnesium, copper, and zinc content. Leukocyte separation was performed by density gradient centrifugation on Ficoll-Hypaque, followed by washing steps to remove platelet contamination and hypotonic lysis to remove red cell contamination. Polarized Zeeman-effect atomic absorption spectroscopy was used for trace metal analysis.
Decreased plasma magnesium levels were observed in the diabetic children. This finding is well documented and probably due to increased urinary magnesium loss from glycosuria. Lymphocyte magnesium levels in the diabetic children were also …

The effect of a cardiac education program in terms of changes in knowledge, state anxiety and stress were examined for persons suffering a myocardial infarction for the first time. In addition, the relationship between stress and trait anxiety and changes in knowledge and state anxiety were examined. A sample of 20 patients were selected from two medium sized hospitals in Southeastern Virginia. Statistically significant differences at the .01 level were found between the means of the pretest and posttest scores of the experimental group for knowledge and state anxiety using the t test for dependent groups. Human serum high density lipoprotein subclasses, HDL2 and HDL3 , were isolated by preparative salt density gradient ultracentrifugation and further analyzed by electrophoresis on a 4 to 15% concentration gradient polyacrylamide gel.
The separation of the major classes of serum lipoproteins was achieved after a single ultracentrifugation for 272,000 g at 15°c in a swinging bucket rotor. High resolution concentration gradient gel electrophoresis was found to be particularly suitable for the separation of lipoproteins. The isolation of HDL by precipitation methods, heparin-manganese and dextran-magnesium have also been evaluated in this study, and were found not to completely separate …

Duck plague virus causes a hemorrhagic disease affecting the family Anatidae, resulting in 60-80% mortality in captive flocks. Plaque and neutralization assays for Anatid herpesvirus (AHV) require 5-10 days for completion and are restricted by the seasonal availability of fertile duck eggs. The enzyme linked innnunosorbent assay (ELISA) is rapid and specific and avoids the use of living cells. The Holland strain of AHV was grown on duck embryo fibroblasts and purified by banding on CsCl gradients. The resulting virus was used to raise antisera in prebled New Zealand white male rabbits. Duck plague is a hemorrhagic, necrotic disease found exclusively in the order Anseriformes, family Anatidae (ducks, geese, swans). The etiologic agent has been reported to be a herpes-like virus. The Holland strain of duck plague was found to induce cytopathic effects in cell culture (perinuclear vacuolation followed by lysis). A simple reproducible plaque assay for quantitating the virus was developed. Under conditions of the plaque assay, one-hit kinetics were observed. The host range of duck plague virus in cell culture was found to include the order Galliformes, family Phasianidae (pheasants, quails and partridges). Large artery, pedal small vein, femoral vein and central venous pressures and extremity volume changes were recorded in mongrel dogs in which the hindlimb was perfused at a constant flow. Arterial, pedal venous and femoral venous blood gases were drawn and analyzed for measurements of arterio-venous O2 and CO2 differences. Paw and thigh resistances were calculated. Measurements were repeated following ligation of the iliac and femoral veins, alpha receptor blockade (dibenzyline), and dilation with Vasodilan.
