Influence Of D(+)-Glucosamine On Infection Rates And Parasite Loads In Tsetse Flies (Glossina Spp.) Infected With Trypanosoma Brucei, Steve Mihok, Leonard H. Otieno, Nafisa Darji, David Munyinyi

Teneral Glossina morsitans centralis, G. m. morsitans and G. pallidipes were infected with three different clones of Trypanosoma brucei in blood containing d(+)-glucosamine, an inhibitor of tsetse midgut lectin. On average, 5 days of d(+)-glucosamine treatment tripled infection rates, without affecting the proportion of infections that matured. Total infection rates were equal in males and females, but twice as many infections matured in males. Counts of parasites in the guts and salivary glands of 277 flies revealed order of magnitude differences among flies, with females consistently having 2-3-times as many parasites as males. The large T antigen encoded by simian virus 40 (SV40) plays essential roles in the infection of permissive cells, leading to production of progeny virions, and in the infection of nonpermissive cells, leading to malignant transformation. Primary mouse embryo fibroblasts (MEFs) are nonpermissive for SV40, and infection by wild-type SV40 leads to immortalization and transformation of a small percentage of infected cells. We examined the ability of an extensive set of mutants whose lesions affect SV40 large T antigen to immortalize MEFs. We found that immortalization activity was retained by all mutants whose lesions are located upstream of codon 346. …

Performance testing of beef bulls was initiated to improve productivity of Arkansas beef cattle through breeding. This cooperative testing program provides cattlemen with reliable information for identifying animals with superior breeding value for the traits of production measured in the test. By using the information as a basis for herd sire selection, a breeder can greatly increase the chances of obtaining a bull that will sire rapid-gaining, more efficient, highquality calves. A Flow Cytometric Analysis Of Immunocyte Populations In Peripheral Blood And Pleural Effusions Of Patients With Cancer, Mandy L. Bohn

Two-color flow cytometric analysis was performed on paired samples of peripheral blood (PB) and pleural effusions (PE) of patients with metastatic malignancies. The purpose of these analyses was to test the hypothesis that there are significant differences in the distribution of immunocyte populations in the PE compared to that of the PB.

The majority of the pleural fluid immunocytes were CD3+ cells. The ratio of CD4+ to CD8+ cells was higher in the PE compared to PB. Levels of CD8+, CD19+ and CD14+ cells were not significantly different in the PE compared to the levels in the PB. CD16+CD56+ cells …

The pulps of mandibular molars of 15 athymic and 15 conventional rats were surgically exposed and left open to their oral flora. Each group was divided into 3 subgroups of 5 animals each. The rats were sacrificed after their pulps were exposed for 2, 4, or 8 weeks. After fixing, decalcifying, and embedding, the specimens were sectioned and stained with H and E. They were then examined under a microscopic grid and quantified by percentages of surface areas of bone, connective tissue, bone marrow, intrabony spaces, periapical lesions, and numbers of osteoclasts. A study of the growth of Merino weaner flocks over summer and autumn in Western Australia 's eastern wheatbelt during 1989 and 1990 has identified efficient weaner management strategies, and provided a basis for future research and extension needs of this area. The average liveweight of several weaner flocks was unexpectedly low at the start of summer. Weaner flocks may need to be supplemented before the start of summer to prevent undue weight loss at this time.

A pilot Pig Herd Health Management Scheme co-ordinated by the Department of Agriculture has stimulated much increased involvement of veterinarians in the Western Australian pig industry. Co-ordinated by Ashley Mercy, principal veterinary officer (epidemiology) horn 1985 to 1989, the scheme demonstrated the value of having regular visits from veterinary consultants and in implementing programs to prevent disease and production problems occurring in herds.

Viruses with defective genomes have been identified in association with virtually every major family of viruses and have been widely utilized as tools for investigating virus functions in animal cell culture systems (Perrault, 1981). It is generally thought that defective interfering viruses (DIs) arise through deletion, rearrangement, or recombination of a competent viral genome. DIs tack the ability for independent existence relying on their parental helper viruses to supply factors required for replication, maturation, and/or encapsidation (Huang and Baltimore, 1977). Infection Rates In Glossina Morsitans Morsitans Fed On Waterbuck And Boran Cattle Infected With Trypanosoma Congolense, Steve Mihok, Reardon O. Olubayo, David F. Wesonga

Teneral Glossina morsitans morsitans were fed on waterbuck (Kobus defassa) and Boran cattle (Bos indicus) infected experimentally with Trypanosoma congolense clone IL2895. Infection rates in tsetse varied from 9 to 31% when fed on cattle, and from 2 to 59% when fed on waterbuck. In waterbuck, infections were often detected through the development of parasites in tsetse at times when parasitaemia could not be detected through microscopic examination of blood. Male and female, and 1- and 2-day-old flies were equally susceptible to infection on both hosts. Infection in tsetse was associated with a 14% absolute reduction in survival during the …

Pathogenicity Of Tsetse-Transmitted Trypanosoma Congolense For Waterbuck (Kobus Defassa) And Boran Cattle (Bos Indicus), R. O. Olubayo, S. Mihok, D. F. Wesonga, E. R.M. Mbwabi

Five waterbuck (Kobus defassa) and four Boran cattle (Bos indicus) were infected with Trypanosoma congolense IL2895 using Glossina morsitans morsitans. At the same time, two waterbuck and two cattle were inoculated intravenously with bloodstream forms. With both methods of challenge, cattle had short prepatent periods followed by a continuous high parasitaemia. All cattle became severely anaemic and had to be treated with trypanocidal drugs to prevent death. In contrast, tsetse and intravenous challenge of waterbuck resulted in a long prepatent period, followed by brief, intermittent levels of low parasitaemia, and eventual selfcure. RNA molecules with enzymatic activity (ribozymes) targeted to the genome of HIV-1 were developed and demonstrated to mediate site-specific cleavage of the HIV-1 RNA in vitro and to inhibit viral proliferation intracellularly. Two classes of ribozymes that incorporate a "hammerhead" catalytic center were utilized, eotide elements in the viral sequence to provide half of the The first class used nuclcatalytic center.
