Ctrp3 And Alcoholic Liver Disease In Female Mice, Callie Root
Ctrp3 And Alcoholic Liver Disease In Female Mice, Callie Root
Undergraduate Honors Theses
C1q TNF Related Protein 3 (CTRP3), is a cytokine that is primarily secreted from adipose tissue, which classifies it as an adipokine. Our previous research has shown that CTRP3 prevents alcoholic fatty liver disease (ALD) in male mice. However, even when accounting for confounding factors such as absolute and relative alcohol intake, females are more sensitive to the effects of consumption compared to male mice. Therefore, the goal of this project was to determine whether CTRP3 prevented ALD in female mice. Methods: Female wild type (WT) and female CTRP3 transgenic over expressing (Tg) mice were fed an ethanol containing liquid …

Investigating The Interactions Between Individual Calmodulin And Hiv-1 Protein Domains, Riley K. Kendall, Jerry Larue
Student Scholar Symposium Abstracts and Posters
The World Health Organization found that 37.9 million people were living with HIV by the end of 2018. HIV is a virus that weakens the immune system through viral replication and the destruction of CD4+ T-cells, which are white blood cells that detect infection and make antibodies. A cure for HIV has not yet been discovered. HIV-1 contains a Gag polyprotein which regulates the stages of viral replication. Previous studies suggest that the myristoyl group of a matrix protein peptide found on the Gag polyprotein, MA, forms a complex with a calcium-binding, multifunctional regulatory protein called Calmodulin (CaM). CaM …

Volatile Organic Compound Exposure And Cardiometabolic Syndrome Risk In A Nationally Representative Cohort., Stacey Lane Konkle
Electronic Theses and Dissertations
BACKGROUND: The relative importance of environmental exposures such as volatile organic compounds (VOCs) is one of the paramount public health priorities of our time, yet is presently unstudied. VOCs are ubiquitous in the environment and have been associated with numerous adverse health effects, including a number of cardiovascular and metabolic effects that are components of Cardiometabolic Syndrome (CMS). OBJECTIVES: To examine the relationship between CMS and individual-level exposures to VOCs, measured as urinary metabolites of VOCs (UM-VOCs), in a nationally representative sample. METHODS: Associations between urinary biomarkers of exposure to 19 parent VOCs and CMS were assessed using the National …
Tobacco-Derived Aldehydes: Platelet Activation, Thrombosis, And The Role Of Trpa1., Andre Dwayne Richardson
Tobacco-Derived Aldehydes: Platelet Activation, Thrombosis, And The Role Of Trpa1., Andre Dwayne Richardson
Electronic Theses and Dissertations
Cigarette smoking is the single largest risk factor for cardiovascular disease (CVD) pathophysiology. Numerous researchers have shown potential associations between aldehydes in tobacco-derived aerosols from mainstream cigarette smoke (MCS) or electronic cigarettes (e-cigs) and their cardiotoxicity by damaging blood vessel endothelium. The severity of exposure to these toxicants can furthermore lead to adverse cardiovascular outcomes such as myocardial infarctions, stroke, coronary and peripheral artery disease, or atherosclerosis due to blood clots, a pro-thrombotic event. However, the mechanisms by which levels of harmful and potentially harmful constituents (HPHCs) such as aldehydes induce thrombosis are not well-known. Of specific interest, evidence has …

Characterization Of A Mycoplasma Pneumoniae Cards Toxin Mutant, Nikaash Pasnoori
Honors Scholar Theses
Mycoplasma pneumoniae is a high-burden pathogen which causes mild to significant infections of the respiratory system. According to the CDC, an estimated two million cases occur yearly in the United States alone, demonstrating the widespread effect of the pathogen. In addition to being the cause of respiratory infections, M. pneumoniae has also been implicated in exacerbating pre-existing asthma conditions. These morbidities make finding a vaccine candidate a vital part of easing the healthcare burden caused by the pathogen. The current mechanism of infection is unknown, but recent evidence points to the Community Acquired Respiratory Distress Syndrome (CARDS) toxin as being …

Optical Clearing Reveals Tnbs-Induced Morphological Changes Of Vglut2-Positive Nerve Endings In The Colorectum, Shivam Patel
Honors Scholar Theses
Sensitization of colorectal afferents and colorectal hypersensitivity have been observed in a mouse model of post-infectious irritable bowel syndrome via intracolonic treatment of 2,4,6-trinitrobenzenesulfonic acid (TNBS). In this study, we investigated the distribution and morphology of microscopic colorectal afferent endings before and after intracolonic treatment of TNBS. We genetically labeled predominantly extrinsic colorectal afferents using the vesicular glutamate transporter type 2 (VGLUT2) promoter. Then, we used an optical tissue clearing method of whole-mount colorectum to image labeled VGLUT2-nerve endings that are otherwise obscured in untreated samples. We used vector path tracing to quantify the density and degree of curliness of …

Tobacco Product-Derived Aldehydes: Effects On Circulating Angiogenic Cells & Implications For Cardiovascular Disease., Jordan B. Lynch
Electronic Theses and Dissertations
Cardiovascular disease (CVD) is the leading cause of death worldwide. Tobacco smoke is the most significant modifiable risk factor in CVD development and contains numerous toxic compounds, including aldehydes, which have been linked to CVD. Formaldehyde, acetaldehyde, acrolein, and crotonaldehyde have been identified as significant contributors to cigarette-induced health effects, but the mechanism of these effects is not well understood. These aldehydes are also found in aerosols produced by e-cigarettes (e-cigs). The goal of this dissertation was to explore the systemic, hematological, and endothelium-related effects of exposure to tobacco products and constituent aldehydes and to identify potential mechanisms of injury. …

Dual Role Of Antimicrobial Peptide Cathelicidin In Alcohol-Associated Liver Disease., Fengyuan Li
Electronic Theses and Dissertations
Patients with alcohol-associated liver disease (ALD) have high morbidity and mortality in its severe forms. One of the major features in different forms of ALD is the altered gut microbiota. Antimicrobial peptides (AMPs) play a crucial role in maintaining gut microbiota homeostasis. LL-37 (known as CRAMP in mouse) is the sole member of the human cathelicidin family and has piqued great research interest for its dual role in modulating microbiota and the immune response in metabolic diseases. Inflammasome activation is an important component of the liver pathophysiology in ALD and requires two signals for its full activation to induce the …
Elucidating The Developmental Defects In Zebrafish Associated With The Cardiac Drug Verapamil, Blake Stephan Justis
Elucidating The Developmental Defects In Zebrafish Associated With The Cardiac Drug Verapamil, Blake Stephan Justis
Graduate Theses/Dissertations
Birth defects are abnormalities in a developing organism that lead to a malformation in structure or function. Over half of birth defects have no determined cause; however, known causes occur by genetic anomalies, exposure to environmental agents (a.k.a. teratogens), or multifactorial reasons. To explain the unknown causes of birth defects, an area of focus in this study is to identify potential teratogens. Identifying these teratogens, is key to preventing future birth defects. An obvious source of teratogens in pregnant women would be that of pharmaceuticals. Thus, a main goal of this study is to identify drugs that cause birth defects. …
The Effects Of Zoledronate And Sleep Deprivation On The Distal Femur Trabecular Thickness Of Ovariectomized Rats: Application Of Different Statistical Methods, Erin Nolte
Student Scholar Symposium Abstracts and Posters
Osteoporosis is a disease that causes the degradation of bone, leading to an increased risk of fracture. 1 in 3 women over the age of 50 will be affected by Osteoporosis. This study aims to understand how bone is affected by sleep deprivation in estrogen-deficient rats, and how Zoledronate might negate the inimical effects of sleep deprivation on bone. As bone mineral density (BMD) is a crude evaluation of the architectural changes seen in Osteoporosis, trabecular thickness may serve as a better single evaluation of bone health. 31 Wistar female rats were ovariectomized and separated into 4 random groups. The …
Molecular Mechanisms Of Antimicrobial Resistance In Multi-Drug Resistant Enterococci, Ayesha Khan
Molecular Mechanisms Of Antimicrobial Resistance In Multi-Drug Resistant Enterococci, Ayesha Khan
Dissertations and Theses (Open Access)
Antibiotic resistance is a major global public health threat. Enterococci are recalcitrant, nosocomial pathogens that can be intrinsically resistant to valuable antibiotics, like beta-lactams, or evolve resistance to all existing antimicrobials. The LiaFSR system regulates resistance to cell membrane (CM) stressors like daptomycin (DAP), a front-line drug for multi-drug resistant infections. DAP resistance (DAP-R) in E. faecalis is mediated by CM phospholipid alterations. Emergence of DAP-R often leads to b-lactam resensitization, a phenomenon called the seesaw effect. The molecular mechanism of DAP-R and the seesaw effect are unknown. Here we show that LiaX is a surface exposed protein whose C-terminal …

Factors That Impact Uptake Of Carrier Screening By Male Reproductive Partners Of Female Prenatal Patients, Wendi Betting
Dissertations and Theses (Open Access)
Carrier screening is a genomic technology that is used to identify individuals who are carriers of autosomal recessive conditions. Despite published recommendations, the majority of male partners do not complete carrier screening after their female partner is identified to be a carrier. Previous studieshave examined reasons why women elect or decline carrier screening, but there have been few published studies that examine factors that influence a male partner’s decision to elect or decline carrier screening, particularly when the female has been identified as a carrier. The aim of the study was to determine the factors that influence the uptake of …

Heterogeneous Nuclear Ribonucleoprotein K (Hnrnp K) Overexpression And Its Interaction With Runx1 Rna In Acute Myeloid Leukemia, Marisa Aitken
Dissertations and Theses (Open Access)
Acute myeloid leukemia (AML) is an often devastating hematologic malignancy with 5-year overall survival lingering near 20%. Acquiring a deeper understanding of molecular underpinnings of leukemogenesis will provide a basis for developing more effective therapeutic strategies for patients with AML.
Here, we identified overexpression of hnRNP K as a recurrent abnormality in a subset (~20%) of AML patients. High levels of this RNA-binding protein associated with inferior clinical outcomes in de novo AML. Thus, to evaluate its putative oncogenic capacity in myeloid disease, we overexpressed hnRNP K in murine hematopoietic stem and progenitor cells isolated from fetal liver cells (FLCs). …
Food Safety Risk In An Indoor Microgreen Cultivation System, Gina Marie Misra
Food Safety Risk In An Indoor Microgreen Cultivation System, Gina Marie Misra
Graduate Theses and Dissertations
Microgreens are immature sprouts of edible plants, sharing some similarities with sprouted seeds and petite leafy greens. Since they are most often grown in containers in buildings or greenhouses, they present a new area for food safety research at the intersection of the built environment and produce farming. Contamination by human pathogens has been extensively studied in other types of produce typically eaten raw, including sprouted seeds, which have been implicated in numerous outbreaks of salmonellosis over the last several decades. There is a paucity of knowledge about the microgreen sector of the fresh-cut industry; thus, it was determined that …

Synthesis, Cytotoxicity And Apoptosis Studies On Nickel And Zinc Derivatives Of Testosterone-N4-Substituted-Thiosemicarbazones, Mok Piew Heng
Student Works (2020-2029)
The side effects of cisplatin such as toxicities are mainly due to the lack of selectivity of this chemotherapeutic agent. In order to increase the selectivity of an anticancer agent, hormone molecule that targets a specific receptor may be utilized. This research aimed to prepare cytotoxic Schiff base compounds (and their nickel and zinc complexes) with testosterone and derivatives of thiosemicarbazide, which are potentially selective towards cancerous cells. Three Schiff base ligands (TM, TF, and TP) were made from the conjugation of testosterone with three derivatives of thiosemicarbazide (N4= methyl, fluorophenyl, and ethylphenyl) and their respective nickel (NM, NF, and …

Evaluation Of Anthelmintic Therapies In A Fall Calving Beef Cowherd, Laine Zammit
Graduate Theses and Dissertations
Internal parasitism inevitability prompts economic loss in beef cattle production by decreasing growth performance and reproductive traits. Today, the most widely used class of anthelmintic used to treat parasitism, is the macrocyclic lactone. Many studies have conflicting results on the efficacy of macrocyclic lactones (ML) efficacy against internal parasitism. The objective of this study was to evaluate the effectiveness of moxidectin and eprinomectin, two of the MLs, on cow performance. Multiparous fall calving, crossbred beef cows (n = 106) were allocated randomly to 1 of 3 anthelmintic treatments: 1) Negative control (CON), in which cows did not receive an anthelmintic, …

Section of Neurology
Coronavirus disease (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS CoV-2) has turned out to be a formidable pandemic. Upcoming evidence from confirmed cases of COVID-19 suggests an anticipated incursion of patients with neurological manifestations in the weeks to come. An expression of the angiotensin-converting enzyme 2 (ACE 2), the cellular receptor for SARS-CoV-2 over the glial cells and neurons have made the brain a potential target. Neurotoxicity may occur as a result of direct, indirect and post-infectious complications. Attention to neurological deficits in COVID-19 is fundamental to ensure appropriate, timely, beneficial management of the affected patients. Most …

Has Covid-19 Impacted Price-Weight Relationships And Value Of Gain?, Elliott James Dennis
Extension Farm and Ranch Management News
First paragraph:
In the beginning of COVID-19, much of the attention was focused on managing the redirection of meat product from food service to retail stores. This past month has keenly focused on packing plant closures due to COVID-19 cases among workers and how to manage the supply of fat cattle already ready for slaughter. Fat cattle available for slaughter either cannot get bids or bids are significantly below breakeven prices leaving feedlots with decisions to be made about marketing and placements. The April 2020 Cattle on Feed report (https://usda.library.cornell.edu/concern/publications/m326m174z) revealed March placement decisions. Placements were down about …

Objective: Animal African trypanosomiasis (AAT) is a life-threatening vector-borne disease, caused by trypanosome parasites, which are principally transmitted by tsetse flies. In Kenya, the prevalence of drug-resistant trypanosomes in endemic regions remains poorly understood. The objective of this study was to establish AAT point prevalence, drug susceptibility of associated trypanosomes, and measure infectivity by multiple AAT mammalian hosts to tsetse flies in Shimba hills, a resource-poor region with high bovine trypanosomiasis prevalence and morbidity rates at the coast of Kenya. We collected tsetse flies using traps (1 Ngu and 2 biconical), and then sorted them on sex and species. Trypanosomes …
Fact Or Myth: The Flu Can Be Caused By The Flu Vaccine, Arleigh Wood
Fact Or Myth: The Flu Can Be Caused By The Flu Vaccine, Arleigh Wood
Virtual 2020 Spring Student Showcase for Research and Creative Inquiry
To many, the flu vaccine seems like a threat to an otherwise healthy individual. People often get the flu after receiving the flu shot, which does not promote confidence in the general public. These and other reasonings have created a myth that the flu vaccine can cause the flu. Understanding how the vaccination is created and how it interacts with the immune system helps to realize why the flu vaccine cannot cause the virus. For the vaccine to work, adaptive immunity must be activated by the introduction of the viral particles in a safe manner. An effective immune response from …

Syphilis is an STI that has recently made a resurgence in homosexual populations. When infected, a patient has two options: seek treatment immediately or do not seek treatment. I used a system of differential equations that includes susceptible, exposed, treatment receptive, treatment non-receptive, cured, and dead state variables in order to determine the relationships among transmission rate, treatment options, and death rate. My model suggests that an individual’s treatment option has a larger effect on death rate than the transmission rate inherent to the disease. The model also contains two sets of equilibria: an unstable trivial disease free condition and …

Ebola Virus Disease (EVD) is a rare but severe disease that is transmitted among humans through direct-contact with, and close proximity to, infected bodily fluids. From 2014-16, West Africa experienced the largest Ebola outbreak ever recorded, infecting over 28,000 people, and killing over 11,000. Although the symptoms of EVD are treatable, the disease can be extremely deadly, with an average of 50% EVD cases resulting in fatality. In areas where healthcare is scarce and vaccinations are not readily available, the practices of social distancing and self-quarantining have been shown to be highly effective in combating the spread of EVD. To …

465— Modeling Vaccine Efficacy For Tuberculosis In A Prison Population, Kaitlyn Mundackal
GREAT Day Posters
Tuberculosis is a highly contagious disease and is particularly problematic in confined communities such as prisons. I simulated how Tuberculosis moves through a prison population and tested how much vaccination effort is needed to control its spread. To explore this, I tested adding ever increasing numbers of randomly placed edges in a network and determined the size of the largest component. Afterwards, I removed edges in the model using two different methods, one illustrating if the edges were removed randomly and the other starting with prisoners that had the most connections, to simulate the effect of vaccination. My results show …

Schistosomiasis is a leading parasitic infection that debilitates many individuals in sub Saharan Africa. Because this parasitic infection involves a snail as an intermediate host, we use a native prawn species as a natural predator to decrease the population of this snail community. This study focuses on Tomefa, a primarily fishing peri-urban community located oi the outskirts of Accra, the capital city of Ghana. A survey was also given to the local community measuring economic characteristics. In the laboratory, Macrobrachium spp prawn exhibit preference of Bulinus snails over Biomphalaria snails while non-parasitic snails were consumed in higher proportion to parasitized …
The Role Of Diet In The Onset Of Depression: A Biochemical Connection Between Nutrition And Mental Health, Katlin Wildeman
The Role Of Diet In The Onset Of Depression: A Biochemical Connection Between Nutrition And Mental Health, Katlin Wildeman
Senior Honors Theses
Depression is a major clinical concern, having a complex onset and the presence of multiple, often unidentifiable causes. Depression affects millions of individuals worldwide, with a high prevalence in regions of the world with a Western-style diet as compared to regions with a Mediterranean diet. A Western-style diet consists of foods high in sugar, fat, and processed meats and grains, whereas the Mediterranean diet contains significantly more vegetables, fruits, lean meats, and whole grains. The link between diet and mental health disorders has implications for individuals of all ages who are hesitant to turn to medication. In addition to presenting …
Antidepressants Versus Music Therapy: Which Treatment Is Better For Treating Depression In Parkinson's Disease?, Emaan Dawood
Antidepressants Versus Music Therapy: Which Treatment Is Better For Treating Depression In Parkinson's Disease?, Emaan Dawood
Virginias Collegiate Honors Council Conference
Music and medicine are merging to treat depression in Parkinson’s disease (PD). Antidepressants, the current standard, demonstrate mixed effects in clinical trials, warranting other treatment options. Here, I compare music therapy to antidepressants in the treatment of depression in PD via literature review. Studies of depressed PD patients undergoing music therapy and/or antidepressants were reviewed with attention to variables including depression (on a patient-reported outcome) that were measured pre- and post- therapy. In the music studies, improvements were seen in depression. Methodological inconsistencies across these studies, a paucity of studies, and small sample sizes, however, necessitate more research to reach …
