All organisms face a constant barrage of environmental stresses. Single-cell organisms such as Saccharomyces cerevisiae, or common Baker’s yeast, must rely solely on cellular responses in order to survive. This response must occur in a rapid and highly coordinated manner to quickly inhibit all unnecessary processes and shuttle all available resources to those necessary for survival. One method that cells utilize for rapid protein regulation is the use of post-translational modifications. Clinicians, professional athletes, and recreational athletes are interested in supplementation that up-regulates nitric oxide (NO) production in blood vessel endothelium, increasing arterial vasodilation. Benefits from these supplements include improvements in blood pressure, muscle hyperemia, and exercise performance. Citrulline Malate (CM) is a pre-workout ingredient, popular for its ability to increase exercise performance and blood serum concentrations of L-arginine, resulting in NO production. Recently, Inositol-Stabilized Arginine Silicate (ASI, Nitrosigine) has been added to many of the most popular pre-workout blends, following a group of studies showing ASI increases serum arginine and reduces post-workout muscle damage. Ipomoeassin F is a flagship congener of a resin glycoside family that inhibits growth of many tumor cell lines with only single-digital nanomolar IC50 values. However, biological and pharmacological mechanisms of ipomoeassin F have been undefined. To facilitate exploration of the biological and pharmacological properties, we performed sophisticate SAR (Structure–activity relationship) studies of ipomoeassin F to understand its pharmacophore and structure properties so that we can design favorable probes for further biological investigation. There is an urgent need for novel treatments for Candida infections. The utility of antimicrobial peptides for antifungal therapy has garnered interest in recent years. One promising family of peptides is the Histatins, a family of naturally-occurring peptides secreted into the oral cavity that display antimicrobial activity. Histatin 5 is a twenty-four amino acid peptide with strong antifungal activity. Studies from our laboratory have identified a small histatin-derived peptide, KM29, that yields fungicidal activity 10-fold greater than Histatin 5 against multiple Candida species. Obesity prevalence in the United States continues to increase and is associated with health consequences such as type 2 diabetes, hypertension, atherosclerosis, and hyperlipidemia. Among many contributing factors to obesity, fructose may be one of the major reasons as it disrupts the antioxidant system thereby resulting in an accumulation of reactive oxidative species and leading to obese conditions. Amyloid Beta (Aβ) was the major focus of this study. It is a peptide that is present in the brain with a high tendency to self-aggregate. When this protein aggregates, it forms oligomers and protofibrils which in turn are deposited as senile plaques in the brain. The reason for the concern with these plaques is their association with the neurological disorder Alzheimer’s disease. Candida albicans is a normal inhabitant of the skin and mucosal membranes of humans, however, in individuals with depressed immune systems or disrupted cutaneous flora, Candida can overgrow and cause serious infection. Candida infection is the fourth leading cause of nosocomial infection in the United States. These infections are often associated with longer hospital stays and higher mortality. Current drug therapies for this infection are largely ineffective due to the increased drug resistance of Candida species, and for some therapeutics, high levels of drug toxicity to humans. Purpose: The growing amount of clinical resistance observed in current antifungal drugs and in anti-HIV pharmaceuticals is a concern in the medical community. The purpose of this study is to develop a mild synthetic process for biomedically relevant thiazolyl aryl ketones that can be used to develop antifungal and anti-HIV drugs. We hypothesized that the proposed synthetic technique would be more efficient, produce fewer unwanted byproducts, and be more tolerant of functional groups than existing methods.

Arterial compliance (i.e., arterial elasticity) is a major contributor to the pathophysiology of cardiovascular disease (CVD) and has been recognized as an independent risk factor for CVD. Smokers have poor arterial compliance compared to non-smokers. In non-smokers, passive heat stress has been shown to acutely improve arterial compliance. It is unknown how passive heat stress affects arterial compliance in smokers. The purpose of this pilot study was to test the hypothesis that passive heat stress improves arterial compliance in smokers to a greater extent than non-smokers. The intracellular bacterial agent of Q fever, Coxiella burnetii, replicates within a phagolysosomelike parasitophorous vacuole (PV) in human macrophages and delivers effector proteins to the host cytosol via a Dot/Icm type IV secretion system (T4SS). The T4SS effectors are critical for PV formation and prevention of host cell death that allows sufficient time for bacterial replication. Recruitment of ubiquitin-related components to the C. burnetii PV is also predicted to be involved in PV formation and bacterial replication and is likely controlled by effector proteins. Synthetic cannabinoids (SC's) began to gain popularity around the world in 2009. Since then, many of the compounds have been outlawed and methods developed to detect them and their metabolites using mass spectrometry. Our work investigated the possibility of developing a colorimetric presumptive test. The SC JWH-019 was synthesized and its ketone targeted as a possible reaction site. Many SC's contain ketones and thus a reaction at this site would be applicable to many of the compounds. Due to significant lipid and carbohydrate production as well as other useful properties such as high production of useful biomolecular substrates (e.g., lipids) and the ability to grow using non-potable water sources, algae are being explored as a potential high-yield feedstock for biofuels production. In both natural and engineered systems, algae can be exposed to a variety of environmental conditions that affect growth rate and cellular composition. Context: Maintaining thermal balance under heat-stress depends on appropriate increases in sweating and skin blood flow (cutaneous vasodilation). Given the multiple effects on nicotine on the body, it is unknown if sweating and cutaneous vasodilation are impaired, or possibly enhanced in smokers during heat stress. Objective: To examine the effects of passive heating on thermoregulatory responses (i.e., sweating and cutaneous vasodilation) in smokers versus non-smokers. Design: 1 passive heat trial per subject. Dietary conjugated linoleic acid (CLA) is known to be effective in avoiding many obesity related diseases. Conjugated linoleic acid is a product of ruminant fermentation and 3.4 g/day are needed to obtain the clinical benefits. However, it is difficult to obtain sufficient CLA to realize these benefits from a healthy diet containing dairy and beef products, without increasing levels of dietary cholesterol and saturated fat. A 20% CLA soy oil with low saturated fat and no cholesterol has been produced by photoisomerization of linoleic acid in the triacylglyceride oil. A 31p, 13c, And 1h Nmr Study Of The Direct Interaction Of Cocaine Hc1 And Magnesium Atp, Gene Lee Bangs, Roger M. Hawk, Debi Patangia

In vivo 31P NMR studies recently have shown that cocaine causes an imbalance of the free magnesium in the brain which results in pH lowering, ischemia, and even death. This direct interaction with the free Mg+2 inthe brain also affects the Ca+2 balance which controls arterial and vascular contraction. This research has addressed the mechanism of the cocaine interaction withmagnesium adenosine 5-triphosphate (ATP) using 31P, 13C, and 1H NMR using a Bruker 200 MHz nuclear magnetic resonance (NMR) system. Data are presented and discussed which shows that cocaine and ATP form a complex species which directly affects the NMR spectra.

The conformation of the nitro group of nitroaromatic compounds relative to the aromatic ringsystem is suggested to affect their metabolic activation and mutagenicity. We have recently showed the nitrophenylfuran skeletal muscle relaxant, dantrolene, tobe a potent mutagen inSalmonella. Synthesis of or^o-substituted analogues of dantrolene was achieved in an effort to alter the conformation of the nitro group ina manner that willdecrease the mutagenicity.
