Cold atmospheric plasma, including plasma jet and surface plasma, can promote the apoptosis of cancer cells without causing significant damage to surrounding normal cells, which was hopeful to be applied to the clinical cancer therapy. Intense pulsed electric fields (PEF) are a novel modality for the efficient and targeted ablation of tumors by electroporation. The major adverse side effects of PEF therapies are strong involuntary muscle contractions and pain. Nanosecond-range PEF (nsPEF) are less efficient at neurostimulation and can be employed to minimize such side effects. We quantified the impact of the electrode configuration, PEF strength (up to 20 kV/cm), repetition rate (up to 3 MHz), bi- and triphasic pulse shapes, and pulse duration (down to 10 ns) on eliciting compound action potentials (CAPs) in nerve fibers. We developed two models of chemically induced chronic lung injury and pulmonary fibrosis in mice (intratracheally administered hydrochloric acid (HCl) and intratracheally administered nitrogen mustard (NM)) and investigated male-female differences. Female mice exhibited higher 30-day survival and less weight loss than male mice. Thirty days after the instillation of either HCl or NM, bronchoalveolar lavage fluid displayed a persistent, mild inflammatory response, but with higher white blood cell numbers and total protein content in males vs. females. Furthermore, females exhibited less collagen deposition, milder pulmonary fibrosis, and lower Ashcroft scores. After instillation of either HCl or NM, all animals displayed …

Exposure to hydrochloric acid (HCl) represents a threat to public health. Children may inhale higher doses and develop greater injury because of their smaller airways and faster respiratory rate. We have developed a mouse model of pediatric exposure to HCl by intratracheally instilling p24 mice (mice 24 days old; 8–10 g) with 2 µL/g 0.1 N HCl, and compared the profile of lung injury to that in HCl-instilled adults (10 weeks old; 25–30 g) and their age-matched saline controls. Earplugs are a common form of protection for workers exposed to hazardous noise levels. Their comfort directly impacts the effective protection by influencing their consistent and correct use. Nevertheless, comfort definition may vary according to the studies. Thus, a previous review of the literature has shown that to improve our understanding of perceived comfort and to reduce measurement variability, it is advisable to consider comfort through a multidimensional construct (physical, acoustical, functional and psychological). The occlusion effect is commonly experienced as the altered perception of one’s own physiological noise when the earcanal entrance is blocked. Objectively, this phenomenon corresponds to an acoustic pressure increase in the occluded earcanal. The occlusion effect originates from the earcanal wall normal vibration and depends on the spatial distribution of the latter. At low frequencies, this spatial distribution can be characterized by the position of its centroid along the earcanal middle axis. The use of earplugs is commonly associated with an increased perception of the bone-conducted part of one's own physiological noise. This phenomenon is referred to as occlusion effect and is most prominent at low frequencies. The occlusion effect is commonly experienced by in-ear device wearers as an increased loudness sensation of bone-conducted low frequency sounds. A widespread theory proposed by Tonndorf and based on a simplified electro-acoustic model describes the phenomenon as the removal of the open earcanal high-pass filter effect due to a perfect or partial occlusion. However, this filter has not been clearly defined and several ambiguities remain. Revisiting the model, a second order high-pass filter effect for the volume velocity transferred between the earcanal wall and the eardrum is highlighted. A novel monopolar electroporation system and methodologies were developed for in vivo electroporation intended for potential clinical applications such as gene therapy. We hypothesized that an asymmetric anode/cathode electrode applicator geometry could produce favorable electric fields for electroporation, without the typical drawback associated with traditional needle and parallel plate geometries. Three monopolar electrode applicator prototypes were built and tested for gene delivery of reporter genes to the skin in a guinea pig model. Gliomas are primary brain tumors that originate from glial cells. Classification and grading of these tumors is critical to prognosis and treatment planning. The current criteria for glioma classification in central nervous system (CNS) was introduced by World Health Organization (WHO) in 2016. Combining vibrating mesh nebulizers with additional new technologies leads to substantial improvements in pharmaceutical aerosol delivery to the lungs across therapeutic administration methods. In this dissertation, streamlined components, aerosol administration synchronization, and/or Excipient Enhanced Growth (EEG) technologies were utilized to develop and test several novel devices and aerosol delivery systems. The study aims were to develop fracture prediction models by using machine learning approaches and genomic data, as well as to identify the best modeling approach for fracture prediction. The genomic data of Osteoporotic Fractures in Men, cohort Study (n = 5130), were analyzed. After a comprehensive genotype imputation, genetic risk score (GRS) was calculated from 1103 associated Single Nucleotide Polymorphisms for each participant. Data were normalized and split into a training set (80%) and a validation set (20%) for analysis. The objective of this research project is to evaluate the efficiency of traditional chemical tracing compared to dual-viral tracing for labeling long ascending propriospinal neurons (LAPNs) in the uninjured rat spinal cord, and to develop a MATLAB program which will accurately quantify this labeling.

To evaluate chemical tracing, Fluoro-Ruby (FR) was injected at the level of the axon terminals, and the number of ipsilateral labeled cell bodies at lumbar level was quantified. Weight-sharing is one of the pillars behind Convolutional Neural Networks and their successes. However, in physical neural systems such as the brain, weight-sharing is implausible. This discrepancy raises the fundamental question of whether weight-sharing is necessary. If so, to which degree of precision? If not, what are the alternatives? The goal of this study is to investigate these questions, primarily through simulations where the weight-sharing assumption is relaxed. Purpose: We describe a shape-aware multisurface simplex deformable model for the segmentation of healthy as well as pathological lumbar spine in medical image data.

Approach: This model provides an accurate and robust segmentation scheme for the identification of intervertebral disc pathologies to enable the minimally supervised planning and patient-specific simulation of spine surgery, in a manner that combines multisurface and shape statistics-based variants of the deformable simplex model. Hypothesis: To characterize anatomical measurements and shape variation of the facial nerve within the temporal bone, and to create statistical shape models (SSMs) to enhance knowledge of temporal bone anatomy and aid in automated segmentation.

Background: The facial nerve is a fundamental structure in otologic surgery, and detailed anatomic knowledge with surgical experience are needed to avoid its iatrogenic injury. Trainees can use simulators to practice surgical techniques, however manual segmentation required to develop simulations can be time consuming. Increased levels of heat shock protein 90 (HSP90) have been recently implicated in the pathogenesis of pulmonary fibrosis and the use of HSP90 inhibitors constitutes a potential therapeutic approach. Similarly, acute exposure to nitrogen mustard (NM) is related to the development of chronic lung injury driven by TNF-α, TGF-β, ERK and HSP90. Thus, we developed a murine model of NM-induced pulmonary fibrosis by instilling C57BI/6J mice with 0.625 mg/kg mechlorethamine hydrochloride. The objective occlusion effect induced by bone-conducted stimulation refers to the low frequency acoustic pressure increase that results from occluding the ear canal opening. This phenomenon is commonly interpreted as follows: the bone-conducted sound “leaks” through the earcanal opening and is “trapped” by the occlusion device. This instinctive interpretation misrepresents the fundamental mechanism of the occlusion effect related to the earcanal impedance increase and already highlighted by existing electro-acoustic models. Existing methods in neuromorphic olfaction mainly focus on implementing the data transformation based on the neurobiological architecture of the olfactory pathway. While the transformation is pivotal for the sparse spike-based representation of odor data, classification techniques based on the bio-computations of the higher brain areas, which process the spiking data for identification of odor, remain largely unexplored. Au Québec, environ 360 000 travailleurs sont exposés quotidiennement à des niveaux de bruit susceptibles d’engendrer des problèmes d’audition. Les protecteurs auditifs individuels sont souvent utilisés pour pallier ce problème. Cependant, l’inconfort lié à l'utilisation de ces protecteurs limite la durée du port et donc l'efficacité de la protection. This proposed project will be developing an artificial reverse shoulder arthroplasty joint that focuses on improvements to the Zimmer Biomet Comprehensive Reverse System by freating a unique feature, or features, to the humeral component that focuses on the prevention of dislocation by not limiting the range of motion of men from ages 45 to 65 years.

Pediatric patients with disorders and diseases of the heart have limited options with regards to implantable devices. Many of these implants are ventricular assist devices, which is not always suitable for a patient. Total artificial hearts (TAHs) have supported many adult patients until transplantation, and we believe that they could do the same for pediatric patients. A distinct bioenergetic impairment of heart mitochondrial subpopulations in diabetic cardiomyopathy is associated with obesity; however, many type 2 diabetic (T2DM) patients with high-risk for cardiovascular disease are not obese. A brain tumor is an uncontrolled growth of cancerous cells in the brain. Accurate segmentation and classification of tumors are critical for subsequent prognosis and treatment planning. This work proposes context aware deep learning for brain tumor segmentation, subtype classification, and overall survival prediction using structural multimodal magnetic resonance images (mMRI). We first propose a 3D context aware deep learning, that considers uncertainty of tumor location in the radiology mMRI image sub-regions, to obtain tumor segmentation. An updated classification of diffuse lower-grade gliomas is established in the 2016 World Health Organization Classification of Tumors of the Central Nervous System based on their molecular mutations such as TP53 mutation. This study investigates machine learning methods for TP53 mutation status prediction and classification using radiomics and genomics features, respectively. One of the most challenging problems encountered in deep learning-based brain tumor segmentation models is the misclassification of tumor tissue classes due to the inherent imbalance in the class representation. Consequently, strong regularization methods are typically considered when training large-scale deep learning models for brain tumor segmentation to overcome undue bias towards representative tissue types. However, these regularization methods tend to be computationally exhaustive, and may not guarantee the learning of features representing all tumor tissue types that exist in the input MRI examples.
