Commissioning An Anthropomorphic Spine And Lung Phantom For Remote Dose Verification Of Institutions Participating In Rtog 0631, Douglas F. Caruthers

Dissertations and Theses (Open Access)

The RPC developed a new phantom to ensure comparable and consistent radiation administration in spinal radiosurgery clinical trials. This study assessed the phantom’s dosimetric and anatomic utility. The ‘spine phantom’ is a water filled thorax with anatomy encountered in spinal radiosurgery: target volume, vertebral column, spinal canal, esophagus, heart, and lungs. The dose to the target volume was measured with axial and sagittal planes of radiochromic film and thermoluminescent dosimeters (TLD). The dose distributions were measured with the radiochromic film calibrated to the absolute dose measured by the TLD. Four irradiations were administered: a four angle box plan, a seven …

The Use Of Preclinical Models To Improve The Treatment Of Retinoblastoma, Katie Marie Nemeth May 2010

The Use Of Preclinical Models To Improve The Treatment Of Retinoblastoma, Katie Marie Nemeth

Theses and Dissertations (ETD)

Rodent models play an essential role in the development of new chemotherapeutics and dosing regimes. It is often difficult to carryout a clinical study for pediatric cancers due to the small patient population. Retinoblastoma, a pediatric cancer of the eye, is one example of a pediatric cancer that can benefit from preclinical studies. Over the years various retinoblastoma rodent models have been developed used to test various combination of broad-spectrum systemic chemotherapy. It was found form these studies that the combination of topotecan and carboplatin was effective. However both drugs cause myelosuppression and therefore administrating both of these drugs systemically …

Development And Application Of Computational Tools For The Study And Optimization Of Variable Resolution X-Ray (Vrx) Computed Tomography Scanners, David Alejandro Rendon

Theses and Dissertations (ETD)

The overall goal of this project was to develop and apply important computerized aids for the design and implementation of Variable Resolution X-ray (VRX) CT scanners developed at the University of Tennessee, Memphis. VRX scanners take advantage of the “projective compression” principle that allows the same device to image objects of very different sizes with the same level of detail by adjusting the field of view and the reconstruction resolution.

The first part of this project aimed to develop a set of computational tools specifically tailored for the design, implementation and study of VRX scanners. This included creating a reconstruction …

Preosteoblast Behavior On A Synthetic Calcium Phosphate Scaffold Coated With Cell Derived Matrix, Daniel A. Young May 2010

Preosteoblast Behavior On A Synthetic Calcium Phosphate Scaffold Coated With Cell Derived Matrix, Daniel A. Young

Theses and Dissertations (ETD)

Bone grafts are commonly used for the treatment of large bone defects. Biodegradable, synthetic bone grafts combined with cells and growth factors as a tissue engineered construct can provide benefits over the common autografts and allografts currently used. Calcium phosphate (CaP) materials offer high biocompatibility and chemical similarity to natural bone tissue. Biodegradable beta-tricalcium phosphate (β-TCP) scaffolds with an interconnected porous network have been created to promote tissue ingrowth and regeneration in vitro and in vivo. Extracellular matrix (ECM) coated on titanium and polymer scaffolds has been shown to improve cell response to those material surfaces. Here, we attempted …

Mechanism-Based Strategies To Enhance The Actions Of A, Fabiola C. Gomez May 2010

Mechanism-Based Strategies To Enhance The Actions Of A, Fabiola C. Gomez

Dissertations and Theses (Open Access)

Heat shock protein 90 (HSP90) is an abundant molecular chaperone that regulates the functional stability of client oncoproteins, such as STAT3, Raf-1 and Akt, which play a role in the survival of malignant cells. The chaperone function of HSP90 is driven by the binding and hydrolysis of ATP. The geldanamycin analog, 17-AAG, binds to the ATP pocket of HSP90 leading to the degradation of client proteins. However, treatment with 17-AAG results in the elevation of the levels of antiapoptotic proteins HSP70 and HSP27, which may lead to cell death resistance. The increase in HSP70 and HSP27 protein levels is due …

Characterization And Optimization Of Antigen-Specific T Cell Responses During Ex Vivo Expansion Of Melanoma Tumor-Infiltrating Lymphocytes, Yufeng Li May 2010

Characterization And Optimization Of Antigen-Specific T Cell Responses During Ex Vivo Expansion Of Melanoma Tumor-Infiltrating Lymphocytes, Yufeng Li

Dissertations and Theses (Open Access)

Treatment of metastatic melanoma with tumor reactive T cells (adoptive T cell therapy, ACT) is a promising approach associated with a high clinical response rate. However, further optimization of this treatment modality is required to increase the clinical response after this therapy. ACT in melanoma involves an initial phase (pre-REP) of tumor-infiltrating lymphocyte (TIL) expansion ex vivo from tumor isolates followed by a second phase, “rapid expansion protocol” (REP) generating the billions of cells used as the TIL infusion product. The main question addressed in this thesis was how the currently used REP affected the responsiveness of the CD8+ T …

Influence Of Anchoring On Miscarriage Risk Perception Associated With Amniocentesis, Regina A. Nuccio

Dissertations and Theses (Open Access)

INFLUENCE OF ANCHORING ON MISCARRIAGE RISK PERCEPTION ASSOCIATED WITH AMNIOCENTESIS Publication No. ___________ Regina Nuccio, BS Supervisory Professor: Claire N. Singletary, MS, CGC Amniocentesis is the most common invasive procedure performed during pregnancy (Eddleman, et al., 2006). One important factor that women consider when making a decision about amniocentesis is the risk of miscarriage associated with the procedure. People use heuristics such as anchoring, the action of using a prior belief regarding the magnitude of risk as a frame of reference for new information to be synthesized, to better understand risks that they encounter in their lives. This study aimed …

The Effects Of Cellular Theta Breathing Meditation On Cell Mediated Immune Response: A Controlled, Randomized Investigation Of Altered Consciousness And Health, Marjorie D. Hardgrave

UNLV Theses, Dissertations, Professional Papers, and Capstones

Medical anthropology is well positioned to make contributions to consciousness research based on biocultural approaches that integrate methodologies from the biological, behavioral and social sciences to explore aspects of human health. The ubiquity and perseverance of health related activities involving altered states of consciousness (ASC) across cultures past and present suggest that these potentials are deeply rooted in human sociocultural evolution. Analyzing the relationship between immune function and meditative ASC represents an effort to empirically investigate the adaptive value of these human potentials.

A controlled, randomized investigation of two meditation practices was conducted at the University of Nevada, Las Vegas …

Optimization Algorithms For Functional Deimmunization Of Therapeutic Proteins, Andrew S. Parker, Wei Zheng, Karl E. Griswold, Chris Bailey-Kellogg

Dartmouth Scholarship

To develop protein therapeutics from exogenous sources, it is necessary to mitigate the risks of eliciting an anti-biotherapeutic immune response. A key aspect of the response is the recognition and surface display by antigen-presenting cells of epitopes, short peptide fragments derived from the foreign protein. Thus, developing minimal-epitope variants represents a powerful approach to deimmunizing protein therapeutics. Critically, mutations selected to reduce immunogenicity must not interfere with the protein's therapeutic activity.

Segmentation Of Thermographic Images Of Hands Using A Genetic Algorithm, Payel Ghosh, Judith Gold, Melanie Mitchell Jan 2010

Segmentation Of Thermographic Images Of Hands Using A Genetic Algorithm, Payel Ghosh, Judith Gold, Melanie Mitchell

Computer Science Faculty Publications and Presentations

This paper presents a new technique for segmenting thermographic images using a genetic algorithm (GA). The individuals of the GA also known as chromosomes consist of a sequence of parameters of a level set function. Each chromosome represents a unique segmenting contour. An initial population of segmenting contours is generated based on the learned variation of the level set parameters from training images. Each segmenting contour (an individual) is evaluated for its fitness based on the texture of the region it encloses. The fittest individuals are allowed to propagate to future generations of the GA run using selection, crossover and …

CEST is a molecular imaging technique that allows indirect detection of protons associated with mobile proteins. GlycoCEST is a variant of CEST for imaging tissue glycogen, the storage form of glucose. With glycoCEST, the -OH protons of glycogen are saturated, transfer the saturation to bulk water by way of chemical exchange which reduces the bulk water signal in proportion to the glycogen content. the purpose of this study was to determine the feasibility of glycoCEST imaging in human skeletal muscle at 7T. Our findings, although preliminary, suggest that glycoCEST imaging at 7T can be used to image muscle glycogen.

The Power Of Empowerment: An Icf-Based Model To Improve Self-Efficacy And Upper Extremity Function Of Survivors Of Breast Cancer, Mary Insana Fisher, Dana Howell

Physical Therapy Faculty Publications

Breast cancer is one of the most frequently diagnosed cancers among women. Breast cancer treatments often negatively impact the function of the arm, and quality of life and upper extremity function does not always return to a prediagnosis level. Survivors of breast cancer may also experience feelings of diminished self-efficacy related to functional deficits resulting from their physical limitations. The International Classification of Functioning (ICF) provides a framework for rehabilitation practitioners to address physical and psychological impairments, activity limitations, and participation restrictions. Patient outcomes may be improved by fostering self-efficacy through empowerment. This paper explores how the ICF model and …

Excitoxicity In Retinal Ischemia And Treatment Using Non-Competitive Receptor Antagonists, Jacob Rube Jan 2010

Excitoxicity In Retinal Ischemia And Treatment Using Non-Competitive Receptor Antagonists, Jacob Rube

The Science Journal of the Lander College of Arts and Sciences

The following is an excerpt from the article: Ischemia is defined as an inadequacy of blood flow to tissue. Ischemia can deprive tissue of oxygen and metabolic substrates and it can also prevent the removal of waste products. If the ischemia is maintained over enough time the tissue will lose its homeostasis and eventually die causing an infarct. Retinal ischemia occurs when the blood supply to the retina does not meet the metabolic needs that are required to sustain the retina. This can lead to retinal damage and severe vision loss. Ischemia is caused by occluded blood vessels.

Cardiac Tissue Regeneration - The Use Of Stem Cells, Chavy Friedlander Jan 2010

Cardiac Tissue Regeneration - The Use Of Stem Cells, Chavy Friedlander

The Science Journal of the Lander College of Arts and Sciences

The following is the introduction to the article: Cardiovascular disease is the leading cause of illness and fatality. Diseases such as coronary artery disease, myocardial infarction and subsequent congestive heart failure pose a serious problem to much of the population in the Unites States and all over the world. The heart’s compromised function that results from the above conditions is so influential as the heart has little capacity to repair its lost tissue; once the cardiac myocytes are destroyed its effects are amplified by a drastic decrease in cardiac function. Until recently with the advent of stem cell technology, researchers …

Small Intestine Submucosa For Repair Of Injured Rabbit Cornea: A Pilot Study, Mark Nicholas Perenich

PCOM Biomedical Studies Student Scholarship

Corneal injuries blind approximately 15 million people worldwide annually. Current methods of treatment have several disadvantages with respect to cost and availability. This study evaluated a small intestine submucosa (SIS) xenograft treatment for a severe corneal injury. SIS could be a favorable alternative to the current treatment methods, because it is inexpensive to produce and abundant where pigs are consumed. A severe ocular injury was created in the right eye of 5 rabbits by n-heptanol and mechanical debridement of the cornea and limbus. The cornea was then evaluated with fluorescence stain and a SIS xenograft was used for treatment. Clinical …

Electromyographic Analysis Of Trunk Muscle Activation During A Throwing Pattern Following Rotator Cuff Mobilization, Aubrey L. Doede Jan 2010

Electromyographic Analysis Of Trunk Muscle Activation During A Throwing Pattern Following Rotator Cuff Mobilization, Aubrey L. Doede

CMC Senior Theses

Correct muscular activation of the body segments during an overhand throw is achieved when movement originates in the larger and more proximal legs and trunk and moves sequentially to the smaller, distal segments of the shoulder and arm. This sequence permits angular velocity to transfer progressively through the throw as part of an open kinetic chain. The athlete can summate angular velocity and segmental forces only if he is able to create a separation between the body segments during the movement pattern, and this separation is thus essential to effective segmental sequencing for activation of the trunk muscles to occur …

Quantification Of Circulating Melanoma Cells, Jamie Freeman Jan 2010

Quantification Of Circulating Melanoma Cells, Jamie Freeman

Theses : Honours

Current prognostic techniques for Cutaneous Malignant Melanoma (CMM), a highly aggressive and drug resistant skin cancer, are inadequate at managing the disease and identifying early stage patients requiring treatment. It is thought that Circulating Tumour Cells (CTCs), which circulate in patient blood after being shed from solid tumours, may be useful in enhancing prognostic techniques and it has previously been shown in other malignancies that the presence of CTCs in patient blood is associated with poor prognosis. In CMM, CTCs can be detected through RT-PCR for melanoma associated markers, although this technique does not allow CTCs to be quantified. In …

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

Inflammatory responses to implanted biomedical devices elicit a foreign body fibrotic reaction that limits device integration and performance in various biomedical applications. We examined chronic inflammatory responses to microgel conformal coatings consisting of thin films of poly(N-isopropylacrylamide) hydrogel microparticles cross-linked with poly(ethylene glycol) diacrylate deposited on poly(ethylene terephthalate) (PET). Unmodified and microgel-coated PET disks were implanted subcutaneously in rats for 4 weeks and explants were analyzed by histology and immunohistochemistry. Microgel coatings reduced chronic inflammation and resulted in a more mature/organized fibrous capsule. Microgel-coated samples exhibited 22% thinner fibrous capsules that contained 40% fewer cells compared to unmodified PET disks. …
