Diseases caused by vector-borne plant pathogens cause adverse impacts on yield resilience, food security, and farmer livelihoods, which are bound to aggravate under global change. Biological control is routinely discounted as a mitigation strategy for plant diseases, partially due to scarce and inconclusive empirical support. Here, using curated field survey data for 58 persistently or semi-persistently transmitted pathogens, we employ a multi-method approach to assess the role of resident (i.e., naturally occurring) biological control agents in these pathosystems. Our meta-analyses show how in planta pathogen incidence is strongly affected by vector abundance and infectivity. Meanwhile, biological control agent density negatively …

Phosphorylation of the JNK (pJNK) protein promotes an immunosuppressive tumor microenvironment (TME), enhancing aggressiveness in inflammatory triple-negative breast cancer (TNBC). This study evaluated the role of JNK signaling using a gene signature. RNA sequencing was performed on 347 TNBC tumors from the phase 3 International Breast Cancer Study Group (IBCSG) 22-00 trial, which evaluated adjuvant low-dose cyclophosphamide and methotrexate (CM). Immune-related tumors were identified by TNBC subtype or tumor-infiltrating lymphocytes (TILs). Associations between JNK and outcomes were analyzed using Cox models. Low pJNK levels were associated with better disease-free survival (DFS) in immune-related tumors. These tumors also had lower Treg …

Evaluating Current Practices In Operative Nontechnical Skills Training And Feedback In General Surgery Residency: A Mixed Methods Approach, Madeline Cloonan
Evaluating Current Practices In Operative Nontechnical Skills Training And Feedback In General Surgery Residency: A Mixed Methods Approach, Madeline Cloonan
Theses & Dissertations
Background: Non-technical skills (NTS) are the interpersonal and cognitive abilities that are critical to accomplishing high-risk tasks. Deficiencies in NTS, including communication, teamwork, leadership, and decision-making, are linked to errors in the operating room. Structured training and feedback in NTS remain inconsistent and have not been integrated into daily training. This dissertation addresses this gap by evaluating current NTS training and feedback practices and developing a rubric to enhance the effectiveness of NTS feedback.
Methods: We conducted five complementary studies to assess current NTS training and feedback. The first was a systematic review aimed at identifying intraoperative NTS training interventions …
Colostrum Extracellular Vesicle Are Neuroprotective In The Mptp Intoxicated Mouse Model Of Parkinson's Disease, Samia Akter
Colostrum Extracellular Vesicle Are Neuroprotective In The Mptp Intoxicated Mouse Model Of Parkinson's Disease, Samia Akter
Theses & Dissertations
Parkinson’s Disease (PD) is the second most common neurodegenerative disorder. Dopaminergic neuronal loss in the substantia nigra and the formation of Lewy bodies are the disease pathological hallmarks. We demonstrate that bovine colostrum extracellular vesicles (EVs) induce anti-inflammatory and neuroprotective activities during MPTP (methyl-4-phenyl-1,2,3,6-tetrahydropyridine) intoxication. Treatment these mice with colostrum EVs reduced the extent of activated microglia and neurodegenerative activities. RNA sequencing and transcriptomic analysis revealed a significant decrease in pro- inflammatory responses, as well as reduced recruitment and activation of immunocytes in affected brain tissues. Decreased expression of pro-inflammatory cytokines and chemokines, as well as suppressed canonical inflammasomes, was …
Study Of Peptide Amphiphiles As Antimicrobial Agents, Aramis J. Pereira
Study Of Peptide Amphiphiles As Antimicrobial Agents, Aramis J. Pereira
Theses & Dissertations
The global antimicrobial resistance (AMR) crisis presents a severe public health threat, with drug-resistant infections now the third leading cause of mortality worldwide. This escalating problem, driven by the rapid proliferation of multidrug-resistant organisms (MDROs) like the "ESKAPE" pathogens, severely limits treatment options and imposes substantial economic burdens. The crisis is further exacerbated by a limited pipeline of new antimicrobial agents, as most recently approved drugs are modifications of existing classes rather than novel compounds. Peptide amphiphiles (PAs) offer a promising alternative due to their membrane-targeting mechanism, which reduces the likelihood of resistance development. These molecules are amphiphilic and positively …

Oncoplastic Nipple-Sparing Mastectomy And Immediate Reconstruction In Non-Ideal Candidates, Aaron Dadzie, Yossef Alsabawi, Sonia Y. Khan, Paul Berry
School of Medicine Publications
Mastectomies have long been used as a surgical treatment for malignant and benign disorders of the breast, both as a therapy and prophylaxis in high-risk patients. A nipple-sparing mastectomy (NSM) allows for the removal of most of the glandular and ductal tissues while preserving the surface architecture of the nipple-areola complex (NAC). This report summarizes the current mastectomy practices and suggests an expansion of the established surgical selection criteria for NSM in the context of immediate implant-based breast reconstruction. We present two cases in which a novel hybrid oncoplastic approach was used and discuss its safety and efficacy when performing …

The organizational principles of nephronal segments are based on anatomical and physiological attributes that are linked to the homeostatic functions of the kidney. Recent molecular approaches have uncovered layers of deeper signatures and states in tubular cells that arise at various time points on the disease trajectory. Here, we introduce an analytical pipeline of multiplexed spatial protein imaging integrated with RNA expression to characterize proximal tubular subpopulations and neighborhoods in human kidney tissue. We demonstrate that, in reference tissue, a large proportion of S1 proximal tubular epithelial cells expresses thymus antigen 1 (THY1), a mesenchymal stromal and stem cell marker …

Esophageal adenocarcinoma is increasingly prevalent and is thought to arise from Barrett's esophagus (BE), a metaplastic condition in which chronic acid and bile reflux transforms the esophageal squamous epithelium into a gastric-intestinal glandular mucosa. The molecular determinants driving this metaplasia are poorly understood. We developed a human BE organoid biobank that recapitulates BE's molecular heterogeneity. Bulk and single-cell transcriptomics, supported by patient tissue analysis, revealed that BE differentiation reflects a balance between SOX2 (foregut/esophageal) and CDX2 (hindgut/intestinal) transcription factors. Using squamous-specific inducible Sox2-KO (Krt5CreER/+ Sox2Δ/Δ ROSA26tdTomato/+) mice, we observed increased basal proliferation, reduced squamous differentiation, and expanded metaplastic glands at …

Acinetobacter baumannii causes prolonged infections that disproportionately affect immunocompromised populations. Our understanding of A. baumannii respiratory pathogenesis relies on an acute murine infection model with limited clinical relevance that employs an unnaturally high number of bacteria and requires assessment of bacterial load at 24-36 h post-infection. Here, we demonstrate that low intranasal inoculums in tlr4 mutant mice allows for infections lasting at least 3 weeks. Using this "chronic infection model" we determine the adhesin InvL is a virulence factor required during later stages of infection, despite being dispensable in the early phase. We also demonstrate that the chronic model enables …

Patterns Of Ophthalmology-Related Interfacility Transfers To The Emergency Department By Temporal Trends, Laura E. Ramirez, Michael Waxman, Denis R. Pauze, Edward J. Wladis
Journal of Academic Ophthalmology
Background: Interfacility transfer patients are associated with inferior outcomes, higher costs, and emergency department (ED) overcrowding. Previous studies have analyzed the general trends influencing interfacility transfers, but the patterns affecting ophthalmology-related transfer rates remain unclear. The purpose of this study was to identify and quantify the patterns of interfacility ophthalmology-specific transfers to the ED by temporal trends and to determine the type of ophthalmic services required by the transferred population.
Methods: All patients transferred to the ED of a single level 1 trauma center because of ophthalmology-related complaints from 2016 to 2023 were analyzed retrospectively. Transfer and nontransfer …
