Ovarian Cancer Survivorship Checklist, Emily Lott
Seton Hall University Dissertations and Theses (ETDs)
Background: The cancer survivorship population has been growing significantly in the last few years due to advancements in treatments such as immunotherapy, targeted medications and other types of chemotherapy. However, these survivors can have lifelong symptom burdens due to their diagnoses and adverse effects of their curative therapies. Symptoms are not always physical and can include emotional distress. Ovarian cancer patients in particular tend to have symptom complications because of their disease process. Symptoms include pain, constipation, neuropathy and fatigue. Ovarian cancer patients also experience a high level of cancer relapses. Early detection is key in cancer recurrences for patients …

The intricate relationship between COVID-19 and diabetes has garnered increasing attention within the medical community. Emerging evidence suggests that individuals with diabetes may experience heightened vulnerability to COVID-19 and, in some cases, develop diabetes as a post-complication following the viral infection. Additionally, it has been observed that patients taking cough medicine containing steroids may face an elevated risk of developing diabetes, further underscoring the complex interplay between these health factors. Based on previous research, we implemented deep-learning models to diagnose the infection via chest x-ray images in coronavirus patients. Three Thousand (3000) x-rays of the chest are collected through freely …

Our ability to sense and move our bodies relies on proprioceptors, sensory neurons that detect mechanical forces within the body. Different subtypes of proprioceptors detect different kinematic features, such as joint position, movement, and vibration, but the mechanisms that underlie proprioceptor feature selectivity remain poorly understood. Using single-nucleus RNA sequencing (RNA-seq), we found that proprioceptor subtypes in the Drosophila leg lack differential expression of mechanosensitive ion channels. However, anatomical reconstruction of the proprioceptors and connected tendons revealed major biomechanical differences between subtypes. We built a model of the proprioceptors and tendons that identified a biomechanical mechanism for joint angle selectivity …

Human noroviruses (HuNoVs) are the leading cause of acute gastroenteritis. In immunocompetent hosts, symptoms usually resolve within 3 days; however, in immunocompromised persons, HuNoV infection can become persistent, debilitating, and sometimes life-threatening. There are no licensed therapeutics for HuNoV due to a near half-century delay in its cultivation. Treatment for chronic HuNoV infection in immunosuppressed patients anecdotally includes nitazoxanide, a broad-spectrum antimicrobial licensed for treatment of parasite-induced gastroenteritis. Despite its off-label use for chronic HuNoV infection, nitazoxanide has not been clearly demonstrated to be an effective treatment. In this study, we standardized a pipeline for antiviral testing using multiple human …

A comprehensive atlas of cis-regulatory elements and their dynamic activity is necessary to understand the transcriptional basis of cellular structure maintenance, metabolism, and responses to the environment. Here we show, using matched single-nucleus chromatin accessibility and RNA-sequencing from juvenile male C57BL6 mice, an atlas of accessible chromatin regions in both normal and denervated skeletal muscles. We identified cell-type-specific cis-regulatory networks, highlighting the dynamic regulatory circuits mediating transitions between myonuclear types. Through comparison of normal and perturbed muscle, we delineated the reprogramming of cis-regulatory networks in response to denervation, described the interplay of promoters/enhancers and target genes. We further unveil a …

Thesis/ Dissertation Defenses
Pharmacogenomics (PGx) is a relatively new field of study. It links genetics to pharmacology since it deals with the influence of the genetic makeup of the individual on their ability to respond to specific medications. Some of the most important genes in this field, dubbed very important pharmacogenes (VIPs), belong to the cytochrome P450 (CYP) superfamily of drug metabolizing enzymes. The two members of this family that are the main focus of this thesis are CYP2C19 and CYP2D6. They play major roles in the metabolism of numerous medications, and it is therefore imperative that variations within those genes in various …

Journal of the Saudi Heart Association
A 35-year-old male, with a medical history of acute myocarditis, presented with palpitations. Further investigation revealed non-sustained ventricular tachycardia and a slightly reduced left ventricular systolic function. Cardiac magnetic resonance showed extended late gadolinium enhancement of the left ventricle and fat infiltration. Genetic testing was positive for a pathogenic desmoplakin mutation, fulfilling the criteria of arrhythmogenic left ventricular cardiomyopathy. In conclusion, the authors described a case of a mimicked acute myocarditis at a young age in a patient with an arrhythmogenic left ventricular cardiomyopathy. Therefore, the genetic study is essential for both diagnosis and management.

Axenfeld–Rieger anomaly (ARA) is a specific ocular disorder that is frequently associated with other systemic abnormalities. PITX2 and FOXC1 variants explain the majority of individuals with Axenfeld–Rieger syndrome (ARS) but leave ~30% unsolved. Here, we present pathogenic/likely pathogenic variants in nine families with ARA/ARS or similar phenotypes affecting five different genes/regions. USP9X and JAG1 explained three families each. USP9X was recently linked with syndromic cognitive impairment that includes hearing loss, dental defects, ventriculomegaly, Dandy–Walker malformation, skeletal anomalies (hip dysplasia), and other features showing a significant overlap with FOXC1-ARS. Anterior segment anomalies are not currently associated with USP9X, yet our cases …

Liquid biopsy, through isolation and analysis of disease-specific analytes, has evolved as a promising tool for safe and minimally invasive diagnosis and monitoring of tumors. It also has tremendous utility as a companion diagnostic allowing detection of biomarkers in a range of cancers (lung, breast, colon, ovarian, brain). However, clinical implementation and validation remains a challenge. Among other stages of development, preanalytical variables are critical in influencing the downstream cellular and molecular analysis of different analytes. Although considerable progress has been made to address these challenges, a comprehensive assessment of the impact on diagnostic parameters and consensus on standardized and …

Rab GTPases are important regulators of intracellular vesicular trafficking. RAB5C is a member of the Rab GTPase family that plays an important role in the endocytic pathway, membrane protein recycling and signaling. Here we report on 12 individuals with nine different heterozygous de novo variants in RAB5C. All but one patient with missense variants (n = 9) exhibited macrocephaly, combined with mild-to-moderate developmental delay. Patients with loss of function variants (n = 2) had an apparently more severe clinical phenotype with refractory epilepsy and intellectual disability but a normal head circumference. Four missense variants were investigated experimentally. In vitro biochemical …

This study aimed to determine whether serum metabolite and hormones of ewes with low body condition scores subjected to the melatonin plus progesterone-based protocol (MPE) would be similar to a single progesterone-based protocol (PE) and whether melatonin ear implant supplementation could increase pregnancy rate. Ninety suckled Merino ewes with singleton lamb were equally allocated in two experimental groups (MPE, PE) and a control group (CON) on days 53 ± 3 (D0) postpartum. Ewes in MPE group were subjected to exogenous melatonin plus progesterone-based protocol [Melatonin (day 0) + P4 insertion (day 35) + 500 IU eCG (day 42)]. Ewes in …

Comparison Of Carcass Characteristics, Meat Quality, And Fatty Acid Composition In Slowand Fast-Growing Broilers At Different Slaughter Weights, Büşra Yaranoğlu, Hi̇lal Çapar Akyüz, Esi̇n Ebru Onbaşilar
Turkish Journal of Veterinary & Animal Sciences
This study aimed to compare the slaughter, meat quality characteristics, and fatty acid composition of fast-growing (FAG) and slow-growing (SWG) broilers at different slaughter weights. In the experiments, a total of 90 carcasses were used: 45 SWG (Hubbard-Isa Red JA) and 45 FAG broilers (Ross 308) with 15 carcasses from each slaughter weight group (1500 ± 50 g, 2000 ± 50 g, 2500 ± 50 g). Hot carcass yield and cold carcass yield detected in the FAG broilers were higher than in SGW broilers (p < 0.001). As the weight of slaughter increased, hot carcass and cold carcass yield increased (p < 0.001). The breast percentage was significantly lower in the SWG broilers compared to the FAG broilers (p < 0.001). In contrast, thigh, wing, and abdominal fat percentages were higher in the SWG broilers (p < 0.001). The breast percentage increased as the slaughter weight increased (p < 0.001). SWG broilers had significantly lower pH, except for the pH 24 of the thigh meat (p < 0.01). The L*, a*, and b* values of the SWG broilers were lower than the FAG broilers (p < 0.01). In terms of a* values determined initially and at the 24th h in the breast and thigh meat, the 1500 g slaughter weight group had by far the highest value (p < 0.01). SWG broilers had significantly lower values in terms of cooking loss, water-holding capacity, and drip loss (p < 0.001). C18:2ω6 was detected at higher amounts in the FAG broilers (p < 0.05). The FAG broilers had higher PUFA, desired fatty acids, PUFA/SFA, and thrombogenic index values (p < 0.05). The results show that FAG broilers can meet the strong worldwide demand for meat quantity and quality. Slaughter weight changes only affected the quantity of the meat.

Testicular dysfunction, a severe secondary complication of diabetes, negatively affects reproductive health. Ghrelin is a peptide that plays a role in many metabolic events, such as stimulating growth hormones and regulating appetite, blood pressure, and reproduction. Metformin has antiinflammatory and antioxidant properties and is widely used to treat type 2 diabetes. Therefore, in this study, to investigate the effects of ghrelin and metformin on testicular function in experimental diabetic rats given streptozotocin, placed on a high-fat diet, and that had their sperm parameters evaluated, testosterone hormone, caspase 3 and iNOS expression, and histopathological examinations were performed. In total, 38 Wistar …

Arum rupicola Boiss var. rupicola is a preferred ingredient in traditional medicine in various countries, including Türkiye. The current study aims to establish the antiparasitic effect of A. rupicola Boiss var. rupicola on the eggs and larvae of T. canis in vitro. An ovicidal effect was not detected in the plant extracts, and embryogenesis in the T. canis eggs was not affected by the leaves' ethanolic extract and the distilled water extract of the rhizome during the incubation periods (6, 12, and 24 h). The larvicidal activity was observed in both extracts on the T. canis larvae after hatching. All …
