The Alleviating Effects Of Epidermal Growth Factor On An In Vitro Oral Mucositis Model In The Presence Of Fusobacterium Nucleatum, Tien Thi Trieu Truong

Chulalongkorn University Theses and Dissertations (Chula ETD)

Objectives: 5-fluorouracil (5-FU) is commonly used as a chemotherapy drug for head and neck cancers. However, it often produces an adverse cytotoxic effect on the oral epithelia leading to oral mucositis (OM) in cancer patients. Thus, reversing this cytotoxic effect locally with novel bioactive drugs or factors is necessary. This study aimed to investigate the rescue effect of Nicotiana benthamiana plant-derived epidermal growth factor (P-EGF) on human normal oral keratinocytes (NOKs) exposed to 5-FU-induced cytotoxicity in the presence of Fusobacterium nucleatum (Fn). Materials and Methods: Cytotoxicity and ATP-dependent proliferation and metabolism of NOK monolayer cultures were assessed after 48 hours …

Genetic Heterogeneity Of Glucose 6-Phosphate Dehydrogenase (G6pd) Regulatory Region In G6pd Deficient Patients, Lawrence Billy Vasco Djama

Chulalongkorn University Theses and Dissertations (Chula ETD)

G6PD deficiency is a genetic disorder caused by either suboptimal function of the G6PD enzyme or instability of the G6PD gene, resulting in insufficient production of the G6PD protein. This study investigates mutations in the regulatory regions of the G6PD gene using Nanopore MinION sequencing, as mutations outside the coding region can also affect G6PD activity. Blood samples from 89 individuals (73 adults and 16 newborns) with G6PD deficiency or intermediate levels but unidentified coding sequence mutations were analyzed. Genomic DNA was screened for mutations, enriched using degenerate oligonucleotide-primed PCR (DOP-PCR), and sequenced using the Oxford MinION platform. Bioinformatic analyzes …

Healthy ageing has become a goal to help older people maintain good-quality longevity. Researchers have found that epigenetic, environmental, and behavioural lifestyle factors influence the variation of lifespan and healthspan. This study aims to evaluate the age-associated epigenetic biomarkers among older people living in four major regions of Thailand, along with their association with lifestyle and sociodemographic characteristics. 214 participants passed the criteria and enrolled from the southern, northeastern, central, and northern areas of Thailand. Sociodemographic characteristics were recorded through interviews, and blood samples were collected to evaluate blood chemistry and epigenetic markers. SIRT1 and HMGB1 proteins in plasma were …

Volatile Organic Compound Profiles Identifiled In Vaginal Discharge Of Bitches During Estrous Cycle, Amornthep Preechatangkit Jan 2023

Volatile Organic Compound Profiles Identifiled In Vaginal Discharge Of Bitches During Estrous Cycle, Amornthep Preechatangkit

Chulalongkorn University Theses and Dissertations (Chula ETD)

Volatile organic compounds (VOCs) are the metabolites of microorganisms. VOCs are low molecular weight and easily evaporate at room temperature, all odorants are VOC. VOCs are different depending on health status and reproductive status. The objective of this study was to profile the VOCs in bitches during estrous cycle, and to compare VOCs between stages of estrous cycle and between pre-ovulatory, ovulation and post-ovulatory periods. A total of 23 normal, healthy bitches requested for ovulation timing as part of the routine breeding management were used in this study. All animals had a previous history of normal estrous cycle without any …

Inhibition Of Chondrocyte Apoptosis Via Fas/Fasl System In Knee Osteoarthritis Patients, Ousakorn Kamenkit Jan 2023

Inhibition Of Chondrocyte Apoptosis Via Fas/Fasl System In Knee Osteoarthritis Patients, Ousakorn Kamenkit

Chulalongkorn University Theses and Dissertations (Chula ETD)

Osteoarthritis (OA) is a degenerative rheumatic disease characterized by articular cartilage degeneration. This articular cartilage degradation is positively correlated with chondrocyte apoptosis. The Fas/Fas ligand (FasL) system is part of the extrinsic apoptosis pathway that regulates cell death through the binding of Fas on target cells to FasL on effector cells. Although Fas+ chondrocytes have been identified and the Fas/FasL system has been implicated in this apoptotic process, the apoptosis through chondrocyte-to-FasL+ cell interaction remains unclear. Therefore, this study aims to investigate Fas/FasL-mediated chondrocyte apoptosis focusing on cell-to-cell interactions. In our study, we conducted ex vivo experiments using flow cytometry …

Reproductive Performance Of Gilts And Sows In Gestation Group Housing And Free Farrowing System In Tropical Climate, Natchanon Dumniem

Chulalongkorn University Theses and Dissertations (Chula ETD)

Animal welfare is a rising topic in the swine industry, resulting in the adoption of alternative housing systems. However, the knowledge of management practices to mitigate the deleterious effects of these systems on reproductive performance in tropical environments is limited. This thesis aims to determine the impacts of alternative housing systems on the reproductive performance of sows and gilts, comprising 4 parts. i) To compare reproductive performance among conventional crates (CC), group housing gestation pen with temporary farrowing crate (PC), and group housing gestation pen and free-farrowing pen (PP). On average, the farrowing rate of sows in the CC group …

Analytical Performance Of Progesterone Testing Using Surface Plasmon Field-Enhanced Fluorescence Spectroscopy Technology In Dogs, Pimuk Sungprasit Jan 2023

Analytical Performance Of Progesterone Testing Using Surface Plasmon Field-Enhanced Fluorescence Spectroscopy Technology In Dogs, Pimuk Sungprasit

Chulalongkorn University Theses and Dissertations (Chula ETD)

A new clinical chemistry analyzer using surface plasmon field-enhanced fluorescence spectroscopy (SPFS) has been introduced into veterinary diagnostic laboratories as a point-of-care unit, enabling rapid turnaround times for both diagnosis and follow-up. Consequently, the test results must be accurate, precise, and reliable. This research aims to evaluate the method performance of the analyzer. The evaluation was conducted according to Clinical Laboratory Improvement Amendments (CLIA) guidelines using method validation. The performance evaluation included studies on imprecision (within-day and between-day), method comparison, recovery tests, and linearity for determining progesterone concentration. The method validation results showed good and acceptable precision. Serum spiking concentrations …

Metagenomic Analysis Using Oxford Nanopore For Fungal Identification In Hemoculture Sample, Nattapong Langsiri Jan 2023

Metagenomic Analysis Using Oxford Nanopore For Fungal Identification In Hemoculture Sample, Nattapong Langsiri

Chulalongkorn University Theses and Dissertations (Chula ETD)

Currently, with advancements in medical technology, the mortality rate from fungal infections has also increased, particularly since the late 20th century. The incidence of invasive fungal infections, especially bloodstream fungal infections, is rising. However, laboratory diagnosis of fungi from clinical specimens still faces significant limitations. Diagnosing fungi based on morphological characteristics to identify species level can take weeks, which is not aligned with the severity of the disease. Current molecular biology techniques also have limitations, such as PCR success rates and mixed infections. With significant advancements in sequencing technology leading to third-generation sequencing, there are notable benefits. Third-generation sequencing, or …
