Identification And Characterization Of Cyps Induced In The Drosophila Antenna By Exposure To A Plant Odorant, Shane R Baldwin, Pratyajit Mohapatra, Monica Nagalla, Rhea Sindvani, Desiree Amaya, Hope A Dickson, Karen Menuz

Department of Medicine Faculty Papers

Degrading Enzymes (ODEs). However, their contribution to olfactory signaling in vivo is poorly understood. This is due in part to the challenge of identifying which of the dozens of antennal-expressed CYPs might inactivate a given odorant. Here, we tested a high-throughput deorphanization strategy in Drosophila to identify CYPs that are transcriptionally induced by exposure to odorants. We discovered three CYPs selectively upregulated by geranyl acetate using transcriptional profiling. Although these CYPs are broadly expressed in the antenna in non-neuronal cells, electrophysiological recordings from CYP mutants did not reveal any changes in olfactory neuron responses to this odorant. Neurons were desensitized …

Cellular Therapy Updates In B-Cell Lymphoma: The State Of The Car-T, Zachary D Crees, Armin Ghobadi

2020-Current year OA Pubs

Non-Hodgkin Lymphoma accounts for >460,000 cases and >240,000 deaths globally and >77,000 cases and >20,000 deaths in the U.S. annually, with ~85% of cases being B-cell malignancies. Until recently, patients with relapsed/refractory B-cell lymphoma following standard chemotherapy in combination with anti-CD20 monoclonal antibodies and autologous stem cell transplantation experienced a median overall survival (OS) of <6 months. However, with the approval of four different CD-19 CAR-T therapies between 2017 and 2021, approximately 60-80% of patients receiving CAR-T therapy now achieve an objective response with >3 years median OS. Here, we review the current state of the art of CD19 CAR-T therapies for B-cell lymphomas, focusing on current updates in US FDA-approved products, along with their associated efficacy and toxicities. Lastly, we highlight a selection of promising clinical developments in …

Timing Of Antibiotic Therapy In The Icu, Marin H Kollef, Andrew F Shorr, Matteo Bassetti, Jean-Francois Timsit, Scott T Micek, Andrew P Michelson, Jose Garnacho-Montero

2020-Current year OA Pubs

Severe or life threatening infections are common among patients in the intensive care unit (ICU). Most infections in the ICU are bacterial or fungal in origin and require antimicrobial therapy for clinical resolution. Antibiotics are the cornerstone of therapy for infected critically ill patients. However, antibiotics are often not optimally administered resulting in less favorable patient outcomes including greater mortality. The timing of antibiotics in patients with life threatening infections including sepsis and septic shock is now recognized as one of the most important determinants of survival for this population. Individuals who have a delay in the administration of antibiotic …

Faculty Research 2021

BACKGROUND: Genomics-driven discoveries of microbial species have provided extraordinary insights into the biodiversity of human microbiota. In addition, a significant portion of genetic variation between microbiota exists at the subspecies, or strain, level. High-resolution genomics to investigate species- and strain-level diversity and mechanistic studies, however, rely on the availability of individual microbes from a complex microbial consortia. High-throughput approaches are needed to acquire and identify the significant species- and strain-level diversity present in the oral, skin, and gut microbiome. Here, we describe and validate a streamlined workflow for cultivating dominant bacterial species and strains from the skin, oral, and gut …

Athletic Training Educators’ Perceptions Of Interprofessional Education And Educational Strategies Used To Infuse Ipe Within Athletic Training Programs: A Mixed Methods Approach, Christina Orozco Nevers

Seton Hall University Dissertations and Theses (ETDs)

BACKGROUND: As athletic trainers (ATs) educators and professionals we recognize the importance of preparing our students to practice as part of an interprofessional team and acknowledge that this training must begin while they are in the academy. Although there is lots of information about interprofessional education (IPE) in various other healthcare professions (HCPs), there is limited information about how AT educators are infusing IPE into the curriculum. To maximize the development of interprofessional teaming practices in healthcare we must first understand the most effective ways to infuse IPE in AT professional programs given how compact the curriculum is. Educators must …

Genz And Mendell-Elston Estimation Of The High-Dimensional Multivariate Normal Distribution, Lucy Blondell, Mark Kos, John Blangero, Harald H. H. Goring

School of Medicine Publications

Statistical analysis of multinomial data in complex datasets often requires estimation of the multivariate normal (MVN) distribution for models in which the dimensionality can easily reach 10–1000 and higher. Few algorithms for estimating the MVN distribution can offer robust and efficient performance over such a range of dimensions. We report a simulation-based comparison of two algorithms for the MVN that are widely used in statistical genetic applications. The venerable Mendell- Elston approximation is fast but execution time increases rapidly with the number of dimensions, estimates are generally biased, and an error bound is lacking. The correlation between variables significantly affects …
