Magnetic hyperthermia (MH) harnesses the heat-releasing properties of superparamagnetic iron oxide nanoparticles (SPIONs) and has potential to stimulate immune activation in the tumor microenvironment whilst sparing surrounding normal tissues. To assess feasibility of localized MH in vivo, SPIONs are injected intratumorally and their fate tracked by Zirconium-89-positron emission tomography, histological analysis, and electron microscopy. Experiments show that an average of 49% (21–87%, n = 9) of SPIONs are retained within the tumor or immediately surrounding tissue. In situ heating is subsequently generated by exposure to an externally applied alternating magnetic field and monitored by thermal imaging. Tissue response to hyperthermia, …

Evaluating The Use Of Behavioral Skills Training To Teach Substitute Caregivers To Identify Hazards, Carlos Abarca Mar 2021

Evaluating The Use Of Behavioral Skills Training To Teach Substitute Caregivers To Identify Hazards, Carlos Abarca

USF Tampa Graduate Theses and Dissertations

The leading cause of death for children across the world is unintentional injuries (UNICEF 2001). Hazards such as accessible pools, poisons, and small ingestible items are the leading causes of unintentional injuries. Behavioral interventions such as Project 12-Ways/Safe Care have been used to teach parents how to be proactive in structuring a home free of accessible hazards by teaching the parents to identify and remove hazards in their home. Though the Project 12-Ways/Safe Care model has over 30 years of literature supporting its efficacy, the model has not been tested with substitute caregivers who often play a critical role in …

Marchigian Sardinian alcohol-preferring (msP) rats serve as a unique model of heightened alcohol preference and anxiety disorders. Their innate enhanced stress and poor stress-coping strategies are driven by a genetic polymorphism of the corticotropin-releasing factor receptor 1 (CRF1) in brain areas involved in glucocorticoid signaling. The activation of glucocorticoid receptors (GRs) regulates the stress response, making GRs a candidate target to treat stress and anxiety. Here, we examined whether mifepristone, a GR antagonist known to reduce alcohol drinking in dependent rats, decreases innate symptoms of anxiety in msPs. Male and female msPs were compared to non-selected Wistar counterparts across three …

The Effects Of Myelin On Macrophage Activation Are Phenotypic Specific Via Cpla2 In The Context Of Spinal Cord Injury Inflammation, Timothy J. Kopper, Bei Zhang, William M. Bailey, Kara E. Bethel, John C. Gensel

Physiology Faculty Publications

Spinal cord injury (SCI) produces chronic, pro-inflammatory macrophage activation that impairs recovery. The mechanisms driving this chronic inflammation are not well understood. Here, we detail the effects of myelin debris on macrophage physiology and demonstrate a novel, activation state-dependent role for cytosolic phospholipase-A2 (cPLA2) in myelin-mediated potentiation of pro-inflammatory macrophage activation. We hypothesized that cPLA2 and myelin debris are key mediators of persistent pro-inflammatory macrophage responses after SCI. To test this, we examined spinal cord tissue 28-days after thoracic contusion SCI in 3-month-old female mice and observed both cPLA2 activation and intracellular accumulation of lipid-rich myelin …

Comparison Of The Effectiveness Of Disinfectant-Impregnated Wipes Versus Detergent Wipes For Surface Decontamination, Jacob Amadin

USF Tampa Graduate Theses and Dissertations

Over time, there is an increased need to adopt more advanced and effective ways of disinfection. The emergence of the coronavirus-2019 (COVID-19) pandemic reminded the world on the importance of disinfection. Disinfection has been a routine in the healthcare setting. However, with the current pandemic problem, it is more prominently and persistently done in the domestic setting and other business settings beyond healthcare. As such, the use of most disinfectants and detergent wipes to achieve a near sanitized environment has become a common practice around the globe. This research presents the efficacy of disinfectant impregnated versus detergents wipes through comparison. …

Postoperative Pharmacologic Anticoagulation Following Temporal Lobe Resection Of A Gliosarcoma In A Hypercoagulable Patient, Alec Kellish, Siyuan Yu, Mark Heslin, Brian Gable, Gabrielle Hassinger

Cooper Rowan Medical Journal

Gliosarcomas are a rare subtype of glioblastomas associated with high rates of malignancy-associated venous thromboembolism (VTE). VTE risk is further increased in hypercoagulable patients upon discontinuing pharmacologic anticoagulation for surgery. We present a 60-year old obese male with history of hypercoagulability on apixaban who developed extensive thrombosis following resection of a gliosarcoma. Prior to temporal lobe resection, apixaban was discontinued and an IVC filter placed. On postoperative day 4, imaging revealed thrombosis above the IVC filter extending to the bilateral common, internal and external iliac, and femoral veins, requiring immediate anticoagulation and suction thrombectomy. Clinicians must balance the risk of …

Cell−surface polysaccharides are essential to many aspects of physiology, serving as a highly conserved evolutionary feature of life and as an important part of the innate immune system in mammals. Here, as simplified biophysical models of these sugar coatings, we present results of molecular dynamics simulations of hyaluronic acid and heparin brushes that show important effects of ion pairing, water dielectric decrease, and coion exclusion. As in prior studies of macromolecular crowding under physiologically relevant salt concentrations, our results show equilibria with electroneutrality attained through screening and pairing of brush anionic charges by monovalent cations at the atomistic detail. Department of Pathology and Laboratory Medicine

Background: The 2014 WHO Classification of ovarian neoplasms introduced a new entity of seromucinous tumors associated with endometriosis. These tumors encompassed a spectrum from benign to malignant and included seromucinous cystadenoma/ cystadenofibroma, seromucinous borderline tumor/atypical proliferative seromucinous tumor and seromucinous carcinoma. However, the 2020 WHO Classification of Female Genital Tumours removed seromucinous carcinomas as a distinct entity and recategorized them as Endometrioid carcinomas with mucinous differentiation. Here we describe clinico-morphologic features of seromucinous tumors recategorizing cases originally diagnosed as seromucinous carcinoma in light of 2020 WHO classification and present detailed review of literature.
Methods: Slides of seromucinous tumors were reviewed. …

Understanding The Prevalence Of Compassion Fatigue, Compassion Satisfaction, And Resilience In Primary Care, Christina M. O'Connell

Doctor of Nursing Practice Scholarly Projects

Compassion fatigue (CF), its components of burnout (BO) and secondary traumatic stress (STS), and compassion satisfaction (CS) affect healthcare providers and staff and can dramatically influence patient care and access to primary care services. Resilience is a tool which serves as a protective factor that can mitigate CF and enhance CS. There is a gap in research regarding the relationship between compassion fatigue, compassion satisfaction, and resilience among primary care providers and frontline staff. Additionally, the current COVID-19 pandemic has revealed healthcare’s deficits related to staff and provider burnout, a myriad of workforce safety issues, disparities in care, and a …

There are currently limited Food and Drug Administration (FDA)-approved drugs and vaccines for the treatment or prevention of Coronavirus Disease 2019 (COVID-19). Enhanced understanding of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) infection and pathogenesis is critical for the development of therapeutics. To provide insight into viral replication, cell tropism, and host-viral interactions of SARS-CoV-2, we performed single-cell (sc) RNA sequencing (RNA-seq) of experimentally infected human bronchial epithelial cells (HBECs) in air-liquid interface (ALI) cultures over a time course. This revealed novel polyadenylated viral transcripts and highlighted ciliated cells as a major target at the onset of infection, which we …

Faculty Research 2021

Employers have begun to offer voluntary workplace genomic testing (wGT) as part of employee wellness benefit programs, but few empirical studies have examined the ethical, legal, and social implications (ELSI) of wGT. To better understand employee perspectives on wGT, employees were surveyed at a large biomedical research institution. Survey respondents were presented with three hypothetical scenarios for accessing health-related genomic testing: via (1) their doctor; (2) their workplace; and 3) a commercial direct-to-consumer (DTC) genetic testing company. Overall, 594 employees (28%) responded to the survey. Respondents indicated a preference for genomic testing in the workplace setting (70%; 95% CI 66-74%), …

Combining Multicolor Fish With Fluorescence Lifetime Imaging For Chromosomal Identification And Chromosomal Sub Structure Investigation, Archana Bhartiya, Ian Robinson, Mohammed Yousuf, Stanley W. Botchway

Centre for Regenerative Medicine & Stem Cell Research

Understanding the structure of chromatin in chromosomes during normal and diseased state of cells is still one of the key challenges in structural biology. Using DAPI staining alone together with Fluorescence lifetime imaging (FLIM), the environment of chromatin in chromosomes can be explored. Fluorescence lifetime can be used to probe the environment of a fluorophore such as energy transfer, pH and viscosity. Multicolor FISH (M-FISH) is a technique that allows individual chromosome identification, classification as well as assessment of the entire genome. Here we describe a combined approach using DAPI as a DNA environment sensor together with FLIM and M-FISH …

Faculty Research 2021

Immune cell activation assays have been widely used for immune monitoring and for understanding disease mechanisms. However, these assays are typically limited in scope. A holistic study of circulating immune cell responses to different activators is lacking. Here we developed a cost-effective high-throughput multiplexed single-cell RNA-seq combined with epitope tagging (CITE-seq) to determine how classic activators of T cells (anti-CD3 coupled with anti-CD28) or monocytes (LPS) alter the cell composition and transcriptional profiles of peripheral blood mononuclear cells (PBMCs) from healthy human donors. Anti-CD3/CD28 treatment activated all classes of lymphocytes either directly (T cells) or indirectly (B and NK cells) …

Adolescent alcohol use is one of the strongest predictors for the development of an alcohol use disorder (AUD). Notably, this period of risk coincides with the development of affective disorders, which disproportionately impact and drive problematic drinking behavior in women. Stress is a particularly salient factor that drives relapse during periods of abstinence. Previous work in our lab has shown that adolescent intermittent ethanol vapor (AIE) produces sex-dependent changes in glutamatergic activity in the bed nucleus of the stria terminalis (BNST) and behavioral outcomes following acute restraint stress in adulthood. In females, AIE disrupts group 1 metabotropic glutamate (mGlu1/5) receptor …

Family-Based Caregiving: Does Lumping Asian Americans Together Do More Harm Than Good?, Suryadewi E. Nugraheni, Julia F. Hastings

Journal of Social, Behavioral, and Health Sciences

Asian American family caregivers have gained increased attention due to the need to provide life-sustaining aid at home given the rising numbers of older adults. This article reflects upon caregiving-related research studies that have overlooked the circumstances Asian American caregivers bring to the home-care context. Policies written to address community needs tend to omit the social circumstances many Asian American caregivers must face when trying to take advantage of programs and services. For example, the eligibility requirements fail to recognize distinctive cultural values embedded within the caregiving processes. Further, most Asian American data is aggregated. Aggregating data by ethnicity limits …
