The Effect Of Walking Speed And Magnitude Of Perturbation On Compensatory Responses, Farahnaz Fallah Tafti, Jenna Yentes Mar 2021

The Effect Of Walking Speed And Magnitude Of Perturbation On Compensatory Responses, Farahnaz Fallah Tafti, Jenna Yentes

UNO Student Research and Creative Activity Fair

Imposing a perturbation can induce an unstable condition during gait. Measuring the compensatory responses may be an accurate indicator of the ability to control stability. Responses to a perturbation can be quantified through the perturbed walking pattern deviations from an unperturbed condition. The aim of this study was to determine the effect of perturbation magnitude or speed of walking on compensatory responses to lateral perturbations during walking. Twenty healthy young participants recruited to this study and they were perturbed with the medium magnitude of perturbation, while walking on the treadmill at three different speeds (slow, preferred, and fast). They also …

Metabolism And Oncology: The Role Of Nutrition In Improving Cancer Outcomes, Michael A. Chapek Mar 2021

Metabolism And Oncology: The Role Of Nutrition In Improving Cancer Outcomes, Michael A. Chapek

University Honors Theses

Despite significant advances in oncologic treatment, underlying cancer-associated metabolic derangements and nutritional needs remain largely neglected in cancer care. Cancer cachexia and protective metabolic changes exhibited by cancerous cells continue to pose formidable barriers to improving therapeutic outcomes and quality of life for patients. Cancer has traditionally been viewed as a proliferative disease caused by genetic mutations, but newer perspectives suggest that it is primarily a metabolic disease. The present paper discusses the etiology of cachexia and sarcopenia, and nutritional interventions that can be offered to patients suffering from these wasting disorders. The role of inflammation in cancer and the …

Colon adenocarcinoma is a common cause of cancer-related deaths worldwide. Epithelial-mesenchymal transition is a major regulator of cancer metastasis, and increased understanding of this process is essential to improve patient outcomes. Long non-coding RNA (lncRNA) are important regulators of carcinogenesis. To identify lncRNAs associated with colon carcinogenesis, we performed an exploratory differential gene expression analysis comparing paired colon adenocarcinoma and normal colon epithelium using an RNA-sequencing data set. This analysis identified lncRNA ZFAS1 as significantly increased in colon cancer compared to normal colon epithelium. This finding was validated in an institutional cohort using laser capture microdissection. ZFAS1 was also found …

Properties Of Multivesicular Release From Mouse Rod Photoreceptors Support Transmission Of Single-Photon Responses, Cassandra L. Hays, Asia L. Sladek, Greg D. Field, Wallace B. Thoreson

Journal Articles: Ophthalmology

Vision under starlight requires rod photoreceptors to transduce and transmit single-photon responses to the visual system. Small single-photon voltage changes must therefore cause detectable reductions in glutamate release. We found that rods achieve this by employing mechanisms that enhance release regularity and its sensitivity to small voltage changes. At the resting membrane potential in darkness, mouse rods exhibit coordinated and regularly timed multivesicular release events, each consisting of ~17 vesicles and occurring two to three times more regularly than predicted by Poisson statistics. Hyperpolarizing rods to mimic the voltage change produced by a single photon abruptly reduced the probability of …

Using True Experiments To Study Culture: Manipulations, Measurement Issues, And The Question Of Appropriate Control Groups, Christine Ma-Kellams Mar 2021

Using True Experiments To Study Culture: Manipulations, Measurement Issues, And The Question Of Appropriate Control Groups, Christine Ma-Kellams

Faculty Research, Scholarly, and Creative Activity

Social group memberships are primarily studied in quasi-experimental contexts, but how can culture, class and gender be manipulated in true experimental designs? This review highlights the different empirical strategies that can be used to manipulate “culture” as it relates to race/ethnicity (activation of thinking styles, language, and priming of cultural constructs), class (social standing, group status, or perceived social status), and gender (role salience, gender identity, sex hormone administration). I review measurement issues related to manipulation checks and the problem of what construct is tapped by the manipulation, appropriate control groups, and intersectional identities or group memberships.

Serial block-face scanning electron microscopy (SBF-SEM) allows for the collection of hundreds to thousands of serially-registered ultrastructural images, offering an unprecedented three-dimensional view of tissue microanatomy. While SBF-SEM has seen an exponential increase in use in recent years, technical aspects such as proper tissue preparation and imaging parameters are paramount for the success of this imaging modality. This imaging system benefits from the automated nature of the device, allowing one to leave the microscope unattended during the imaging process, with the automated collection of hundreds of images possible in a single day. However, without appropriate tissue preparation cellular ultrastructure can …

Loss Of Bone Morphogenetic Protein-Binding Endothelial Regulator Causes Insulin Resistance, Hua Mao, Luge Li, Qiying Fan, Aude Angelini, Pradip K Saha, Huaizhu Wu, Christie M Ballantyne, Sean M Hartig, Liang Xie, Xinchun Pi

Faculty, Staff and Students Publications

Accumulating evidence suggests that chronic inflammation of metabolic tissues plays a causal role in obesity-induced insulin resistance. Yet, how specific endothelial factors impact metabolic tissues remains undefined. Bone morphogenetic protein (BMP)–binding endothelial regulator (BMPER) adapts endothelial cells to inflammatory stress in diverse organ microenvironments. Here, we demonstrate that BMPER is a driver of insulin sensitivity. Both global and endothelial cell-specific inducible knockout of BMPER cause hyperinsulinemia, glucose intolerance and insulin resistance without increasing inflammation in metabolic tissues in mice. BMPER can directly activate insulin signaling, which requires its internalization and interaction with Niemann-Pick C1 (NPC1), an integral membrane protein that …

Data For "Subtype-Selective Positive Modulation Of Sk Channels Depends On The Ha/Hb Helices", Miao Zhang, Meng Cui Mar 2021

Data For "Subtype-Selective Positive Modulation Of Sk Channels Depends On The Ha/Hb Helices", Miao Zhang, Meng Cui

Pharmacy Faculty Data Sets

In the activated state of small-conductance Ca2+-activated potassium (SK) channels, calmodulin interacts with the HA/HB helices and the S4-S5 linker. CyPPA potentiates SK2a and SK3 channel activity but not the SK1 and IK subtypes. Here, we report that the subtype-selectivity of CyPPA relies on the HA/HB helices. Mutating residues in the HA (V420) and HB (K467) helices of SK2a channels to their equivalent residues in IK channels diminished the potency of CyPPA. CyPPA elicited prominent responses on mutant IK channels with an arginine residue in the HB helix substituted for its equivalent lysine residue in the SK2a channels …

Structural And Functional Features Of The P2x4 Receptor: An Immunological Perspective, Jean M. Kanellopoulos, Cássio Luiz Coutinho Almeida-Da-Silva, Sirje Rüütel Boudinot, David M. Ojcius

All Dugoni School of Dentistry Faculty Articles

Extracellular nucleotides are important mediators of activation, triggering various responses through plasma membrane P2 and P1 receptors. P2 receptors are further subdivided into ionotropic P2X receptors and G protein-coupled P2Y receptors. P2X4 is an ATP-gated cation channel broadly expressed in most tissues of the body. Within the P2X family, P2X4 has a unique subcellular distribution, being preferentially localized in lysosomes. In these organelles, high ATP concentrations do not trigger P2X4 because of the low pH. However, when the pH increases to 7.4, P2X4 can be stimulated by intra-lysosomal ATP, which is in its active, tetra-anionic form. Elucidation of P2X4, P2X3 …

Faculty Research 2021

Despite the correlation of clinical outcome and molecular subtypes of high-grade serous ovarian cancer (HGSOC), contemporary gene expression signatures have not been implemented in clinical practice to stratify patients for targeted therapy. Hence, we aimed to examine the potential of unsupervised matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI-IMS) to stratify patients who might benefit from targeted therapeutic strategies. Molecular subtyping of paraffin-embedded tissue samples from 279 HGSOC patients was performed by NanoString analysis (ground truth labeling). Next, we applied MALDI-IMS paired with machine-learning algorithms to identify distinct mass profiles on the same paraffin-embedded tissue sections and distinguish HGSOC subtypes by …

Examining The Mechanistic Roles Of Integrin Alpha-6 In Cancer Metastasis., Chase T. Clark

Honors College Theses

Metastasis- the spread of cancer cells from the primary tumor to the surrounding tissues- is responsible for 90% of cancer deaths. Integrin alpha-6 (ITGA6) is a specific transmembrane cell surface protein that functions in cell surface adhesion and signaling. ITGA6 is upregulated in many types of cancers and promotes the migration and invasion of cancer cells to surrounding tissues. It is my objective to determine the mechanism through which ITGA6 facilitates the migration of cancer cells through the extracellular matrix (ECM). These experiments helped to establish the role of ITGA6 in tumor development and provide focus for possible chemotherapeutic treatment. …

Does Religious Belief Affect Attitudes Towards Mental Illness?, Robert B. Conway

Honors College Theses

There has been an increase in recent years in the effort to raise awareness of mental health issues, particularly suicide. However, awareness does not equate to understanding and there has been little in the way of public education. One of the biggest obstacles faced by those suffering from a mental illness is the public stigma associated with it. Given the high percentage of the population in the South who identify as being religious, this study looked at the relationship between religiosity and public stigma towards mental illness. Two hundred forty-two university students drawn from undergraduate biology and nursing classes completed …
