Background. Ethanol (EtOH) consumption is known to affect multiple organs; this is unsurprising, as the concentration of EtOH in the blood at relevant doses reaches the millimolar range. The overarching goal of this dissertation was to elucidate mechanisms of alcohol-induced organ injury, specifically the effects of alcohol on the hepatic extracellular matrix (ECM) proteome, the alcoholic hepatitis (AH) plasma peptidome, and the effects of alcohol on the renal cortex proteome and transcriptome. Mental health is an all-encompassing term that focuses on a person’s ability to achieve work-life balance inclusive of positive mental health and mental ill-health. Current trends suggest an overall increase in mental ill-health for undergraduate and graduate students. McMillan and Chavis’ sense of community theory provided the theoretical framework for this study. This study aimed to examine whether speech-language pathology graduate students’ sense of classroom community and perceived learning predict anxiety levels while controlling for demographic variables. Background: Porphyromonas gingivalis, a major pathogen associated with chronic periodontitis, secretes variety of proteins, majority of which begins with glutamine. Several of these proteins were found with pyroglutamate (pGlu) at N-terminus suggesting the presence of this posttranslational modification pathway in P.gingivalis. The observation that N-terminal glutamine is over-represented as the first amino acid after signal peptide cleavage, and subsequent confirmation of pGlu formation on the nascent protein via mass spectrometry, led us to conclude that an enzyme must be present as the executor of this reaction. Muscle injury is a common side effect of radiation treatment for head and neck cancer. To increase understanding of muscle injury related dysfunction, we investigated the effects of oral swallowing function after cryoinjury to mylohyoid muscle in rats. The hypothesis is that injury to the mylohyoid delays the temporal licking pattern, resulting in aberrant drinking behaviors. Six rats received bilateral mylohyoid injuries by applying a 3mm cryoprobe. Licking behavior was measured by electrophysiological recordings of rhythmic tongue movements in a ten-minute drinking session taken pre-and post-injury (one-and two-weeks). Environmental pollution is not distributed equally across members of society. Low-income individuals are more likely to live near waste sites and other sources of pollution, and, therefore, face greater exposure to environmental health hazards. One such community in Louisville, Kentucky, the Riverside Gardens neighborhood, consists of approximately 300 homes that are surrounded on three sides by industry and industrial waste, including a remediated Environmental Protection Agency Superfund site, a former coal-burning power plant and coal ash storage site, and multiple chemical, rubber, and plastic manufacturing companies. Bis-thiosemicarbazones (BTSC) and their metal chelates have properties that are useful in several different scientific fields. These systems have already received attention in major fields of biology and engineering. Hydrogen evolution reaction (HER) catalysts need to be cheap and operate under minimal overpotentials with a long lifetime. The treatment of cancer requires, novel agents that have potent cytotoxic activity against cancer cells while displaying minimal side effects. This study’s purpose was to determine a possible difference in hemoglobin A1c levels and sport participation in children with type 1 diabetes. Patients with provided consent via an iPad electronic informed consent/assent (preamble) completed a physical activity and sport participation survey. The survey was linked to their medical chart including demographic and diabetes medical history (e.g. HbA1c). The analyses provided a sample of 120 participants, 40 in the no sport participation group and 78 in the sport participation group with 56 females and 64 males, with an average age12.94±2.8 years. When treating patients with existing restorations in which indirect restoration placement is indicated and crown lengthening surgery is contraindicated, dentists may choose to finish the crown margin on an existing restoration. The interface between the two dental restorations (resin composite and ceramic crown) may be more susceptible to microleakage and fluid staining, reducing restoration longevity. As the field of Public Health has shifted from a biomedical paradigm to a social ecological approach, focusing on health equity and social determinants of health, it has increasingly acknowledged the roles of culture and trauma in health outcomes. The field has responded by modifying some research practices, but continues to prioritize conventional methodologies—with few options designed to foreground trauma- and cultural-responsiveness. This thesis describes the development of a novel lectin-Fc fusion protein and its antiviral and anti-cancer activity. The molecule, Avaren-Fc (AvFc), is a fusion of a variant of the actinomycete lectin actinohivin (Avaren) and the Fc region of human IgG1, and is selective for the terminal α1,2-mannose residues found at the ends of high-mannose-type glycans that can be found on the surface of certain heavily glycosylated viruses and cancer cells. Bullying is a pervasive societal issue that is consistently linked to negative outcomes that are emotionally, socially, behaviorally, and medically related. Most youth will encounter this negative life event in their childhood. The purpose of this dissertation was to use a diathesis-stress model perspective to assess the relations between the negative life event of bullying involvement, youths’ mental health, and youth weight status. This paper depicts a U.S. veteran and graduate student’s experience using meditation, artmaking, and journaling to target the effects of trauma and its symptoms of post-traumatic stress disorder and dissociation. This is a self-study utilizing art as the main method for communication and knowing. Methods: An online questionnaire was developed and used to collect data. A mixed-mode approach was followed to recruit the participants. We tested the hypothesis that whole‐body passive heat stress reduces arterial stiffness in older adults. At preheat stress (baseline) and when core temperature was elevated by 0.6 ± 0.2°C (mild) and 1.2 ± 0.3°C (moderate), arterial stiffness was measured in eight healthy younger (26 ± 5 years) and eight healthy older (70 ± 4 years) adults in the supine position. Arterial stiffness was estimated from carotid‐to‐femoral pulse wave velocity (cfPWV, applanation tonometry). cfPWV was higher at baseline in older adults (8.8 ± 2.3 m/sec vs. 5.6 ± 0.9 m/sec, P < 0.01) and this difference was maintained throughout passive heat stress (P < 0.01). cfPWV did not change (P ≥ 0.49) with passive heat stress in either …

Growing evidence that speech perception tasks elicit sensorimotor activity, and that this activity varies due to context, sex, cognitive load, and cognitive ability. However, it is unknown as to whether the sex of the speaker and demands of the task differentially effect males and females during speech perception tasks. In this article, the investigators present a new method using a deep learning approach to diagnose schizophrenia. In the experiment presented, the investigators have used a secondary dataset provided by National Institutes of Health. The aforementioned experimentation involves analyzing this dataset for existence of schizophrenia using traditional machine learning approaches such as logistic regression, support vector machine, and random forest. This is followed by application of deep learning techniques using three hidden layers in the model. The reaction between transglutaminase factor XIII (FXIII) and fibrin is a key process in the final stages of blood coagulation. The αC region of fibrin contains three reactive glutamines (Q237, Q328, and Q366) which are crosslinked by FXIII, as well as a FXIII anchoring site (E396). Previous work has helped elucidate the FXIII-αC interaction, but much is still unknown about the key residues in the αC region.

Background: Human papillomavirus (HPV) vaccination has immense potential to prevent HPV-related infections but vaccination rates remain suboptimal. Nurses could potentially increase vaccination rates by providing recommendations and addressing vaccine concerns. As student nurses’ transition into practice, it is important to identify factors that could influence their attitudes and uptake of the HPV vaccine; however, these factors remain unknown.

Background: Disparities in early language development have been noted for children from low-income homes, which can impact their educational and health trajectory. The quantity and quality of language exposure in the home environment influences a child’s language development. Adult attachment style is a predictor of parenting behavior and could impact critical interactions between the parent and child.
