Pras: Predicting Functional Targets Of Rna Binding Proteins Based On Clip-Seq Peaks., Jianan Lin, Yuping Zhang, Wayne N Frankel, Zhengqing Ouyang

Faculty Research 2019

RNA-protein interaction plays important roles in post-transcriptional regulation. Recent advancements in cross-linking and immunoprecipitation followed by sequencing (CLIP-seq) technologies make it possible to detect the binding peaks of a given RNA binding protein (RBP) at transcriptome scale. However, it is still challenging to predict the functional consequences of RBP binding peaks. In this study, we propose the Protein-RNA Association Strength (PRAS), which integrates the intensities and positions of the binding peaks of RBPs for functional mRNA targets prediction. We illustrate the superiority of PRAS over existing approaches on predicting the functional targets of two related but divergent CELF (CUGBP, ELAV-like …

Faculty Research 2019

The Long interspersed nuclear element 1 (LINE-1) is a primary source of genetic variation in humans and other mammals. Despite its importance, LINE-1 activity remains difficult to study because of its highly repetitive nature. Here, we developed and validated a method called TeXP to gauge LINE-1 activity accurately. TeXP builds mappability signatures from LINE-1 subfamilies to deconvolve the effect of pervasive transcription from autonomous LINE-1 activity. In particular, it apportions the multiple reads aligned to the many LINE-1 instances in the genome into these two categories. Using our method, we evaluated well-established cell lines, cell-line compartments and healthy tissues and …

Transcriptome Profiling Of Brain Myeloid Cells Revealed Activation Of Itgal, Trem1, And Spp1 In Western Diet-Induced Obesity., Hongtian Yang, Leah C. Graham, Alaina M Reagan, Weronika Grabowska, William H. Schott, Gareth R Howell

Faculty Research 2019

BACKGROUND: Environmental factors are critical in the development of age-related cognitive decline and dementia. A western diet (WD) can cause nutrient deficiency and inflammation that could impact cognition directly. It is increasingly recognized that innate immune responses by brain myeloid cells, such as resident microglia, and infiltrating peripheral monocytes/macrophages may represent an essential link between a WD, cognitive decline, and dementia. Our previous data demonstrated that chronic consumption of a WD induced inflammation through brain myeloid cells in aging mice and a mouse model of Alzheimer's disease (AD). However, the subtypes of myeloid cells that contribute to the WD-induced inflammation …

The Relationship Of Motivational Climates, Mindsets, And Goal Orientations To Grit In Male Adolescent Soccer Players, Erin Albert, Trent A. Petrie, E. Whitney G. Moore Aug 2019

The Relationship Of Motivational Climates, Mindsets, And Goal Orientations To Grit In Male Adolescent Soccer Players, Erin Albert, Trent A. Petrie, E. Whitney G. Moore

Kinesiology, Health and Sport Studies

Across a variety of domains, such as in academics and in the military, grit is a significant predictor of performance, even after controlling for dimensions of innate ability. However, little is known about how grit develops or the psychosocial factors that may contribute to its presence (i.e. motivational climates, mindsets, and goal orientations). Although sport is a natural context in which to examine grit, both as a predictor and an outcome, few studies have done so. Thus, within the framework of achievement motivation theory, we examined the relationships of the perceived motivational climate created by coaches (task-involving vs. ego-involving), athletes’ …

The aim of this study was to investigate the effects of regular post-exercise whole-body cryotherapy (WBC) on physiological and performance adaptations to high-intensity interval training (HIT). In a two-group parallel design, twenty-two well-trained males performed four weeks of cycling HIT, with each session immediately followed by 3 min of WBC (−110 °C) or a passive control (CON). To assess the effects of WBC on the adaptive response to HIT, participants performed the following cycling tests before and after the training period; a graded exercise test (GXT), a time-to-exhaustion test (Tmax), a 20-km time trial (20TT), and …

Life Satisfaction And Healthcare Utilization Among Immigrants To The United States, Merritt Christian Rachel Harrelson Aug 2019

Life Satisfaction And Healthcare Utilization Among Immigrants To The United States, Merritt Christian Rachel Harrelson

LSU Master's Theses

Previous research suggests a relationship between life satisfaction and healthcare utilization or HCU (Habibov & Afandi, 2016; Kim, Park, Sun, Smith, & Peterson, 2014). However, no study was found that examined this relationship among elderly American immigrants. The purpose of this study was to determine whether life satisfaction is linked to HCU for this population. The researcher of the current study applied binary logistic regression to data taken from the Health and Retirement Survey to determine whether changes in life satisfaction could be linked to variance in HCU across seven domains, including hospitalization, overnight nursing home stays, doctors visits, dentist …

Multiple Sclerosis (MS) is an autoimmune disease of the central nervous system with prominent neurodegenerative components. The triggering and progression of MS is associated with transcriptional and epigenetic alterations in several tissues, including peripheral blood. The combined influence of transcriptional and epigenetic changes associated with MS has not been assessed in the same individuals. Here we generated paired transcriptomic (RNA-seq) and DNA methylation (Illumina 450 K array) profiles of CD4+ and CD8+ T cells (CD4, CD8), using clinically accessible blood from healthy donors and MS patients in the initial relapsing-remitting and subsequent secondary-progressive stage. By integrating the output of a …

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

Contact sites of endoplasmic reticulum (ER) and mitochondria locally convey calcium signals between the IP3 receptors (IP3R) and the mitochondrial calcium uniporter, and are central to cell survival. It remains unclear whether IP3Rs also have a structural role in contact formation and whether the different IP3R isoforms have redundant functions. Using an IP3R-deficient cell model rescued with each of the three IP3R isoforms and an array of super-resolution and ultrastructural approaches we demonstrate that IP3Rs are required for maintaining ER-mitochondrial contacts. This role is independent of calcium fluxes. We also show that, while each isoform can support contacts, type …

Human Polyomavirus Jcpyv And Its Role In Progressive Multifocal Leukoencephalopathy And Oncogenesis, Luis Del Valle, Sergio Piña-Oviedo

School of Medicine Faculty Publications

The human neurotropic virus JCPyV, a member of the family, is the opportunistic infectious agent of Progressive Multifocal Leukoencephalopathy (PML), a fatal disease seen in severe immunosuppressive conditions and, during the last decade, in patients undergoing immunotherapy. JCPyV is a ubiquitous pathogen with up to 85% of the adult population word-wide exhibiting antibodies against it. Early experiments demonstrated that direct inoculation of JCPyV into the brain of different species resulted in the development of brain tumors and other neuroectodermal-derived neoplasias. Later, several reports showed the detection of viral sequences in medulloblastomas and glial tumors, as well as expression of the …

Regulation of the development, production, marketing, and sales of medical pharmaceuticals and devices in the United States fall under the regulatory functions of the Food and Drug Administration (FDA). The current FDA approval process takes an average of 10 years from start to completion, and costs over $100 million. As a result, companies use many different methods to find additional use of their drugs through marketing directly to the physician, or recycling of previously approved drug moieties. In this work, an evaluation of the in vitro and ex vivo biocompatibility of polymers and drugs in different phases of FDA approval …

Organic Food Health Benefits Have Been Hard To Assess, But That Could Change, Cynthia Curl Aug 2019

Organic Food Health Benefits Have Been Hard To Assess, But That Could Change, Cynthia Curl

Public and Population Health Faculty Publications and Presentations

“Organic” is more than just a passing fad. Organic food sales totaled a record US$45.2 billion in 2017, making it one of the fastest-growing segments of American agriculture. While a small number of studies have shown associations between organic food consumption and decreased incidence of disease, no studies to date have been designed to answer the question of whether organic food consumption causes an improvement in health.

I’m an environmental health scientist who has spent over 20 years studying pesticide exposures in human populations. Last month, my research group published a small study that I believe suggests a path forward …
