Objective: Organ radiation dose from a CT scan, calculated by CT dosimetry software, can be combined with cancer risk data to estimate cancer incidence resulting from CT exposure. We aim to determine to what extent the use of improved anatomical representation of the adult human body “phantom” in CT dosimetry software impacts estimates of radiation dose and cancer incidence, to inform comparison of past and future research.
Methods: We collected 20 adult cases for each of three CT protocols (abdomen/pelvis, chest and head) from each of five public hospitals (random sample) (January-April inclusive 2010) and three private clinics (self-report). Organ …

We have previously demonstrated that the poly-arginine peptide R18 can improve histological and functional outcomes following traumatic brain injury (TBI) in the Sprague–Dawley rat. Since D-enantiomer peptides are often exploited in pharmacology for their increased stability and potency, the present study compared the effects of R18 and its D-enantiomer, R18D, following TBI in the Long-Evans rat. Following a closed-head impact delivered via a weight-drop apparatus, peptide was administered at a dose of 1000 nmol/kg at 30 min after TBI. Treatment with R18D, but not R18 resulted in significant reductions in sensorimotor (p = 0.026) and vestibulomotor (p = 0.049) deficits …

Plasmodium falciparum histone deacetylases (PfHDACs) are an important class of epigenetic regulators that alter protein lysine acetylation, contributing to regulation of gene expression and normal parasite growth and development. PfHDACs are therefore under investigation as drug targets for malaria. Despite this, our understanding of the biological roles of these enzymes is only just beginning to emerge. In higher eukaryotes, HDACs function as part of multi-protein complexes and act on both histone and non-histone substrates. Here, we present a proteomics analysis of PfHDAC1 immunoprecipitates, identifying 26 putative P. falciparum complex proteins in trophozoite-stage asexual intraerythrocytic parasites. The …

The aim of this study was to examine the content validity, construct validity and reliability of the newly developed Basketball Jump Shooting Accuracy Test (BJSAT). Basketball athletes from different playing levels (State Basketball League [SBL], n = 30, age: 22.7 ± 6.1 yr; SBL Division I, n = 11, age: 20.6 ± 2.1 yr) completed four separate trials of the BJSAT with each trial consisting of shot attempts from two- and three-point distances at pre-determined court locations. Each shot attempt was scored utilising a criteria where higher scores were given when greater accuracy was exhibited. The BJSAT detected a significant, …
Mechanism Of Nanogp8 In Glioblastoma Multiforme, Jonhoi Smith
Mechanism Of Nanogp8 In Glioblastoma Multiforme, Jonhoi Smith
Electronic Theses and Dissertations
Glioblastoma multiforme (GBM) is an incurable brain tumor, with patients only expecting to live 15 to 16 months post-diagnosis with the most current treatments options including surgery, chemotherapy, and radiation. How GBM resist treatment is still not very well known. However, cancer stem cells (CSCs), a subset of cells in GBM tissue considered responsible for therapeutic resistance and the poor patient prognosis. NANOG, a homeobox gene, is responsible for maintaining pluripotency of embryonic stem cells and is observed in CSCs. NANOGP8, a retrogene in the NANOG family is expressed in CSCs and provides cells with stem-like characteristics previously observed in …

Development Of A Functional Testing Platform For The Sensory Segment Of The Neuromuscular Reflex Arc, Alisha Colon
Electronic Theses and Dissertations
Investigations of human biology and disease have been hindered by the use of animal models. The information obtained from such studies often results in clinically irrelevant results and drug trial failures. Additionally, several governing bodies have been formulating legislation to move away from animal models and toward more ethical and efficient testing platforms for drug discovery and cosmetic research. As an answer to these issues, "body-on-a-chip" systems have been a rapidly developing field which easily recapitulates in vivo functionality, providing a more relevant, repeatable, and ethical testing platform to better predict biology. These systems can be used as human-based testing …

Sneathia amnii is a poorly characterized gram-negative anaerobe that commonly colonizes the vagina. It has been linked to many obstetric disorders, including preterm labor, preeclampsia, and chorioamnionitis. S. amnii lyses human red blood cells, and we aimed to identify the hemolysin. We identified two genes that appear to encode transporter and effector components of a two-partner secretion system. The putative effector, which we refer to as SaFHA, contains a domain with amino acid similarity to the filamentous hemagglutinin (FHA) of Bordetella pertussis and its predicted structure suggests it may form a transmembrane channel or pore. Thus, we hypothesized that SaFHA …

Posttraumatic stress disorder (PTSD) and alcohol use disorder (AUD) commonly co-occur, share latent genetic risk, and are associated with many negative public health outcomes. Via a self-medication framework, trauma-related drinking to cope (TRD), an unexplored phenotype to date, may help explain why these two disorders co-occur, thus serving as an essential target for treatment and prevention efforts. This study sought to create a novel measure of TRD and to investigate its indirect influences on the association between PTSD and AUD, as well as its potential shared molecular genetic risk with PTSD in a genetically-informative study of college students. A sample …

Older adults who experience social isolation have higher rates of mortality relative to their counterparts. Social interactions are an important way to combat this isolation. This research aims to better understand how social isolation in older adults living in low-income households in Richmond, Virginia (RVA) is related to their economic, physical, and psychological health status. As part of the iCubed Health and Wellness Aging Core and in collaboration with the Richmond Memorial: East End Housing Coalition for Older Adults, older adults from a selected public housing unit (n=28) self-reported their financial status, experiences with physical and psycho-social health, and feelings …

Graduate Research Posters
The physiological manifestations of posttraumatic stress disorder (PTSD) have been associated with an increase in risk of cardiovascular disease (CVD) independent of negative lifestyle factors. Peripheral vascular dysfunction may be a mechanism by which PTSD increases CVD risk via increases in oxidative stress, inflammation, and/or sympathetic nervous system activity. PURPOSE: This study sought to examine peripheral vascular function in those with PTSD compared to age-matched controls. METHODS: Eight individuals with PTSD (5 women, 3 men; age 22 ± 2 years), and sixteen healthy controls (CON; 10 women, 6 men, 23 ± 2 years), participated in the study. Leg vascular function …

The Role Of Nrf2 Signalling In Cell Proliferation And Tumorigenesis Of Chromium Transformed Human Bronchial Epithelial Cells, Marco Antonio De Freitas Clementino
Theses and Dissertations--Toxicology and Cancer Biology
Hexavalent Chromium (Cr(VI) induces malignant cell transformation in normal bronchial epithelial (BEAS-2B) cells. Cr(VI)-transformed cells exhibit increased level of antioxidants, are resistant to apoptosis, and are tumorigenic. RNAseq analysis in Cr(VI)-transformed cells showed that expression of transcripts associated with mitochondrial oxidative phosphorylation is reduced, and the expression of transcripts associated with pentose phosphate pathway, glycolysis, and glutaminolysis are increased. Sirtuin-3 (SIRT3) regulates mitochondrial adaptive response to stress, such as metabolic reprogramming and antioxidant defense mechanisms. SIRT3 was upregulated and it positively regulated mitochondrial oxidative phosphorylation in Cr(VI)-transformed cells. Our results suggests that SIRT3 plays an important role in mitophagy deficiency …

The Use Of Mobile Healthcare Clinics To Expand Access To Underserved Populations: A Rapid Review, Alejandro Jimenez
Undergraduate Honors Theses
Access and utilization of health care is essential for the maintenance of overall health and prevention of chronic diseases. Several factors contribute to healthcare access disparities, including race, ethnicity, poverty, and rurality. Mobile healthcare clinics can be used to travel to medically underserved areas and reduce healthcare disparities. This rapid review examined the scope and impact of mobile healthcare units, and their use in reducing healthcare disparities. The exploration was done in 4 stages (1) identification of existing and relevant research studies, (2) selection of studies using prespecified eligibility criteria, (3) extraction of data from collected studies, and (4) summarization …

The last decade has seen a sharp increase in the number of scientific publications describing physiological and pathological functions of extracellular vesicles (EVs), a collective term covering various subtypes of cell-released, membranous structures, called exosomes, microvesicles, microparticles, ectosomes, oncosomes, apoptotic bodies, and many other names. However, specific issues arise when working with these entities, whose size and amount often make them difficult to obtain as relatively pure preparations, and to characterize properly. The International Society for Extracellular Vesicles (ISEV) proposed Minimal Information for Studies of Extracellular Vesicles (“MISEV”) guidelines for the field in 2014. We now update these “MISEV2014” guidelines …

MicroRNAs, non-coding regulators of gene expression, are likely to function as important downstream effectors of many transcription factors including MYB. Optimal levels of MYB are required for transformation/maintenance of BCR-ABL-expressing cells. We investigated whether MYB silencing modulates microRNA expression in Philadelphia-positive (Ph+) leukemia cells and if MYB-regulated microRNAs are important for the "MYB addiction" of these cells. Thirty-five microRNAs were modulated by MYB silencing in lymphoid and erythromyeloid chronic myeloid leukemia-blast crisis BV173 and K562 cells; 15 of these were concordantly modulated in both lines. We focused on the miR-17-92 cluster because of its oncogenic role in tumors and found …

Department of Pharmacology and Experimental Therapeutics Faculty Papers
Decoding health and disease pathways drives healthcare evolution. Historically, therapeutic paradigms have relied on interventions that mitigate symptoms of established diseases. Increasingly, molecular insights into pathophysiology now provide unprecedented opportunities to offer curative solutions or even prevent disease and thereby secure longitudinal wellness. These opportunities extend past individual patients to entire populations and geographies. Moreover, they optimize prospective healthspan across lifespan. Linking discovery science and its translatable innovations beyond reactive disease intervention to proactive prevention will maximize society’s returns creating the greatest benefit for the greatest number of people globally.
Utilizing A Decision Aid In Acute Inpatients To Increase Patient Satisfaction In Communication With The Provider, Elizabeth Fiedler
Utilizing A Decision Aid In Acute Inpatients To Increase Patient Satisfaction In Communication With The Provider, Elizabeth Fiedler
Doctor of Nursing Practice (DNP) Practice Innovation Projects
Introduction. If communication between patients and providers is improved, patients have better outcomes because of adhering to medications and treatments. Evidence Summary. Poor communication can lead to unnecessary readmissions and decreased quality of care. Decision aid tools assist patients in compiling thoughts and questions for providers, lead to a higher understanding of the plan of care, and can help patients to clarify confusing components of the treatment plan. Tools that improve communication between patients and providers lead to an increase in patient satisfaction. The HCAHPS survey is a measurement of patient satisfaction. Gaps in Literature. Social Networks, Identity, Health, And Quality Of Life Among Older Gay And Lesbian Individuals In Rural Environments, Marc Aaron Guest
Social Networks, Identity, Health, And Quality Of Life Among Older Gay And Lesbian Individuals In Rural Environments, Marc Aaron Guest
Theses and Dissertations--Gerontology
The goal of this dissertation was to explore aging lesbian and gay individuals living in rural communities, in terms of their social networks and the relationships between these networks, identity, health, and quality of life. Guiding the study were three overarching questions. Using a multi-method design, the research was grounded within a socio-ecological context and focused on how structural systems create pathways for health and are affected by social position (intersectionality). Participants (n=25) were recruited from Kentucky (n=20), West Virginia (n=3), and Tennessee (n=2). Thirteen participants self-identified as gay and twelve as lesbian. Findings highlight the complexity of the aging …
A Systematic Review Of Medical Students’ And Professionals’ Attitudes And Knowledge Regarding Medical Cannabis As A Proxy For Forecasting Trends In The Medical Community’S Acceptance Of Therapeutic Hallucinogens, Jared Weisman
Pitzer Senior Theses
Background: In recent years, the renewed global interest in both hallucinogens’ and cannabis’ therapeutic properties has resulted in shifting attitudes and legislative policies worldwide. The aim of this systematic review is to explore the existing literature on medical professionals’ and students’ attitudes and knowledge regarding medicinal cannabis (MC) to assess any relevant and significant trends which may forecast analogous trends in the nascent clinical acceptance of hallucinogens.
Methods: Using Google Scholar and PubMed, a literature search was performed to identify studies pertaining to healthcare professionals’ and medical students’ knowledge and attitudes regarding MC. This systematic search yielded 43 studies published …
“Throwing Drops Of Water At A Wildfire”: Patient And Provider Strategies For Navigating Housing Instability And Type 2 Diabetes, Marlene Edelstein
“Throwing Drops Of Water At A Wildfire”: Patient And Provider Strategies For Navigating Housing Instability And Type 2 Diabetes, Marlene Edelstein
Public Health Theses
Type 2 diabetes and housing access are growing independent and intersecting crises in the United States, and unstable housing is associated with poor health outcomes for people with type 2 diabetes. A growing body of evidence points to the challenges of adhering to diabetes management without access to stable housing, and that provider behavior is impacted when their patients face housing instability. In this qualitative study, we investigate the ways that primary care providers and people with type 2 diabetes strategically navigate the challenges posed by homelessness and unstable housing. Semi-structured interviews from two qualitative studies were analyzed. The first …
A Review On Antibiotic Stewardship In Pediatric Primary Care, Paulina Kaminski
A Review On Antibiotic Stewardship In Pediatric Primary Care, Paulina Kaminski
Public Health Theses
Background. Misuse of antibiotics for upper respiratory tract infections (URTIs) in pediatric primary care settings persists despite growing concern over antibiotic resistance. Interventions aimed at improving prescription use, coined Antibiotic Stewardship Programs (ASPs) often target clinicians for improved clinical decision-making. More recently, ASPs have begun educating patients to antibiotic needs, and for pediatric cases, parents who might pressure clinicians into prescribing unneeded antibiotics. While many reviews have targeted adult patients or emergency settings, few evaluate practices for pediatric populations. For this reason, we performed a review of the literature to summarize common strategies used to reduce antibiotic overprescribing for pediatric …
Old & Sexy: Investigating And Reducing Stigmatization Of Sexually-Active Older Persons, Samantha Nicole Levy
Old & Sexy: Investigating And Reducing Stigmatization Of Sexually-Active Older Persons, Samantha Nicole Levy
Public Health Theses
Media often portrays older persons as asexual or perversely sexual. This stigmatization may harm older persons’ sexual, mental, and physical health and well-being. As such, our study aimed to investigate the stigmatization of sexuality in later life and see if it can be reversed. Our four hypotheses included: (1) sexually-active older persons would be more stigmatized than sexually-inactive older persons; (2) negative age beliefs would predict stigmatization of sexually-active older adults; (3) participants would think more negatively about sexually-active older men compared to sexually-active older women; and (4) a brief writing intervention would help improve participants’ views towards sexually-active old …

Comparison Of Traditional And Alternative Curing Ingredients On Curing Reactions In A Model Meat System, Faith D. Rasmussen, Gary A. Sullivan
Nebraska Beef Cattle Reports
To meet consumer trends, alternative curing ingredients are used to replace sodium nitrite and cure accelerators. Due to the complexity of meat, it is challenging to compare traditional and alternative ingredients for curing reactions. Using a model system, sources of nitrite (traditional, sodium nitrite and alternative, cultured celery juice powder), salt, and cure accelerators (traditional, sodium erythorbate, or alternative, cherry juice powder) at ingoing sodium nitrite concentrations of 10, 50, 100, 150, or 200 ppm were evaluated for curing reactions. More complete curing reactions were indicated by a higher concentration of cured meat pigment, and lower sulfhydryl groups. Lower residual …

Nebraska Beef Cattle Reports
A pooled analysis of 21 finishing trials (2002–2016; 1530 animals) with cattle individually fed in Calan gate barns was conducted. Mixed model regression analysis following random coefficient methodology was used to evaluate relationships between performance variables and carcass characteristics. Gain had a greater effect on efficiency (R2 = 0.72) compared to intake (R2 = 0.02). The relationship between gain and efficiency was cubic, while intake had a quadratic relationship. The cubic response of gain relative to efficiency was continually increasing with relatively slight curves in the line heavily influenced by points that lay on the ends of the …
