Background: Surface modification strategies to reduce engineered nanomaterial (ENM) bioactivity have been used successfully in carbon nanotubes. This study examined the toxicity and inflammatory potential for two surface modifications (humic acid and carboxylation) on titanium nanobelts (TNB).

Methods: The in vitro exposure models include C57BL/6 alveolar macrophages (AM) and transformed human THP-1 cells exposed to TNB for 24 hrs in culture. Cell death and NLRP3 inflammasome activation (IL-1β release) were monitored. Short term (4 and 24 hr) in vivo studies in C57BL/6, BALB/c and IL-1R null mice evaluated inflammation and cytokine release, and cytokine release from ex vivo cultured AM. …

This study assessed the effects of Coprinus comatus cap (CCC) on adipogenesis in 3T3-L1 adipocytes and the effects of CCC on the development of diet-induced obesity in rats. Here, we showed that the CCC has an inhibitory effect on the adipocyte differentiation of 3T3-L1 cells, resulting in a significant decrease in lipid accumulation through the downregulation of several adipocyte specific-transcription factors, including CCAAT/enhancer binding protein β, C/EBPδ, and peroxisome proliferator-activated receptor gamma (PPARγ). Moreover, treatment with CCC during adipocyte differentiation induced a significant down-regulation of PPARγ and adipogenic target genes, including adipocyte protein 2, lipoprotein lipase, and adiponectin. Modulation of immune response is important in cancer immunotherapy, vaccine adjuvant development and inflammatory or immune disease therapy. Here we report the development of new immunomodulators via control of shape transition among RNA triangle, square and pentagon. Changing one RNA strand in polygons automatically induced the stretching of the interior angle from 60° to 90° or 108°, resulting in self-assembly of elegant RNA triangles, squares and pentagons. When immunological adjuvants were incorporated, their immunomodulation effect for cytokine TNF-α and IL-6 induction was greatly enhanced in vitro and in animals up to 100-fold, while RNA polygon controls induced unnoticeable effect. The current study examined the predictive value of stressful life experiences and coping strategies on posttraumatic growth and psychopathological symptoms. Using the Traumatic Life Events Questionnaire (TLEQ), the Brief COPE, the Brief Symptom Inventory (BSI), and the Posttraumatic Growth Inventory (PTGI), hierarchical linear regression models were used to explore the contribution of specific coping strategies to predict posttraumatic growth and psychopathological symptoms following exposure to a trauma/stressful life event. We hypothesized that coping styles such as planning, humor, and acceptance would predict more adaptive outcomes, whereas coping styles such as venting, substance use, and denial would predict more maladaptive outcomes. …

IPEC Mini-grant application for funding of UNE student project Homeless Needs Assessment. The Homeless Needs Assessment Project was designed to assess the greatest needs for people experiencing homelessness in Portland, Maine. UNE Students from several healthcare professions partnered together to provide a holistic approach to the community needs with hopes of developing and implementing intervention strategies. Goal was to conduct surveys of at least 100 homeless individuals.

Recognition of the identity of familiar faces in conditions with poor visibility or over large changes in head angle, lighting and partial occlusion is far more accurate than recognition of unfamiliar faces in similar conditions. Here we used a visual search paradigm to test if one class of social cues transmitted by faces—direction of another’s attention as conveyed by gaze direction and head orientation—is perceived more rapidly in personally familiar faces than in unfamiliar faces. Defective mitochondrial distribution in neurons is proposed to cause ATP depletion and calcium-buffering deficiencies that compromise cell function. However, it is unclear whether aberrant mitochondrial motility and distribution alone are sufficient to cause neurological disease. Calcium-binding mitochondrial Rho (Miro) GTPases attach mitochondria to motor proteins for anterograde and retrograde transport in neurons. Using two new KO mouse models, we demonstrate that Miro1 is essential for development of cranial motor nuclei required for respiratory control and maintenance of upper motor neurons required for ambulation. Neuron-specific loss of Miro1 causes depletion of mitochondria from corticospinal tract axons and progressive neurological deficits mirroring …

The ubiquitin-proteasome system is targeted by many viruses that have evolved strategies to redirect host ubiquitination machinery. Members of the genus Chlorovirus are proposed to share an ancestral lineage with a broader group of related viruses, nucleo-cytoplasmic large DNA viruses (NCLDV). Chloroviruses encode an Skp1 homolog and ankyrin repeat (ANK) proteins. Several chlorovirus-encoded ANK repeats contain C-terminal domains characteristic of cellular F-boxes or related NCLDV chordopox PRANC (pox protein repeats of ankyrin at C-terminal) domains. These observations suggested that this unique combination of Skp1 and ANK repeat proteins might form complexes analogous to the cellular Skp1-Cul1-F-box (SCF) ubiquitin ligase complex. …

Background

Varenicline carries a black box warning for neuropsychiatric adverse events.

Objective

We examined varenicline use and past history of major depressive disorder (MDD) on depressive symptoms during smoking cessation.

Method

This is a secondary analysis of two smoking cessation studies in 152 postmenopausal women who received placebo or nicotine patch, or 78 women who received varenicline with relaxation. Lifetime history of MDD (LH-MDD) was assessed at baseline and women with current MDD were excluded. Center for Epidemiologic Study Depression scale (CESD) measured depressive symptoms at baseline, 6 and 12 weeks.

Results

Baseline CESD scores were 5.3 + 4.4. Those …

While the symptomology of underactive bladder (UAB) may imply a primary dysfunction of the detrusor muscle, insights into pathophysiology indicate that both myogenic and neurogenic mechanisms need to be considered. Due to lack of proper animal models, the current understanding of the UAB pathophysiology is limited, and much of what is known about the clinical etiology of the condition has been derived from epidemiological data. We hereby review current state of the art in the understanding of the pathophysiology of and animal models used to study the UAB.

Fatty acid synthase (FASN) is the enzyme that synthesizes fatty acids de novo in human cells. Although FASN is generally expressed at low levels in most normal tissues, its expression is highly upregulated in many cancers. Consistent with this notion, inhibition of FASN activity has demonstrated potential to halt proliferation and induce cell death in vitro and to block tumor growth in vivo. Consequently, FASN is widely recognized as a valuable therapeutic target. Target-specific oral anticoagulants have become increasingly available as alternatives to traditional agents for the management of a number of thromboembolic disorders. To date, the direct Factor Xa inhibitor rivaroxaban is the most widely approved of the new agents. The dosing of rivaroxaban varies and adheres to specific schedules in each of the clinical settings in which it has been investigated. These regimens were devised based on the results of phase II dose-finding studies and/or pharmacokinetic modeling, and were demonstrated to be successful in randomized, phase III studies. In 2012 nearly 1,200 federally funded community health centers were providing access to care for a predominantly low-income population in medically underserved areas across the country. As health insurance coverage expands under the Affordable Care Act (ACA) and the demand for primary care increases, the role of health centers is likely to increase, and the ACA’s large investment in the health center program provides new resources to help meet growing needs.

This brief describes the operational differences among six different types of benefits advisors including: 1) Commercial agents and brokers, 2) Medicaid enrollment brokers, 3) navigators, 4) non-navigator assistance personnel (or in-person assisters), 5) certified applications assisters, and 6) health center outreach and enrollment assistance workers. We will address: 1) the role of each benefits advisor 2) the health plans with which benefits advisors are authorized to work 3) training requirements, 4) compensation 5) conflict of interest requirements, and 6) the impact benefits advisors have on consumer enrollment decisions.

This is the final report for AMAC-2, entitled Australian Medical Assessment Collaboration: from proof of concept to proof of sustainability (OLT project ID12-2482). This project advanced previous work funded by the ALTC and was undertaken from early 2013 to mid 2014. AMAC-2 took the proof of concept achieved through the initial AMAC project with the aim of building an ongoing, sustainable and successful collaboration between medical schools in Australia and New Zealand.

The Australian Medical Assessment Collaboration (AMAC) project, funded by the Office of Learning and Teaching, seeks to provide an infrastructure and a road map to support collaboration between Australian medical schools in matters of assessment. This may not seem very new perhaps, because there are already several collaborations taking place in Australia, and, typically, they relate to joint item banks, (such as the IDEAL consortium), or joint test administration, (such as the International Foundation of Medicine tests). Ethiopia’s population policy specifically aims to reduce TFR from 7.7 to 4.0 and to increase contraceptive use from 4.0% to 44.0% between 1990 and 2015. In 2011, the use of contraceptive methods increased seven-fold from 4.0% to 27%; and the TFR declined by 38% to 4.8. The use of modern contraceptives is, however, much higher in the capital Addis Ababa (56%) and other urban areas but very low in rural areas (23%) far below the national average (27%). Considerable evidence seems to show that emotional and reflex reactions to feared situations are mediated by the amygdala. It might therefore seem plausible to expect that amygdala-coded fear should also influence decisions when animals make choices about instrumental actions. However, there is not good evidence of this. In particular, it appears, though the literature is conflicted, that once learning is complete, the amygdala may often not be involved in instrumental avoidance behaviours. วัตถุประสงค์ เพื่อศึกษาเปรียบเทียบประสิทธิภาพการปิดผนึกคลองรากฟันในฟันกรามล่างที่มีส่วนคอดคลอง รากฟันของเทคนิคการอุดคลองรากฟัน 3 เทคนิคโดยใช้แบบจําลองการรั่วซึมของกลูโคส วัสดุและวิธีการ นําฟันกรามล่างมนุษย์จํานวน 70 ที่ได้รับการระบุและจําแนกประเภทของส่วนคอดคลอง รากฟันด้วยเครื่องไมโครที่มาขยายคลองรากฟันด้านใกล้กลางโดยใช้ไฟล์นิกเกิลไทเทเนียมชนิดหมุนด้วยเครื่อง แบ่งฟันโดยการสุ่มออกเป็น 3 กลุ่มทดลอง (กลุ่มละ 20 ) กลุ่มควบคุมบวกและกลุ่มควบคุมลบ (กลุ่มละ 5 ปี) อุดคลองรากฟันในกลุ่มทดลองโดยใช้เทคนิคแลทเทอรัล คอนเดนเซชั่น แมทซ์เทปเปอร์ซิงเกิลโคน และคอนตินิวอาส เวฟคอมแพคชันด้วยกัตทาเพอร์ชาและเอเอสพลัสซีลเลอร์ หลังการอุดคลองรากฟันตัดรากฟันด้านไกลกลางออก ประเมินการรั่วซึมโดยใช้แบบจําลองการรั่วซึมผ่านของกลูโคสในระยะเวลา 28 วัน ผลการศึกษา ไม่พบการรั่วซึมของกลูโคสในทั้ง 3 กลุ่มทดลอง จากผลดังกล่าวจึงไม่มีการวิเคราะห์ข้อมูลทางสถิติ สรุป เทคนิคการอุดคลองรากฟันที่แตกต่างกันไม่มีผลต่อประสิทธิภาพการปิดผนึกของวัสดุอุดคลองรากฟันในฟันกรามล่างที่มีรูปร่างไม่สม่ําเสมอและมีส่วนคอดคลองรากฟัน การเลือกใช้เทคนิคที่ง่ายกว่าร่วมกับซีลเลอร์ที่มีคุณสมบัติที่เหมาะสมอาจถูกนํามาใช้เป็นทางเลือกในการขุดคลองรากฟันที่มีส่วนคอดคลองรากฟันได้ (ว ทันต จุฬาฯ 2557;37:267-78)
