In this study, we aimed to determine the levels of some biochemical parameters in the seminal plasma of German Shepherd dogs. Seven German Shepherd dogs were used as materials. A total of 35 ejaculates, five from each dog, were collected. The seminal plasma of each ejaculate was separated by centrifugation at 5000 rpm for 10 min. Biochemical analyses of the seminal plasma were performed. In conclusion, the average concentrations of total protein, calcium, phosphorus, magnesium, sodium and potassium were 3.75 ± 0.21 g/dl, 3.22 ± 0.32 mg/dl, 2.49 ± 0.25 mg/dl, 4.50 ± 0.37 mg/dl, 137.25 ± 1.98 mEq/l and …

In this study, the effect of vitamin E usage (75 days, 45 mg/day of vitamin E (DL-\alpha tocopheryl acetate)) on the myofibrillar proteins in male Morkaraman lamb meat was investigated. SDS-PAGE gels indicated that vitamin E supplementation to the diet did not have any effect on actin and myosin; however it had a fractional effect on tropomyosin. For pH values, no differences were observed between the control and vitamin E supplemented groups.

Diabetes mellitus was induced in Wistar-Albino type male rats with a single dose of streptozotocin (60 mg/kg intraperitoneally). The kinetic properties of pyruvate kinase were investigated in normal and diabetic rat liver tissue. In normal rat liver tissue, the incubation period of pyruvate kinase was 15 min and the optimum pH was 8. In diabetic rat liver tissue, the incubation period of pyruvate kinase was 10 min and the optimum pH was 7. The aim of this study was to develop ELISA to examine antibodies against rinderpest virus (RPV) for vaccination programmes in the Malatya and Elazığ regions. Serum samples from 180 clinically healthy cattle in Malatya were obtained in the spring of 1995 and 157 serum samples from cattle were taken in the spring of 1996 in Elazığ. In this study, we aimed to investigate the regional profile of bovine and ovine Brucellosis in Burdur by isolating Brucella spp. in milk and by detecting antibodies to Brucellae in milk samples. For this purpose, 404 milk samples from 101 cows and 226 milk samples from 113 ewes located in 14 different areas of Burdur were collected. The samples were examined by bacteriological and serological methods. Brucella spp. were not isolated from the milk samples. Macroanatomical Studies On The Cranial Cervical Ganglion In Sheep, İsmai̇l Türkmenoğlu, Nejdet Dursun

In this study, the location and relations of the cranial cervical ganglion were investigated. A total of six sheep of different ages and both sexes were used. The cranial cervical ganlion lies medial to the occipitohyoid muscle, ventral to the jugular foramen, lateral to the longissimus capitis muscle and craniomedial to a common mass constituted by the glossopharyngeal, vagal, accessory and hypoglossal nerves and the condyloid artery. The colour and shape of the ganglion are light brown and fusiform respectively.

This study was performed to compare the growth characteristics of Kangal and Akbaş Turkish shepherd dogs. Live weight records of 141 puppies at birth, 86 puppies at weaning, 68 puppies at 6 and 12 months of age and four different body measurements of 68 dogs at different ages were used for determining growth characteristics. The least squares method was used for statistical analysis. The survival of Yersinia enterocolitica strain 0:9 was investigated during the manufacturing and ripening of Mihalıç cheese produced from raw milk. Mihalıç cheese was produced after inoculating 10^5 cfu/ml of Y. enterocolitica strain 0:9 into raw milk. All cheese samples were ripened at + 4 °C for 30 days in 18 to 22% of brine. The objectives of this study were to compare some haemodynamic and clinical parameters and the analgesic, sedative effects of lidocaine and a lidocaine-meperidine combination for caudal epidural analgesia in mares. This study was performed on 12 thoroughbred mares aged 13-24 years. Time to onset of analgesia in the combination group (6.67 ± 0.42 min) was significantly (P < 0.01) lower than in the lidocaine group (12.17 ± 1.05 min). However, duration of analgesia was significantly (P < 0.001) longer for the combination group (162.67 ± 5.67 min) than for the lidocaine group (109.33 ± 3.29 min). Heart and respiratory rates, rectal temperature, pH, pO_2, pCO_2, HCO_3, BE, PCV, THb and O_2Hb values measured before and after administrations in both groups were not significantly different. In conclusion, the combination of lidocaine-meperidine produced analgesia of quicker onset and longer duration than did lidocaine administered alone for caudal epidural analgesia in mares.

Investigations On The Diagnosis Of Pseudotuberculosis Infections In Sheep By Allergic Skin Tests, Yahya Kuyucuoğlu

This investigation was carried out to evaluate the use of allergic skin tests in the diagnosis of pseudotuberculosis in sheep. For this purpose, two different allergens were prepared by two different methods. Allergens were prepared from C. pseudotuberculosis NVH 3368 strain. The study was carried out on farms with a total of 300 sheep in the province of Konya. The effects of two different allergens injected into four different sites in all three groups (infected, suspected and healthy) were examined (P < 0.05). At the end of the allergic skin test it was found that 73 (73%) of infected sheep tested positive. Sixty-one (83.5%) of these 73 were tested by SAT, 60 (82.1%) by postmortem examination and 38 (52.0%) by bacteriological examination (P < 0.05). All tests gave negative results for the healthy group. Moreover, in the infected animals, the sensitivity, specificity and predictive values from SAT, postmortem examination and bacteriological examination were investigated. The values were 100.0%, 29.0% and 75.5% for the axillary site. It was confirmed that the axilla is the best injection site in allergic tests. In conclusion, it was found the allergic skin test was a reliable method for the diagnosis of pseudotuberculosis in sheep in the field.

D1, D2 and D3 diets containing 15.5, 12.5 and 9.0 MJ kg^{-1} digestible energy concentrations were offered to rainbow trout (Oncorhynchus mykiss Walbaum, 1792), and the rates of gastric evacuation and feed intake were determined. Gastric evacuation determinations were achieved by slaughtering eight fish every 6 h until no residue was found in the cardiac stomach. Return of appetite experiments were conducted by re-feeding groups of trout every 6 h following the first feeding. The effects of different daily feeding rates on the specific growth rate and food conversion ratio of Siberian sturgeon (Acipenser baeri) were investigated in order to determine the optimum daily feeding rate for fish with an initial mean weight of 1736 ± 37 g. The relationship of age, growth and length-weight was analyzed for Chalcalburnus mossulensis from the Aşkale region of Karasu River from November 1995 to October 1997. Fish in the sample groups were between 1 and 7 years and the ratio of males to females was 1:1.08. Von Bertalanffy equations were used to find the growth in length for both sexes. This study was performed to examine whether adult fibroblast cells can be manipulated genetically and whether clonal lines derived from those cells can support embryonic development. A primary adult cell line was established from the ear of an aged cow. Adult cells with a plasmid containing the enhancer green fluorescence protein (EGFP) gene, and non-transfected cells were used for cloning. Green fluorescence expression was observed in 35/49 (71.4%) adult clones. The developmental rates of embryos were significantly lower for cell lines expressing EGFP (11.4%) than for non-transfected cells (20.1%, P < 0.05). However, there was no decrease in nuclear transfer (NT) developmental rates (21.5%) when donor nuclei from EGFP transfected cell lines not expressing EGFP but retaining neomycin-resistance gene expression were used as donor nuclei. The NT embryos from transfected and non-transfected cell lines had similar morphology and cell numbers. The results indicated that adult cells can complete clonal propagation including transfection and selection and can be used to produce transgenic NT embryos.

Changes in fatty acid composition were investigated during the early development stages (egg, embryo, alevin and fry) of rainbow trout (Oncorhyncus mykiss). It was found that saturated and monounsaturated fatty acids, i.e., 14:0, 16:1n-7 and 18:1n-9, were utilized to a greater extent as energy substrates. Quantities of 18:3n-6, 18:4n-3, 20:2n-6 and 22:5n-3 fatty acids were significantly reduced (P < 0.05) during early development. The most significant depletion was observed in monounsaturated fatty acids, which fell from 33.89% to 23.41% during development while n-6 polyunsaturated fatty acids fell from 16% to 12.9%.

In this study, findings of the qualities and quantities of commercial and trash catches from beach-seining in coastal waters of the Aegean Sea are presented. A total of 62 finfish and eight invertebrate taxa were identified in catches sampled during May, June, July and September 1996, with 42 taxa being retained and 28 taxa being genuine trash catches. The estimated total ratio of commercial to genuine trash catch (kg) was 1:0.273. The aim of this study was to estimate the carrying capacity of Kesikköprü Dam Lake, Ankara, where cage farms for the intensive culture of rainbow trout are located. For this purpose Dillon and Rigler's phosphorus budget model was applied in a series of steps and the carrying capacity of the lake was found to be 3335 tonnes per year. This estimated value was about 10 times higher than the present production level of the lake. It seems possible to orientate the fish culture in cages in inland waters by using the phosphorus budget model recommended in this study.

Non-steroidal anti-inflammatory drugs (NSAIDs) are in common use worldwide. These drugs may sometimes be used in high or toxic doses by mistake. The aim of this research was to determine the effect of a cage farm with a capacity of about 55 t on the benthic macrofauna in Kesikköprü Dam Lake. At 3 stations (the cage and 15 m and 60 m from the cage) selected for this purpose the abundance and composition of benthic macrofauna were determined for 11 months. The abundance of benthic macrofauna (individuals/m^2) between the stations was statistically significant (P < 0.05). The abundance of benthic macrofauna was higher at the cage station than at the other 2 stations, except in April and November. During the research period the lowest number of individuals (937 individuals/m^2) was found at the second station 15 m from the cage in February; and the highest number (14,091 individuals/m^2) was determined at the cage station in December. Members of the class Pelecypoda were found at all 3 stations. Dreissena, which is a member of the class Pelecypoda, was the most dominant benthic organism (54%), followed by Chrinomus (30%) and Tubifex (11%). At the cage station, while members of the class Pelecypoda were found, no members of the classes Insecta, Oligocheta or Crustacea were found. The highest number of benthic macrofauna genera (7) was found at the station 60 m from the cage in April and November. The lowest number of genera (1) was found at the cage station in July. The results of this study in Kesikköprü Dam Lake reveal that the cage culture has a localizing effect on the abundance and composition of benthic macrofauna.

This study is concerned with the feeding behavior of pikeperch (Sander lucioperca (L., 1758)) living in Hirfanlı Dam Lake constructed on the Kızılırmak River. There were 326 pikeperch caught between August 1996 and July 1997. The fork length and weight of the pikeperch caught ranged between 105 and 529 mm and 12 and 1520 g in the general population. Pikeperch are carnivorous and piscivorous and they eat their own species when nutrients are lacking. The smallest fish-swallowing length was determined to be 145 mm. Ten motile Aeromonads, including one Aeromonas hydrophila ATCC 7512, were used. Of the strains, five were isolated from gulls (one A. hydrophila strain and four A. caviae strains) and four from rainbow trout (three A. hydrophila strains and one A. caviae strain). Whole cell protein profiles of these strains were analyzed by SDS-PAGE. Forty-five pairs were formed in comparing the protein profiles of each strain. Glucose 6-phosphate dehydrogenase (G6PD) was purified from goose erythrocytes and some characteristics of the enzyme were investigated. The purification procedure was composed of 3 steps: hemolysate preparation, ammonium sulfate precipitation, and 2', 5'-ADP Sepharose 4B affinity gel chromatography. Thanks to the 3 consecutive procedures, the enzyme, having a specific activity of 36.2 EU/mg protein, was purified for a yield of 68.79% and 3892 folds; to ascertain enzyme purity, SDS-PAGE was performed.
