Kinetics And Inhibition By Some Amino Acids Of Arginase In Cattle Rumen Tissue, Mi̇ne Eri̇şi̇r, Sema Temi̇zer Ozan

Turkish Journal of Veterinary & Animal Sciences

The effects of some amino acids on arginase activity in cattle rumen tissue were studied. L-amino acids, such as valine, leucine, isoleucine, ornithine, cysteine, proline, lysine inhibited the enzyme activity noncompetitively. On the other hand, histidine and hydroxyproline were not determined to have any inhibitory effect on the enzyme activity. Isoleucine, valine, leucine, cysteine, ornithine were more strongly inhibitory than proline and lysine.

This study was conducted to investigate the possibilities of developing terminal male and female lines for lamb production by the crossing of native breeds, that is, Akkaraman, Sakız (Chios) and Kıvırcık breeds. The Akkaraman sheep is a fattailed breed raised in central Anatolia, while Sakız and Kıvırcık sheep aren non fattailed breeds raised in western and northern-western Anatolia. The Birth rate of Akkaraman x Akkaraman (AA), Sakız x Akkaraman (SA) and Kıvırcık x Akkaraman (KA) breeding groups were generally found as follows: 94.0, 78.6 and 74.0% in 1996; and 93.3, 90.8 and 85.0% in 1997, respectively. Forty-six formalin-fixed paraffin-embedded canine mammary tumors, 15 benign and 31 malignant, were studied immunohistochemically. Mouse monoclonal anti-alpha-cytokeratin 18, mouse monoclonal anti-alpha-cytokeratin (pan cytokeratin), mouse monoclonal anti-alpha-smooth muscle actin and mouse monoclonal anti-vimentin antibodies were used. The ductal and acinar epithelium stained positively for cytokeratin 18 and pan cytokeratin, although there was no immunoreactivity in some tumors. Metaplastic squamous cells in two solid carcinomas were positive for both cytokeratin 18 and pan cytokeratin. The aim of this study was to evaluate heavy metal levels in mussels ( Unio stevenianus Krynicki) obtained from Van Lake. A total of 120 samples representing each of the four seasons was collected between 01.01.1994 and 01.01.1995. The organic parts of the samples were ashed with a mixture of magnesium oxide and magnesium nitrate according to the dry ashing method and arsenic residues were measured by silver diethyldithyocarbamate in UV spectrophotometer; copper, cadmium, zinc and lead levels were also determined using an atomic absorption spectrophotometer. Frozen-thawed fish and fillets are being marketed as fresh fish fillets. One of the methods which helps us to differentiate unfrozen from frozen-thawed fish fillet is the enzymatic method. By means of the freezing and thawing process lysosomal enzymes are released. Lysosomal enzyme activities have the enzymes which decompose protein, carbohydrate, fat and nucleic acids. This study was carried out to investigate the prevalance of leptospirosis in the urine of cattle slaughtered in three major abattoirs in the east of Turkey. A polymerase chain reaction (PCR) based on a pair of genus-specific primers was used to detect leptospiral DNA in the urine samples of 473 cattle, 284 of which were from Elazig, 112 from Malatya and 77 from Diyarbakir. The detection limit of the method was determined to be approximately five bacteria per ml of urine. A five-year-old male Great Dane was presented with nonambulatory tetraparesis and during the drip follow-up period its neurologic status became worse, despite conservative management for 10 days. Ligamentum flavum disease, dorsal bony compression and malformation of the articular process were diagnosed by magnetic resonance imaging (MRI). A dorsal laminectomy was performed. Physiotherapy, including massage, flexion and extension of the limbs, was done twice a day. Voluntary standing and walking were observed during the fourth week after the operation. Pharmacokinetics of oxytetracycline were investigated in 20 sheep following intravenous injection of 40, 80 and 160 mg/kg doses. An increase in concentration of oxytetracycline was found with increasing dose at all the sampling times in each animal. The peak plasma concentration of oxytetracycline attained after intravenous administration of 40, 80 and 160 mg/kg of the drug was 18.67±0.09, 40.63±0.16 and 86.51±0.10 mg/ml, respectively. These plasma concentrations decreased with plasma half lives of 10.88 ± 0.29, 5.87 ± 0.08 and 7.35 ± 0.08 hours respectively. The purpose of the present study was to determine the effects of acute heat stress on the proportions of leucocyte components and hematocrit values in broiler chickens. Exposure of broiler chickens to 39±1 °C for 2 h at 44 d of age caused an increase in rectal temperatures, and in heterophil and basophil ratios. The H/L ratio also increased from 0.25 to 0.43. The exposure of the broilers to acute heat stress resulted in decreased monocyte and lymphocyte proportions whereas the proportion of eosinophil and the hematocrit values were not affected. Hypothyroidism is commonly associated with hyperlipidemia. The aim of this study was to investigate the effect of Propylthiouracil-induced hypothyroidism on the plasma lipid profile in rabbits. This investigation was performed on male, white New Zealand ( Oryctolagus cuniculus huxley) rabbits. Twenty rabbits were used; each of the two groups (hypothyroidism and control) consisted of ten animals. Serosal application of BAC (benzalkonium chloride) to the pyloric antrum of the rat led to a complete loss of the intrinsic innervation of the treated region and also extensive loss of extrinsic sensory projections in the treated and untreated regions of the stomach. Development Of An Enzyme Immunoassay For The Determination Of Ovine Growth Hormone In Plasma, Behi̇ç Serpek, Seyfullah Hali̇loğlu

A competitive enzyme immunoassay for the determination of ovine growth hormone (oGH) was developed. The assay is based on (anti-rabbit IgG) goat IgG-coated microtitreplates, anti-oGH from rabbits, biotinyl-oGH and streptavidin-horseradish peroxidase. The assay is done directly with 40 ml of plasma and the calibration graph (90% relative binding at 0.38 ± 0.02 ng ml -1 and 50% relative binding at 3.80 ± 0.18 ng ml -1 ) is prepared in plasma without measurable endogenous oGH. Recovery in different plasma samples with added oGH amounted to 96.78 - 100.03 %, and all variabilities were < 10.44. The assay shows no significant binding of oFSH, oLH, oTSH and oPRL. The assay detects high and low plasma oGH levels within the physiological variation as well as changes in plasma oGH after stimulation with growth hormone releasing factor. Hence, it offers a reliable alternative radioimmunoassay. However, a reference method providing evidence of the identity of exogenous oGH will be required for proving illegal treatment with oGH and for forensic purposes.

The present study was conducted to determine whether the mushroom Pleurotus florida has potential to increase the digestibility and feeding value of wheat straw in with urea treatment. In the trial the effects of inoculation of P. florida, and 2% urea treatment were examined in a 2 by 2 factorial design. The mushroom was inoculated to urea supplemented or unsupplemented wheat straw by solid-state fermentation. One hundred pneumonic lungs from calves were examined by an immunoper oxidase technique using avidin-biotin- per oxidase complex method in formalin-fixed, paraffin-embedded sections for Haemophilus somnus. In the histological examination of tissue sections, strong positive reactions against H. somnus were detected in 14% of the lungs. H. somnus antigens were observed at the surface and/or within the epithelial cells, macrophages, necrotic alveolar walls, alveolar -bronchial/br onchiolar exudates, fibrins, distended interlobular septa and perivascular areas. In addition, dense antigen accumulations were detected in the necrotic macrophages and neutrophils.

The immunogenicity of Pasteurella haemolyticaserotypes was investigated in an experimental animal model. Rabbits were immunized with bacterins and leukotoxins of selected serotypes of P. haemolytica,and the IgG level and antileukotoxin activities of immune sera were measured. The highest IgG levels against A1, A2, A7 and T4 antigens were detected in rabbits receiving polyvalent combined (bacterin plus leukotoxin) immunogens. Monovalent combined immunogens induced high antibody titers against homologous serotypes, but low titers against heterologous serotypes. The aim of this study was to determine the quality changes in shrimp stored at 4°C. In the study, whole shrimp samples were placed into straphore plates and stored in a refrigerator (+4°C). Each plate contained 250g samples. Every day the samples were analyzed for sensory properties, Total Volatile Basic Nitrogen (TVB-N) content, Trimethylamine Nitrogene (TMA-N) content, pH value and moisture content. According to findings of sensory, physical and chemical analyses the samples were found to be of "good quality" at day 0, "good quality," at day 1 and "spoiled" after 2 days.

The objective of this study is to compare meat yield and chemical composition of blue crabs and sand carbs. The ratio of crude protein (18.93%), dry matter (22.43%) and crude ash (2.34%) in the meat taken from male individuals were found to be higher in sand crabs than in blue crabs, but lipid levels were not statistically different for male individuals of the two species (p>0.05). This study was carried out to investigate the fattening performance, slaughter and carcass characteristics of White Karaman and Chios x White Karaman (F_1 ) crossbred lambs. The data for the study were obtained from 11 purebred and 12 (6 from each genotype) ram lambs for determination of slaughter and carcass characteristics. This study was conducted in pregnant and aborted sheep to determine Toxoplasma gondiiantibodies. Blood samples were collected from a total of 154 sheep from 22 different herds during the period December 1996-January 1997 in Elazig and vicinity. Of these 154 sheep, 56 were pregnant, 57 aborted in the previous year and the remaining 41 sheep aborted within the period of the study. Sera obtained properly from these blood samples were examined by Sabine-Feldman (SF) test. In this study, the qualitative and quantitative aspects of aerobic bacterial flora of the skin, gills, intestine, liver, kidney and spleen of rainbow trout in relation to pond water in two different fish farms were analysed. The bacterial load of pondwater in Seniçbey Farm was 10 2 -10 5 cfu/ml. The objective of this study was to determine blood plasma and hair copper levels in clinicially healthy Swiss Brown and Holstein cows. This study was performed on 50 cows, 3-6 years old. Blood and hair samples were collected in December, March, June and September. Mean plasma copper levels in December, March, June and September were 68.23±2.62, 44.58±1.78, 52.79±2.15 and 52.76±2.59 mg/dl respectively. Also mean hair copper levels were 5.78±0.22, 5.45±0.15, 5.25±0.11 and 5.77±0.15 ppm respectively. Enterotoxaemia Incidence In Small Ruminants In Elazığ And The Surrounding Provinces In 1994-1998, Canan Özcan, Meti̇n Gürçay

132 intestine specimens belonging to small ruminants with suspected enterotoxaemia were sent to the Elazig Veterinary Control and Research Institute from Elazığ and the surrounding provinces in 1994-1998. In bacteriological examinations, it was found that 51 of the 132 specimens were positive and 81 negative. The toxin-neutralisation test was applied for diagnosis. The 51 enterotoxaemia cases were identified as 28 C.welchii Type D, 16 C.welchii Type C, 7 C.welchii Type A.
