Malignant brain tumors are generally lethal soon after their diagnosis, and the five-year survival rate for the patient is low in spite of neurosurgery, radiation therapy, and/or chemotherapy. Gene therapy has the potential to improve patient survival and quality of life by delivering specific combinations of therapeutic genes directly to the tumor for localized treatment.

Glucocorticoids (GCs) inhibit bone formation in vivo and inhibit osteoblast proliferation and collagen synthesis in vitro. These effects may be mediated by alterations in the insulin-like growth factor (IGF) system. In the present study of normal human osteoblast-like (HOB) cells, we tested the hypothesis that dexamethasone (Dex) inhibits the expression of IGF binding protein-5 (IGFBP-5). Dex decreased IGFBP-5 mRNA levels to 54% of control after 4 hr. Dex did not modify the decay of IGFBP-5 mRNA in transcriptionally arrested osteoblast cells. Dex decreased IGFBP-5 hnRNA levels to 67% of control after 2 hr, and the activity of the human IGFBP- …

Angiogenesis is important in health and disease. In particular, exercise training is known to increase skeletal muscle capillarity, providing there is sufficient training intensity. The stimulus for this may be intracellular hypoxia activating angiogenic growth factor gene expression. Acute hypoxia alone has been shown to increase the gene expression of several key angiogenic regulators, e.g. vascular endothelial growth factor (VEGF) and to a lesser degree transforming growth factor-β1 (TGF-β1), basic fibroblast growth factor (bFGF), and thrombospondin-1 (TSP-1), an endogenous negative angiogenic regulator. A possible cause for root canal failure is the persistence of bacteria that have colonized dentinal tubules. To reduce this risk and, if possible, to also shorten the time consuming endodontic therapy, new equipment and materials are constantly being introduced. The aim of the present investigation was to study the effect of disinfection of dentinal tubules by intracanal laser irradiation using an in vitro model. The groups irradiated by various lasers were compared with calcium hydroxide, a material frequently used as intracanal medication between appointments. Osteoporosis is a disease characterized by low bone density and deterioration of mineralized bone leading to enhanced susceptibility to fracture. Several growth factors have been implicated in the coupling of formation to resorption during the bone remodeling process. The net bone forming activity of many growth factors has prompted numerous studies focused on the regulation of osteoblast cell proliferation, differentiation and activity. The center of the brain-death and organ transplantation debate in Japan has always been whether whole brain-death should be regarded as the death of a person. However, the debate has not yet been settled in Japanese society. There are four main reasons why it is said that whole brain-death criteria are incompatible with Japanese society. First, they cannot establish a person's death because the Japanese define it not according to medical facts but according to social relationships. The cluster determinant 4 (CD4) molecule is a transmembrane glycoprotein. CD4 is essential for normal T helper cell function and plays an important role in T cell development and activation. CD4 is encoded by a single gene located on chromosome 6 in the mouse and chromosome 12 in the human. Both human and murine CD4 genes are divided into ten exons spanning more than 25 kb and have a large non-coding region in the first and third intron. CD4 gene expression is controlled primarily at the transcriptional level during T cell development and activation.

A primary role of the IGF-II/M6P receptor is to target lysosomal enzymes from the golgi to the lysosomes. This receptor has distinct binding sites for IGF-II and M6P, however, reciprocal interactions between these ligands have been observed (Kiess et al. 1989, 1990). Since IGF-II modulates the routing of cathepsin D in MCF-7 cells by blocking the intracellular binding of cathepsin D to the IGF-II/M6P receptor (De León et al. 1996), we hypothesized that expressing a mutant form of IGF-II that does not bind the IGF-II/M6P receptor will not interfere with lysosomal enzyme trafficking.

The cardiac correlate of attention is a deceleration in heart rate, controlled by the parasympathetic division of the autonomic nervous system. This has been extensively studied in infants; the findings indicate that this deceleration is affected by a number of factors, several mediated by the vagus nerve. However, the effects of denervation on this response are not known. Studies with adult heart transplant recipients have shown attenuated acceleration in response to stressful mental tasks. Parkinson's disease (P.D.) has long been thought of as a single disorder. Recent research involving neurochemistry, neurophysiology, and detailed analysis of symptoms, have created a basis for questioning the concept that P.D. is a singular disorder. Literature suggests there may be two primary subtypes (with the possibility of others). The two probable subtypes are hyperkinetic, in which tremor is the predominate symptom and akinetic, in which, gait freezing and postural instability predominate.

The objective of this study was to examine the role of gap junctions in the radiation resistance of thyroid follicles. The question of whether gap junctional intercellular communication plays an important role in the “contact effect”, wherein cells in direct apposition are more resistant to the damaging effects of radiation, has been debated for years. We used the Fischer rat thyroid cell line (FRTL-5) to carry out our experiments. This thesis is divided into two parts: 1) characterization of gap junctional properties in FRTL-5 cells and, 2) exposure of these cells to radiation.

Replacement resorption (RR) occurs following severe injury to the periodontal ligament (PDL). It has been suggested that inflammatory reactions precede RR. The purpose of this study was to examine and compare histologically the effect of topical application of two anti-inflammatory medications and stannous fluoride (SF) on the development of RR. Thirty-four roots of beagle dog mandibular 2nd, 3rd, and 4th premolars received root canal therapy. They were then extracted and left to bench dry for one hour. Psychotherapist factors as predictors of client memory recovery of childhood sexual abuse following a period of amnesia were investigated. Marriage and family therapists and clinical psychologists in California were surveyed regarding the degree of research emphasis in academic training, knowledge of memory processes and professional standards of care regarding memory recovery in psychotherapy, degree of ambiguity tolerance, use of memory recovery techniques and client memory recovery. Therapists with greater research emphasis in academic training reported having fewer clients with memory recovery. Also, therapists who were intolerant of ambiguity used memory recovery techniques more than did therapists who were tolerant. Finally, …

The treatment of pediatric dental patients can only be as effective as the extent to which Spanish-speaking parents consent to the dental treatment. Many cancer patients experience adverse chemotherapy-related side effects. The present study examined whether the perception that one can control and effectively cope with her treatment for breast cancer mitigate against any nausea and vomiting that she may experience in anticipation of chemotherapy treatments. Seventy-five women with breast cancer completed the Multidimensional Health Locus of Control Scale - Form C, the Krantz Health Opinion Survey, the Ways of Coping Scale - Revised, and the Morrow Assessment of Nausea and Emesis. OBJECTIVE: The purpose of this study was to compare elderly non-fallers and fallers for differences in the following: (1) mean functional reach (FR), (2) mean anterior limits of stability (LOS), (3) mean posterior LOS, and (4) mean sensory organization test (SOT) composite score. The following correlations were tested for significance separately in the sample of non-fallers and the sample of fallers: (1) FR and anterior LOS, (2) FR and posterior LOS, (3) FR and SOT composite score, (4) anterior LOS and the SOT composite score, and (5) posterior LOS and the SOT composite score.

Epithelial fatty acid-binding protein (E-FABP) is expressed in the dorsal root ganglia following sciatic nerve injury and in migrating and differentiating neurons during neuronal development. It was hypothesized that E-FABP expression is required for the robust outgrowth of axons from developing and regenerating neurons. To test this hypothesis, E-FABP expression in both PC12 cells and primary retinal neurons was examined. In PC12 cells, NGF induces E-FABP mRNA and protein during the period of neurite outgrowth, and E-FABP localizes to the perinuclear cytoplasm, nucleus, and growth cone. This study mainly examined neuromuscular function, occlusal index, and centric relation-centric occlusion (CR-CO) discrepancy of 23 orthodontically treated patients. Phase I, craniofacial anomalous, and orthognathnic surgery cases were excluded from the study. Record taking was done in 2 sessions: (1) at the time of deband and (2) 3-4 months later. Records taken at each session included facial and intraoral photographs, impressions, CR wax bites, a facebow, and surface EMG (sEMG) recordings.

Phytochemicals are naturally-occurring compounds found in plants, many of which are consumed as nutritional supplements by humans. Pycnogenol is a mixture of flavonoid compounds extracted from bark of pine trees (Pinus maritima) and is commercially available in several different pharmaceutical preparations as a free radical scavenger.
Since few scientific studies have documented the pharmacologic effects of pycnogenol, our objective was to characterize the biological activity of pycnogenol. Plaque accumulation, although undesirable, is prevalent in orthodontic patients with poor oral hygiene and is linked with enamel decalcification. A proposed product designed to seal the tooth around the orthodontic bracket was tested for its durability. It also was evaluated for its ability to seal the enamel.

This study was conducted to examine, retrospectively, orthodontic and orthopedic changes resulting from quad helix expansion therapy in mixed and permanent dentition stages. Twenty-one subjects with an average age of 10.2 years who experienced an expansion of at least 3 mm with the quad helix appliance were selected for this study. The subjects were evaluated according to facial type and age for both short and long-term stability. This study is [sic] introduces a new extension of Euclidean Distance Matrix Analysis (EDMA) as applied to growth prediction analysis. Using EDMA eliminates the presupposition of a set growth pattern, which is introduced by traditional superimposition techniques. When EDMA is extended to analyze prediction methodologies, using a non-age-matched growth sample, some shortcomings become evident. These involve bootstrapping techniques, relative difference in growth, and absence of clinical, real world measure. Since the introduction of bonded orthodontic brackets in the early 1960's spontaneous debonding has been a constant nemesis. Most attempts to avoid this problem by orthodontic manufactures have focused on "bond strength" issue.
