Chemiosmotic Properties Of Isolated Secretory Granules From Parotid Glands Of Normal And Isoproterenol Treated Rats, Peter Raymond Arvan

Secretory granule fractions were prepared isoosmotically from the parotid glands of normal rats and from rats that had received repeated injections of isoproterenol. These granules, purified extensively, are stable after isolation, making it possible to study biophysical parameters which may relate to their role in intracellular transport and secretion.Normal parotid granules have an internal pH of (TURN)6.8 and a low ionic permeability with respect to internal buffering capacity. By contrast, granules from the treated rats have an internal pH ranging from (TURN)7.7 in (0.3 M) sucrose media to (TURN)7.1 in (100 mM) KCl media; suggesting a large ionic permeability with …

A reliable technique for obtaining late prophase, prometaphase and greater numbers of metaphase chromosomes is presented. The increased number of mitoses and high resolution chromosomes were obtained using a modification of the typical three-day blood culture technique in which the cell cycle was temporarily blocked at the G1-s border with methotrexate. A high resolution G-band idiogram is presented using a system similar to the standardization of human chromosomes. The use of this technique and system of nomenclature will facilitate routine analyses of horse chromosomes and provide a basis for the identification and recording of chromosome aberrations in this …

A predicted association between family relations and cognitive development and the emergence of adolescent egocentrism was explored in this study. A sample of seventh grade boys (n=131) and girls (n=120) completed Elkind and Bowen's Imaginary Audience Scale (a measure of egocentrism) and selected items from Heilbrun's Parent-Child Interaction Rating Scale and Schaefer's Parent-Behavior Inventory. A modified version of Lawson's Classroom Test of Formal Operations was used to measure cognitive development. Contrary to a theoretical based hypothesis a negative rather than positive relationship was found between level of formal operational thought egocentrism. Protein recovery and coagulation properties of five commercial and fractionated milk clotting enzymes were studied. The fractionated enzymes were Sephadex G-100 fractions of the commercial enzymes. Milk clotting activity of each fraction was tested using Berridge substrate. All fractions from each preparation which had milk clotting activity as measured with the Formagraph were collected and pooled. These samples and the original enzyme preparations were used to coagulate milk. Percent of protein lost in whey was determined by Kjeldahl. Coagulation was followed using a spectrophotometer monitoring changes in apparent absorbance at 600 nm. Curd protein yields using the five original enzyme …

A modification of a linear diffusion test for measuring milk clotting enzymes at concentrations of 1 x 10-4 to 1 x 10-1 chymosin units/ ml was developed to permit quantitative assay of porcine pepsin in Cheddar cheese with a standard deviation of 6%. The amount of porcine pepsin retained in Cheddar cheese curd was dependent on pH of milk at setting. Milk at pH 6.6, 6.4, 6.2, and 6.0 was set with porcine pepsin, acidified with lactic acid and glucono-~-lactone and made into Cheddar cheese. A rugged and sensitive instrument "Vatimer" was developed to evaluate the coagulation properties of milk in cheese vats. Microvolt signals from the instrument varied with standing-wave motion of milk, forces arising from coagulation, curd firming, curd syneresis and also clean-in-place spray forces. Curd firmness at cutting varied three to four fold in commercial Cheddar cheesemaking operations.

The Formagraph instrument was used to select individual cow milk samples with good and poor chymosin-coagulation characteristics from the Utah State University Holstein herd. Blending 50% good-, and 50% poor-coagulating samples did not improve average coagulation properties of poor samples. Higher chymosin concentration caused …

This study was a comparison of the similarities and differences between the registered nurses' perception and clients' perception of human need fulfillment alterations in the client with uterine cancer. The effect of level of educational preparation and number of years work experience on the nurses' perception was also analyzed. The participants were 15 registered nurses and 15 clients who had uterine cancer and were admitted to two acute care hospitals in Tidewater. The purpose of this study was to explore feelings of loneliness among relocated elderly residents of age-segregated, high-rise apartment buildings and any relationship between these subjective feelings of loneliness and certain variables which were hypothesized as predictors of feelings of loneliness. The variables studied in relation to loneliness were cathectic investment, activity, and perceptions of health. Registered nurses in skilled nursing facilities were surveyed using the Six-Dimension Scale of Nursing Performance to determine: (1) how they rated the importance of specific nursing behaviors related to their role, (2) how they rated the frequency engaged in the same specific nursing behaviors related to their role, and (3) if there was a relationship between how they rated the specific nursing behaviors for importance and frequency. Finger tapping and CVC syllable repetition measures of 7 four to eight year old fluent and disfluent children and their mothers were evaluated. Perceptual ratings of speech rate and effort level were also completed by the children, their mothers, and nine graduate students. These procedures were used to test hypotheses that mean interval durations for tapping and for speech, and perceptual ratings of speech rate and effort level are individual in nature.

Bulimia is a complex eating disorder that involves bouts of binge eating which are usually followed by a purge to rid the individual of the unwanted food. This disorder occurs primarily in females and may lead to several medical and psychological complications. The efficacy of two single component treatments and a package intervention designed to decrease variables related to binge eating were investigated with three bulimic females. The results of the research indicated that individuals who were exposed to the package intervention experienced the largest decrements in the binge eating when compared to the single component procedures.

A selective review of functional and anatomical hemispheric asymmetries in humans confirms the doctrine of hemispheric specialization. However, the post-insult phenomenon of recovery of functions challenges theories that posit too strict a correspondence between anatomical structures and functions. Follow-up studies on hemispherectomized and hemidecorticated patients, who later recovered language, support a hypothesis of hemispheric redundancy, rather than "invariant" hemispheric specialization for language. Thus, through an evolutionary selection process, the brain may have developed as two potentially similar organs. Neuropsychological research on recovery phenomena is criticized as neglecting environmental contributions to recovery. Several models of hemispheric specialization are compared with recovery …

Twelve college students volunteered to participate in a special section of an introductory psychology course at Western Michigan University. A multiple baseline design across groups was used to assess differences in reading comprehension between students who received study objectives written by the instructor and students trained to write their own study objectives. Quizzes and examinations were used to assess students' reading comprehension of course materials. A comparison of pre- and post-test scores on the Nelson-Denny Reading Comprehension Test was used to determine whether students had acquired skills which aided their comprehension of other materials. This study's purpose was to determine if a type of cognitive therapy, Brief Perception Therapy, (Bullmer, 1980) could improve interpersonal perceptual accuracy and psychological adjustment in a substance abuse population. Thirty-five patients seeking inpatient treatment were randomly assigned to a treatment and a comparison group. Twenty-five patients completed the three week program, 13 in the treatment group and 12 in the comparison group. The treatment group received Group Brief Perception Therapy (GBPT) and the comparison group received Regular Group Therapy (RP). The diagnosis of schizophrenia has been considered unreliable, especially when derived by psychiatric interview. Thought disorder is one symptom that has been accepted by most clinicians as a major indicator of schizophrenia. The purpose of this study was to examine the relationship between thought disorder and the diagnosis of schizophrenia in a state hospital setting.

Language intervention strategies of the mothers of ten normal and ten language-impaired children were compared. An experimenter asked each child questions of varying difficulty about a storybook, in the presence of the mother, who was free to help her child as needed. Both the children's and mothers' utterances were coded. The frequency and types of mothers' strategies were analyzed. The analysis revealed that mothers of normal children intervened significantly more often than mothers of language -impaired children, when all interventions were considered. However, the differences were not significant when only interventions related directly to the predetermined questions were considered. The …

The purpose of this study was to compare community mental health attitudes of the past and the present, and between mental health workers and the public. The results indicate that the public is better informed and more favorably disposed toward the mentally ill today than was true several decades ago. Some stereotypes persist, however, and the mentally ill are still regarded less favorably than are other disability groups. Mental health workers and the public appear to be similar in their endorsement of a medical approach to treatment, although the former endorse more strongly a community-based treatment orientation. Ninety-four female nurses completed the Trait portion of the State-Trait Anxiety Inventory developed by Spielberger, Gorsuch, and Lushene (1964) and a demographic inventory. The purpose of this study was to investigate the criteria used to select dental hygiene students and to determine the ability of these criteria to predict students' successful completion of a dental hygiene program. One hundred sixteen graduates of the certificate level dental hygiene program at Old Dominion University between 1971- 1981, inclusive, comprised the sample. Investigation of the coenzyme binding site of lactate dehydrogenase was approached by using coenzyme analogs. Inhibition studies of this enzyme using n-alkylphosphates as coenzyme analogs were performed with a fluorometric method. The results showed that all of the n-alkylphosphates were competitive inhibitors with respect to NAD+.

The n-alkyl-ADPs were synthesized from AMP and dicycloheaylammonium salts of n-alkylphosphates. The synthetic compounds were subjected to NMR spectroscopic studies, thin layer chromatography and determination of the ratio of adenosine moiety to phosphate groups. The results indicated that the synthetic compounds were n-alkyl-ADPs.

In a descriptive correlational study, three predictors of appointment keeping behaviors of adults who attend general medical clinics in a primary health care setting were explored. The three selected predictors from the Health Belief Model, a preventive health behavior model, were: (a) perceived barriers to health care, (b) health locus of control (HLC), and (c) satisfaction with health care. Human serum high density lipoproteins (HDL) were isolated by preparative salt density gradient ultracentrifugation and analyzed further by heparin-sepharose affinity chromatography. Separation of the major classes of serum lipoproteins was achieved after a single ultracentrifugation on a salt density gradient at 272,000 x g for 36 hours in a swinging bucket rotor. After isolation, the total HDL was subdivided into HDL containing apoprotein E and HDL without this apoprotein by heparin-sepharose affinity chromatography. Relative amounts of these subclasses were then calculated.

HDL-cholesterol was measured after precipitation of LDL and VLDL by phosphotungstic acid and magnesium. The statistical relationship between HDL …

This study was designed to investigate the effectiveness of dextrose on cardiovascular endurance. The subjects were nineteen male students enrolled at Loma Linda University in Riverside, California. The subjects were exercised to their maximum while being tested on the treadmill.· Subjects were each tested twice. Thirty minutes before exercising, they ingested one hundred grams of dextrose for one test and ten ounces of sugar-free Kool-aid for the second test. The difference in exhaustion times was then recorded.

The data indicated that dextrose ingestion decreased exhaustion time on the treadmill. Subjects were able to exercise for a longer period of time …

Research has shown that women experience fluctuations in body weight (BW) due to water retention and/or other processes at various times in the menstrual cycle. The purpose of this study is to measure women repeatedly throughout the course of one menstrual cycle and

(1) to confirm whether significant fluctuations in weight occur at various times and

(2) to determine whether there are significant differences in the components of body composition by hydrostatic weighing which are related to any changes in BW.

The purpose of this investigation was to replicate the study conducted by Lee and Canter (1971) and Lee (1974a) to determine if a significant difference among the scores in the two studies existed due to geographical location, and to initiate the establishment of norms for the Portland, Oregon geographical area. Forty children, selected on the basis of chronological age (4.0 to 4.11 years), normal receptive vocabulary skills, normal hearing, and a monolingual background, participated as subjects. A language sample of fifty utterances was elicited from each child and analyzed according to the Developmental Sentence Scoring (DSS) procedure.

In the late 1960's, a new method for advanced airway management in the pre-hospital setting, the esophageal obturator airway (EOA), was introduced. Ever since, it has been the subject of much discussion over whether it was an effective replacement or alternative to the device traditionally used for airway protection, the endotracheal (ET) tube.
