Unidirectional Calcium And Nucleotide Fluxes In Sarcoplasmic Reticulum. I. Interpretation Of Flux Ratios For Different Reaction Schemes, J. J. Feher

The relation between unidirectional calcium and nucleotide fluxes was examined for different ATPase reaction schemes of the sarcoplasmic reticulum. The schemes considered differed in the order of sorption and desorption of calcium, ATP, and ADP. The results suggest that the theoretical relation between calcium and nucleotide fluxes depends on the reaction scheme and that experimental measurements can distinguish among them. The results obtained are generally valid and do not depend on assumptions of equilibrium or pseudoequilibrium between intermediate states of the pump.

ABSTRACT

It has been suggested that potentials measured with conventional microelectrodes in chemically or mechanically skinned muscle fibers arise from a Donnan equilibrium due to myofilament fixed charges. This hypothesis was tested in mechanically skinned frog (Rana pipiens) semitendinosus fibers by measuring the distribution potential (Ed) between fiber and bath with 3 M KCI-filled microelectrodes and the K+ activity gradient (a'K/a) with K+ ion-selective microelectrodes (KISE). If skinned fibers are a Donnan system, Ed should become more positive as pH is decreased, altering the fixed charge on the myofilaments. Unidirectional calcium influx and efflux were evaluated in cardiac sarcoplasmic reticulum (SR) by 45Ca-"0Ca exchange at steady state calcium uptake in the absence of calcium precipitating anions. Calcium efflux was partitioned into a pump-mediated efflux and a parallel passive efflux by separately measuring passive efflux referable to the steady state. Unidirectional and net ATP-ADP fluxes were measured using [3H]-ATP -- ADP and [3H]- ADP -- ATP exchanges. Methods are presented that take into account changing specific activities and sizes of the nucleotide pools during the measurement of nucleotide fluxes. Antimalarial activities ofchloroquine, mefloquine, amodiaquine, and quinine in vitro against Plasmodium falciparum were diminished as a consequence ofmembranefiltration. Filtered drug solutions gave ID₅₀ values up to 25-fold greater than those of non-filtered (ethanol-sterilized) drug solutions. Loss ofactivity byfiltration was overcome by increasing the drug concentration prior to filtration. Water solutions filtered through Millex-GSfilter units consistently showed an absorbance maximum at 277 nm, accompanied by a lesser peak at 225 nm. Water filtrates from Nucleopore and Millex-G Vfilters showed no absorbance at 277 nm and only slight absorbance was evidentfor the Gelmanfilter unit. This Article investigates two alternative methods of human conception: Specifically, the artificial insemination of unmarried women for either their own personal purposes of pregnancy without the benefit of marriage or as surrogates for infertile women. Surrogation is evaluated, then, as an analytic complement to the sexual privacy of women who are expressing their sexual freedom through unconventional means to become pregnant.

The conclusion drawn is that an unmarried woman’s fundamental right to privacy or procreation does not encompass a right to either artificial insemination or surrogation. To allow unfettered access to these two methods of conception would - quite simply …

The invention relates to a method for hydrolyzing drug glucuronic acid conjugates present in mammalian body fluids, the conjugates being derived from a narcotic analgesic, antagonist, or agonist-antagonist whose metabolism includes conjugation with glucuronic acid. The method comprises incubating the body fluid sample at from about 60 to about 70° C., for at least about 1 hour, with 3-glucuronidase derived from Patella vulgata, and substantially increases the sensitivity of chromatographic techniques for the detection of morphine and its analogues.

The proper understanding of obesity requires a multifaceted approach. Behavioral considerations of eating and activity patterns do not account for the large between- and within-subjects variance associated with the energy-balance equation. Sources of adaptive and dispositional variance in metabolic rates are reviewed and suggested to be a likely source of importance for the proper conceptualization and intervention of obesity. Five proposed mechanisms of metabolic variation are reviewed with consideration of the supporting evidence for each mechanism. Thirty years have passed since the discovery of Thorazine, a neuroleptic drug, and the drugging of American state mental patients has become commonplace. Part I distinguishes between two approaches to remedy--"structural" and "atomistic"--and, as a basis for testing the two, describes a state hospital's handling of the most serious drug side effect. This account also provides a sense of the dimensions of the drugging problems in state hospitals. Part II explores a family of atomistic remedies. Small resting B lymphocytes are activated when their cell surface immuno-globulin (Ig) receptors interact with specific antigen or anti-Ig antibodies. Such a receptor-ligand interaction leads to enlargement and proliferation of B lymphocytes. These activated B cells can subsequently mature into Ig-secreting cells in the presence of appropriate accessory cell signals (1-3). However, monovalent anti-Ig antibodies cannot induce B cell activation even though they can bind to the surface Ig (4-7). It has thus been concluded that membrane Ig has to be cross-linked to induce early steps in B cell activation (membrane depolarization; 8) as well as later steps (proliferation). T …

Previous literature reviews are updated. Recent findings impugn the WISC-R internal consistency and document item bias for American Indian children. A pattern of Spatial > Sequential > Conceptual > Acquired Knowledge holds across ages, tribes, and heterogeneous referral sources, except for acculturated children. Kaufman's three factors are replaced by Verbal and Performance factors for Papago and Navajo children. A culturally-learned basis for intellectual functioning among traditional children supports alternative assessment functions for traditional reservation lifestyle and for acculturation and entree into mainstream society. Performance measures, SOMPA, Piagetian and Luria-derived tasks may ultimately provide less biased intelligence estimates. Recent legislation outlines ethical practice although …

Too often when development planners or practitioners plan income-generating schemes for women they consider only handicrafts. While in some situations craft production may provide a secure source of income for women, in many cases it results in poor returns and proves more complicated an undertaking than expected. The Bangladesh Rural Advancement Committee (BRAC) is one agency that has developed a successful program of non-craft employment opportunities for women. Some 10,000 poor women have been engaged by BRAC in viable economic schemes: 9,000 in non-craft production. Parental ratings of overactivity are unrelated to teachers' ratings of overactivity or to laboratory measures of inattention. Teachers' ratings of overactivity correlate strongly with teachers' ratings of inattention as well as to laboratory measures of inattention. Teachers' ratings of inattention, however, are unrelated to laboratory measures of inattention when IQ is controlled. There are considerable definitional problems relating to the symptoms of hyperactivity and inattention, making it difficult create a behavioral definition of the psychiatric syndrome of attention deficit disorder.
