Diabetes mellitus (DM) is a major health issue that has reached alarming levels, affecting around 537 million of the global adult population. DM is a group of metabolic diseases characterized by hyperglycemia with disturbance in carbohydrate, fat, and protein metabolism. The proposed therapeutic options for DM are different according to their mechanism of action, and peptide therapy is one of these options. Adropin, a peptide hormone involved in energy homeostasis, has shown promising results in the treatment of diabetes by lowering fasting blood glucose, reducing insulin resistance, and minimizing fat storage. Utilizing fermentation with probiotics offers a promising opportunity to leverage agricultural by-products, such as date palm pomace (DPP), in order to drive forward sustainable food production endeavors. This dissertation aimed to (1) isolate yeast from fermented food products as well as to characterize their probiotic properties; (2) investigate the physiochemical properties of fermented DPP by selected yeast isolates; (3) evaluate the health benefits of non-fermented and fermented DPP samples both pre and post-digestion (bioaccessible portions); and (4) analyze the untargeted metabolites and carbohydrates metabolites in the bioaccessible portions. BACKGROUND: Urinary tract infections (UTIs) are one of the most common bacterial infections worldwide. As global incidences of UTIs caused by multidrug resistant bacteria continue to increase, so too does the demand for novel antimicrobial therapies.

Due to its relatively understudied nature, the urinary microbiome represents a niche with an untapped source of potentially novel antimicrobials (e.g., bacteriocins). Improvements to bacterial culturing and sequencing techniques have highlighted these potential alternative treatments and control strategies to target antibiotic resistant uropathogenic E. coli (UPEC).

The Epstein-Barr Virus (EBV) or human herpesvirus 4 (HHV-4) is the sole etiologic agent of the acute illness in humans described either as infectious mononucleosis (IM), or glandular fever. IM is a self-limiting disease with various but transient symptoms which include fever, fatigue, malaise, sore throat, swollen lymph glands (particularly of the neck), swollen liver and spleen. Diagnosis relies on clear, timely and informative laboratory test results. Patients with primary IM infection often present on investigation with a well-documented population of atypical/plasmacytoid lymphocytes. Neonatal infection is a significant cause of mortality and morbidity in infants. Globally, neonatal pathogens are increasingly becoming multi-drug resistant, indicating the need to discover novel alternative treatment strategies. Nisin is an antimicrobial peptide that exhibits broad-spectrum activity against Gram-positive and Gram-negative bacteria including a wide variety of clinical pathogens. Nisin can be used in combination with antibiotics to improve their efficacy. This study examined the activity of nisin and bioengineered derivatives against multi-drug resistant Streptococcus agalactiae and Staphylococcus capitis isolates and investigated the potential synergy between nisin peptides and selected antibiotics. Bifidobacteria are beneficial commensals of the human gastrointestinal tract and their presence in the gut has been associated with positive health effects on the host. They account for a vast proportion of the infant gut microbiota, when the infant is fed on a milk-based diet, with their number progressively decreasing in adult and elderly.
