Contamination is a major issue in the biopharmaceutical industry. Bioreactors must be meticulously cleaned to remove residual impurities and avoid cross-contamination. Cleaning validation is necessary to ensure residual contaminants are reduced to the lowest acceptable limits. Bacterial cells represent the paramount challenge in manufacturing equipment cleaning because they are composed of insoluble organic contaminants. Total organic carbon (TOC) analysis is a fast, highly sensitive, and inexpensive analytical technique commonly used to evaluate the approximate level of organic contamination.

Background: Hemorrhoids are a common anorectal condition. In Thai traditional medicine, the Bantaoridsidwangthawan formula (BRF) has been used to treat hemorrhoids. We aimed to evaluate the clinical efficacy and satisfaction of BRF versus Micronized purified flavonoid fraction (MPFF) in patients with internal hemorrhoids grades 1 and 2.

Selective targeting of cancer cells via overexpressed cell-surface receptors is a promising strategy to enhance chemotherapy efficacy and minimize off-target side effects. In this study, we designed peptide 31 (YHWYGYTPERVI) to target the overexpressed epidermal growth factor receptor (EGFR) in triple-negative breast cancer (TNBC) cells. Peptide 31 is internalized by TNBC cells through EGFR-mediated endocytosis and shares sequence and structural similarities with human EGF (hEGF), a natural EGFR ligand. Unlike hEGF, peptide 31 does not induce cell migration in TNBC cells. This population-based cohort study examines the appropriateness of antibiotic prescribing in South Carolina via aggregated pharmacy claims data matched with diagnosis codes from medical claims. Inappropriate antibiotic prescribing decreased from 30.2% in 2012 to 22.6% in 2017 (P < 0.001) and was more common in adults >40 years old.

Pholcodine, an anti-tussive medication widely used as an over-the-counter, OTC drug, has recently faced restrictions in several countries. This paper presents a sensitive electrochemical approach for pholcodine detection. The electrochemical method involved fabricating a graphene nanoplatelets electrode, incorporating polythiophene nanospheres polymer to promote electron transfer and increase the activated surface area. Characterization of the fabricated electrode was performed using transmission electron microscopy, ATR-Fourier-transform infrared spectroscopy, X-ray crystallography, X-ray photoelectron spectroscopy, and electrochemical impedance spectroscopy. The addictive effect of opioids impacted 60 million people globally in 2023. This opioid crisis necessitates the development of opioid derivatives that maintain analgesic efficacy while minimizing addictive effects from opioid use. The present work focuses on morphine, a commonly prescribed opioid with high affinity to the mu-opioid receptor (MOR). Morphine binds non-selectively within both healthy and inflamed tissue. By modifying morphine’s structure through fluorination to counteract its non-selective binding, the drug’s pharmacological ability to provide effective pain relief comes with a lower risk of addiction. Many newly approved immediate-release (IR) solid oral drugs are accompanied by broad "do-not-crush" warnings, often without specific explanations. Building on our recent publication of crushability analysis of novel drugs approved between 2020 and 2022, this research extends the assessment to the approvals of 2023. Our goal is to provide healthcare providers with timely recommendations for the safe manipulation of immediate-release tablets and capsules for patients who cannot swallow them whole.

This medication use evaluation found that prescribing practices of DOAC and warfarin did not change after AHA guidance publication. Patients with HFrEF were more often discharged on warfarin, while those with Afib were more often discharged on DOACs. Those discharged on a DOAC had the highest incidence of LV thrombus resolution up to 6 months post-discharge. Physicians within Parkview Health do not routinely utilize a loading dose when discharging a patient on a DOAC.

This study found that anti-pseudomonal (PsA) coverage is overutilized in diabetic foot infection (DFI), even when broad qualifying criteria are applied. The proposed criteria were poor predictors of PsA growth, which was present in a small number of cases. Patients who did grow PsA were somewhat more likely to have severe DFI, history of DFI, and 90-day history of admission or anti-PsA antibiotic use, but many patients without PsA growth also met these criteria.

Acute Ischemic Stroke (AIS) is a leading cause of disability and death in the United States, requiring urgent treatment1,2; earlier treatment improves patient outcomes.

Fibrinolytic agents including alteplase and tenecteplase (TNK) reduce long-term disability in AIS but are associated with hemorrhagic risks3,4.

As of January 2024, Corewell Health East hospitals updated their guidelines that TNK would be the first line agent in the treatment of AIS.

Alteplase is the only approved agent for AIS, but TNK shows similar effectiveness along with shorter preparation and administration time which may ultimately improve door-to-needle time2 .

Continuous overactivation of myofibroblasts leads to excessive deposition of profibrotic proteins, which in turn causes tissue stiffness and, ultimately, organ failure. One of those proteins, Cadherin-11 (CDH11), is reported to be overexpressed in the tissue of fibrotic skin, specifically on myofibroblasts. Similarly, chronic inflammation drives fibrosis progression in autoimmune-related illnesses like systemic sclerosis (SSc). Therefore, it is hypothesized that CDH11 could be a target for cell-specific delivery of antifibrotic therapies to fibrotic skin tissue. Dalbavancin is a lipoglycopeptide approved for the treatment of acute bacterial skin and soft tissue infections caused by gram-positive microorganisms.

Used off-label in the management of infective endocarditis in IV drug users, as well as in osteomyelitis infections.

In February 2022, the University of Rochester Medical Center (URMC) updated their practice guidelines, in accordance with the Infectious Diseases Society of America, to include fidaxomicin as the preferred agent in patients with initial (non-fulminant) or first recurrence of C. difficile infection (CDI).

Health system protocols were updated to include polymerase chain reaction (PCR) testing for patients with suspected CDI followed by confirmation via toxin enzyme immunoassay (EIA).

In the appropriate clinical context, PCR+/EIA+ test confirms active CDI and warrants antibiotic treatment.

PCR+/EIA- test indicates likely colonization and may preclude the need for treatment.
