One Ion To Rule Them All: Combined Antibacterial, Osteoinductive And Anticancer Properties Of Selenite-Incorporated Hydroxyapatite, Vuk Uskoković, Maheshwar Adiraj Iyer, Victoria M. Wu

Pharmacy Faculty Articles and Research

Although hydroxyapatite (HAp) has been doped with dozens of different ions, the quest for an ion imparting a combination of properties conducive to bone healing is still ongoing. Because of its protean potency and the similarity in size and shape to the phosphate tetrahedron, the selenite ion presents a natural ionic substitute in HAp. The incorporation of selenite into synthetic HAp using two different methods – co-precipitation and ion-exchange sorption – was studied for its effect on crystal properties and on a triad of biological responses: antibacterial, anticancer and osteoinductive. Co-precipitation yielded HAp with a higher selenite content than sorption …

Reflections From The Bedsides Of Dying Patients, Hannah Decleene, Patrick Kukulich

Collaborative Healthcare: Interprofessional Practice, Education and Evaluation (JCIPE)

I walked into a room with its lights glaring and television blaring, a room where a man I had never met before would take his final breath two hours later. The attending nurse kindly dimmed the lights and turned off the television. I sat down next to him. I felt panic. I felt sadness, and I felt peace. This individual would have passed without me knowing, yet I was there with him; he wouldn’t be alone.

The No One Dies Alone (NODA) chapter at Jefferson launched in May, 2016. It launched as a vision in which students of Thomas Jefferson …

Camptotheca Decaisne is endemic to southern China. Since 1934, C. acuminata has been widely introduced to many gardens and arboreta in North America, Asia, and Europe as living collections. Our national surveys in China between 1995 and 1998 indicated that the genus. Camptotheca spp. are severely endangered in native range. Our field surveys failed to locate any wild populations of C. acuminata although it is often cultivated as landscape trees in southern China. We could not identify any living trees of C. acuminata var. tenuifolia and var. rotundifolia. We estimated that there are approximately 500 mature trees of C. lowreyana …

Biophysical Approaches Facilitate Computational Drug Discovery For Atp-Binding Cassette Proteins, Steven V. Molinski, Zoltan Bozoky, Surtaj H. Iram, Saumel Ahmadi

Chemistry and Biochemistry Faculty Publications

Although membrane proteins represent most therapeutically relevant drug targets, the availability of atomic resolution structures for this class of proteins has been limited. Structural characterization has been hampered by the biophysical nature of these polytopic transporters, receptors, and channels, and recent innovations to in vitro techniques aim to mitigate these challenges. One such class of membrane proteins, the ATP-binding cassette (ABC) superfamily, are broadly expressed throughout the human body, required for normal physiology and disease-causing when mutated, yet lacks sufficient structural representation in the Protein Data Bank. However, recent improvements to biophysical techniques (e.g., cryo-electron microscopy) have allowed for previously …

Doxorubicin Cytotoxicity In A Human Proximal Tubular Epithelial Cell Line Was Attenuated By The Natural Product Resveratrol, Morghan Schuyler Getty

Theses, Dissertations and Capstones

The cancer chemotherapeutic agent doxorubicin (DOX), Adriamycin, is part of the treatment regimen for breast, ovarian, small cell lung cancer and acute/chronic lymphoid leukemia. Adverse effects associated with DOX are cardiotoxicity and nephrotoxicity. Interventions are needed to reduce DOX nephrotoxicity. Resveratrol (RES) is a phytochemical contained in grapes, berries and nuts, which possesses antioxidant and anticancer properties. This study tested the hypothesis that RES will attenuate DOX renal cytotoxicity in human noncancerous renal proximal tubular epithelial (HK-2) cells and that RES will reduce DOX mediated changes in mitochondrial function. HK-2 cells were plated and grown for 48 hours (h). Cells …

Formulation And Evaluation Of Fast Disintegrating Sublingual Atropine Tablets For The Potential Treatment Of Acute Organophosphates Toxicity, Alhussain Aodah

Student Theses, Dissertations and Capstones

Organophosphates (OP) are chemicals found in certain pesticides and in nerve agents such as sarin. Toxicity with these agents can lead to death. On August 2013, 1,300 civilians were killed using sarin gas in Syria by the Assad regimen. The lack of prepared medical facilities, and the availability of appropriate treatmentsignificantly contributed to the high percent of causality. Further, worldwide pesticides toxicity affects 3 million people annually. Therefore, it is imperative that antidote to treat OP toxicity are readily available in a dosage form that is acceptable cost, effective, and user friendly. Atropine sulfate (AS) is the first-line antidote forOP …

The Anti-Sepsis Activity Of The Components Of Huanglian Jiedu Decoction With High Lipid A-Binding Affinity, Guirong Chen, Yubin Xu, Jing Jing, Brianna Mackie, Xinchuan Zheng, Xu Zhang, Jing Wang, Xuetao Li

Medicinal Chemistry Publications

Huanglian Jiedu Decoction (HJD), one of the classic recipes for relieving toxicity and fever, is a common method for treating sepsis in China. However, the effective components of HJD have not yet been identified. This experiment was carried out to elucidate the effective components of HJD against sepsis. Thus, seven fractions from HJD were tested using a biosensor to test their affinity for lipid A. The components obtained that had high lipid A-binding fractions were further separated, and their affinities to lipid A were assessed with the aid of a biosensor. The levels of LPS in the blood were measured, …

Silver Nanoparticles And Drug Resistant Bacteria, Aliza Eisig Jan 2017

Silver Nanoparticles And Drug Resistant Bacteria, Aliza Eisig

The Science Journal of the Lander College of Arts and Sciences

The scientist who discovered penicillin and its use as an antibiotic, Alexander Fleming, also raised concerns about bacterial resistance. As he predicted, in the twenty first century, the overwhelming use of antibiotics has led to both drug- and multi-drug resistant bacteria. This paper attempts to investigate the antibacterial potential of silver nanoparticles against drug resistant bacteria. By using Touro's online library database, the efficacy of silver nanoparticles as a potential antibacterial agent was comprehensively researched. Using transmission electron microscopy and the disk diffusion method, silver nanoparticles have been found to exert bactericidal effects by adsorbing to the cell surface and …

Variation In Organ-Specific Pik3ca And Kras Mutant Levels In Normal Human Tissues Correlates With Mutation Prevalence In Corresponding Carcinomas, Barbara L. Parsons, Karen L. Mckim, Meagan B. Myers

United States Food and Drug Administration: Publications

Large-scale sequencing efforts have described the mutational complexity of individual cancers and identified mutations prevalent in different cancers. As a complementary approach, allele specific competitive blocker PCR (ACB-PCR) is being used to quantify levels of hotspot cancer driver mutations (CDMs) with high sensitivity, to elucidate the tissue-specific properties of CDMs, their occurrence as tumor cell subpopulations, and their occurrence in normal tissues. Here we report measurements of PIK3CA H1047R mutant fraction (MF) in normal colonic mucosa, normal lung, colonic adenomas, colonic adenocarcinomas, and lung adenocarcinomas. We report PIK3CA E545K MF measurements in those tissues, as well as in normal breast, …

Cyclosporine Exacerbates Ketamine Toxicity In Zebrafish: Mechanistic Studies On Drug–Drug Interaction, Bonnie L. Robinson, Melanie Dumas, Syed F. Ali, Merle G. Paule, Qiang Gu, Jyotshna Kanungo

United States Food and Drug Administration: Publications

Cyclosporine A (CsA) is an immunosuppressive drug commonly used in organ transplant patients to prevent allograft rejections. Ketamine is a pediatric anesthetic that noncompetitively inhibits the calcium-permeable N-methyl-D-aspartic acid receptors. Adverse drug–drug interaction effects between ketamine and CsA have been reported in mammals and humans. However, the mechanism of such drug–drug interaction is unclear. We have previously reported adverse effects of combination drugs, such as verapamil/ketamine and shown the mechanism through intervention by other drugs in zebrafish embryos. Here, we show that ketamine and CsA in combination produce developmental toxicity even leading to lethality in zebrafish larvae when exposure began …

Detection Of Allergen Markers In Food: Analytical Methods, Girdhari M. Sharma, Sefat E. Khuda, Christine H. Parker, Anne C. Eischeid, Marion Pereira

United States Food and Drug Administration: Publications

Food allergens are proteins that are well tolerated by most, but can cause severe reactions in sensitive individuals. Since there is no cure for food allergy, strict adherence to an allergen-free diet is the only safe choice currently available for allergic consumers. Accurate food labeling can help consumers avoid foods containing an allergenic ingredient. Regulatory agencies have mandated the labeling of major food allergens on packaged foods to help with safe food choices. However, the inadvertent presence of an allergen in food due to cross-contact and labeling error can jeopardize consumer health. Analytical methods are developed for allergen detection and …
