Thermodynamic Mechanisms Of Helix Stabilization In A Model Peptide And Protein, Ryan Murray Jan 2022

Thermodynamic Mechanisms Of Helix Stabilization In A Model Peptide And Protein, Ryan Murray

University of the Pacific Theses and Dissertations

Biologics are large, complex therapeutic agents generally produced from living organisms. One group of biologics is peptide/protein based. Biological agents offer unique advantages over traditional therapeutics including longer half-lives, higher specificity, greater efficacy, and reduced off-target effects. However, protein/peptide based drugs suffer from both delivery and stability issues. The higher order of protein structures (secondary, tertiary, etc.) derive ~80% of their conformational stability from paltry hydrophobic effects, with net stabilization of 5-15 kcal/mole observed for many proteins. Loss of conformational stability can lead to increased aggregation, precipitation, and degradation; and reduced activity and side effects. To increase stability and improve …

Peripherally Restricted Opioid Conjugates And Its Use As Pharmacological Probes And Potential Therapeutics, Md Tariqul Haque Tuhin Jan 2022

Peripherally Restricted Opioid Conjugates And Its Use As Pharmacological Probes And Potential Therapeutics, Md Tariqul Haque Tuhin

University of the Pacific Theses and Dissertations

Opioid-induced constipation (OIC) is one of the major adverse effects of opioid analgesics used by millions of patients each year. While progress has been made, there remains a significant unmet medical need in the treatment of OIC. Major gaps remain in our understanding of the role of the gastrointestinal tract and central nervous system (CNS) in precipitating OIC. For the last four decades, numerous investigations to study the sites of action of opioid analgesics have utilized peripherally acting mu-opioid receptor antagonists (PAMORAs), which have been incorrectly believed to have limited penetration across the blood-brain barrier (BBB). Several preclinical and clinical …

Allergic asthma is a chronic inflammatory respiratory disease associated with eosinophilic infiltration, increased mucus production, airway hyperresponsiveness, and airway remodeling. Epidemiologic data reveal that the prevalence of allergic sensitization and associated diseases has increased in the twentieth century. This has been hypothesized to be partly due to reduced contact with microbial organisms (the hygiene hypothesis) in industrialized society. Airway epithelial cells, once considered a static physical barrier between the body and the external world, are now widely recognized as immunologically active cells that can initiate, maintain, and restrain inflammatory responses, such as those that mediate allergic disease. Airway epithelial cells …

Design And Synthesis Of Tetrahydrocurcumin-Amino Acid Conjugates For Cytotoxicity Enhancement Against Brain Cancer C6 Cells, Polsak Teerawonganan

Chulalongkorn University Theses and Dissertations (Chula ETD)

The L-type amino acid transporter I (LAT1) is an attractive target for drug development in glioma with a poor prognosis. LAT1 facilitates the cross-membrane transport of essential amino acids and plays a crucial role in cancer cell proliferation and growth. In this study, a novel series of tetrahydrocurcumin (THC)-amino acid conjugates were synthesized by conjugating four amino acids: glycine (Gly), leucine (Leu), isoleucine (Ilu), and phenylalanine (Phe) to THC via carbamate bonds to improve the anticancer effects of THC by mediating LAT1. The therapeutic efficacy of THC-amino acid conjugates was further examined in C6 glioma cells, including the role of …

Authentication Of Andrographis Paniculata And Herbal Products Using Dna Barcoding And High-Resolution Melting Analysis, Ei Mon Cho

Chulalongkorn University Theses and Dissertations (Chula ETD)

Andrographis paniculata (AP) exhibits a wide range of therapeutic indications and effective treatment, leading to the increment of the demand for this herbal remedy. Due to the limitation of both fresh AP and AP commercial products, this resulted in the discovery of adulterations with bitter-tasting herbs like Tinospora crispa (TC) and Azadirachta indica var. siamensis (AI) with different indications. Therefore, a suitable method for the identification of the herb of interest is required. While conventional authentication methods for AP have limitations, the DNA barcoding technique has become a potential method for identifying medicinal plants in many countries' pharmacopoeias. This study …

The use of monoclonal antibodies (mAbs) specific against immune checkpoints has displayed satisfactory clinical benefits. However, its effectiveness is limited to just a small percentage of cancer patients. In this work, a recombinant DVD-AT2C bispecific antibody (bsAb) was developed for dual inhibition of PD-1/PD-L1 and CTLA-4 axes and produced in plants. Plant system was utilized as an alternative protein production platform. Transient expression of DVD-AT2C targeting PD-L1 and CTLA-4 in Nicotiana benthamiana was determined highest after 4 days of infiltration. The size, assembly, and protein properties of the purified bsAb were …

Development Of Plant-Produced Anti-Il-6r Mab, Namthip Kaewbandit Jan 2022

Development Of Plant-Produced Anti-Il-6r Mab, Namthip Kaewbandit

Chulalongkorn University Theses and Dissertations (Chula ETD)

After the body is invaded by pathogens, it responds by secreting cytokines, for example, interleukin 6 (IL-6), which activates an intracellular signaling pathway involving JAKs/STAT3 through phosphorylation. However, overexpression of pSTAT-3 can lead to a positive feedback loop of IL-6, causing the release of more cytokines and potentially leading to cytokine storm. Thus, inhibition of IL-6 signaling is another important target for Cytokine storm treatment. Tocilizumab, a humanized IL-6R antibody has been approved for RA treatment. There are several concerns about mammalian cell protein expression systems, such as higher production costs and pathogen contamination risks. Recently, plants have become increasingly …

Titania Nanosheets Induce Anticancer Activityin Human Non-Small Cell Lung Cancer Cells, Rapeepun Soonnarong

Chulalongkorn University Theses and Dissertations (Chula ETD)

Metal nanomaterials can enhance the efficacy of current cancer therapies. Here, we show that Ti0.8O2 nanosheets cause cytotoxicity in several lung cancer cells but not in normal cells. The nanosheet-treated cells showed certain apoptosis characteristics. Protein analysis further indicated the activation of the p53-dependent death mechanism. Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) analyses revealed the cellular uptake of the nanosheets and the induction of cell morphological change. Ti0.8O2 nanosheets induce apoptosis through a molecular mechanism involving peroxynitrite (ONOO−) generation. As peroxynitrite is known to be a potent inducer of S-nitrosylation, we further found that the nanosheets mediated …

Identification Of Potential Drug Targets For Covid 19 Patients With Cytokine Storm, Bayu Cakra Buana Jan 2022

Identification Of Potential Drug Targets For Covid 19 Patients With Cytokine Storm, Bayu Cakra Buana

Chulalongkorn University Theses and Dissertations (Chula ETD)

The outcome of SARS-CoV-2 infection may vary depending on gender, lifestyle, and age. The elderly is more likely to have the severe condition than the younger population. However, several young people suffer from severe lung diseases caused by cytokine dysregulation, also known as a "cytokine storm." Cytokine storms are associated with Covid-19 severity and mortality. Identification of potential cytokine storm targets is critical for improving patient survival. Transcriptomic analysis of expression data from publicly available databases could be used to identify differentially expressed genes and pathways. The list of both results was used to narrow down the potential candidates in …

Renieramycin T Derivative Dh_22 Induces Apoptosis In Lung Cancer Cells By Downregulation Of C-Myc Protein, Indiana Gita Anggraeni Jan 2022

Renieramycin T Derivative Dh_22 Induces Apoptosis In Lung Cancer Cells By Downregulation Of C-Myc Protein, Indiana Gita Anggraeni

Chulalongkorn University Theses and Dissertations (Chula ETD)

Lung cancer is a prevalent cause of death, indicated by an elevated c-Myc level. We synthesized a new derivative of renieramycin T (RT), named DH_22, and tested it for anti-cancer activities in human lung cancer cells. In the cell viability and nuclear staining tests, DH_22 was discovered to be cytotoxic, with an IC50 of around 13 μM; and found to mediate apoptosis in the lung cancer cells. Meanwhile, in a mechanistic sense, DH_22 contributed to the activation of p53-dependent apoptosis and decreased the cellular level of c-Myc. The p53-dependent mechanism was indicated by an increase of p53, an induction of …

Chemometric Attenuated Total Reflectance Fourier Transform Infrared Spectroscopy Method For Routine Screening Of Paracetamol, Ibuprofen, And Aspirin Adulteration In Herbal Medicines, Mario Theodore

Chulalongkorn University Theses and Dissertations (Chula ETD)

Simple and reliable screening tools for herbal medicine drug adulteration are required, especially in rural areas where complex instrumental methods like HPLC face challenges such as logistics and complex operational processes. Attenuated Total Reflectance Fourier Transform Infrared (ATR-FTIR) is a good identification tool in drug quality control, and its operation is much simpler compared to HPLC, as it requires less sample preparation and has a simple operation. To improve its selectivity in analyzing multicomponent samples like herbal medicine, ATR-FTIR could be combined with chemometric methods such as Linear Discriminant Analysis (LDA). In order to establish itself as a routine testing …

Development Of Surrogate Sars-Cov-2 Virus Neutralization Test Method Using Plant-Produced Recombinant Proteins, Perawat Jirarojwattana Jan 2022

Development Of Surrogate Sars-Cov-2 Virus Neutralization Test Method Using Plant-Produced Recombinant Proteins, Perawat Jirarojwattana

Chulalongkorn University Theses and Dissertations (Chula ETD)

Detecting immunity against SARS-CoV-2 is vital for evaluating vaccine response and natural infection, but a conventional virus neutralization test (cVNT) requires BSL3 and live viruses, and a pseudo-virus neutralization test (pVNT) needs specialized equipment and trained professionals in BSL2. The surrogate virus neutralization test (sVNT) was developed to overcome these limitations. This study explored the use of angiotensin converting enzyme (ACE2) produced from Nicotiana benthamiana for the development of an affordable neutralizing antibodies detection assay. The results showed that the plant-produced ACE2-His can bind to the receptor binding domain (RBD) of the SARS-CoV-2, and was used to develop sVNT with …

Development Of Plant-Produced Subunit Vaccines To Elicit The Immunogenicity Against Sars-Cov-2 Variant Viruses, Narach Khorattanakulchai

Chulalongkorn University Theses and Dissertations (Chula ETD)

Since the emergence of the coronavirus pandemic in 2019 (COVID-19), the development of effective vaccines to combat the infection has accelerated worldwide. While the severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2) with mutations in the receptor-binding domain (RBD) with high transmissibility, enhanced infectivity, and immune escape from vaccination is also predominantly emerging. Effective vaccines against variant of concern (VOC) and optimized booster vaccination strategies are thus highly required. Here, the gene encoding seven different RBD (ancestral (Wuhan), Alpha, Beta, Gamma, Kappa, Delta, and Epsilon variants) fused with the fragment crystallizable region (Fc) of human IgG1 (RBD-Fc) was constructed and cloned into …

A Cost-Utility Analysis Comparing Transcatheter And Surgical Pulmonary Valve Replacement Among Indonesia Patients With A History Of Newborn Right Ventricular Outflow Tract Correction, Andi Nurul Annisa

Chulalongkorn University Theses and Dissertations (Chula ETD)

Transcatheter pulmonary valve replacement (TPVR) was launched since 2000. It indicated for cardiac patients who have the right ventricular outflow tract obstruction (RVOT) and a prior surgery history. TPVR have a shorter hospital stay, fewer complications, and a less-invasive method. However, TPVR is more expensive than surgical pulmonary valve replacement (SPVR). This raises access concerns for low income countries. There are no cost-effectiveness studies in Indonesia, and limited efficiency evidence due to rare disorder. It is a challenge in building the appropriate access strategies. Therefore, this study aims to investigate the cost-utility, resulted in total costs, quality-adjusted life-years (QALYs), life-years …
