Production And Characterization Of Monoclonal Antibodies Against Botulinum Toxin Produced In Nicotiana Benthamiana, Kornchanok Sangprasat
Chulalongkorn University Theses and Dissertations (Chula ETD)
Botulism is an uncommon but serious neurological condition caused by the botulinum toxin produced by the gram-positive bacterium named Clostridium botulinum. Common symptoms of botulism include muscle cramps, nausea, vomiting, diarrhea, difficulty swallowing, respiratory failure, muscle weakness, and potentially death. Currently, two types of antitoxin are used: equine-derived heptavalent antitoxin and human-derived immunoglobulin (BabyBIG®). However, treatment with heptavalent antitoxin may lead to hypersensitivity reactions, while BabyBIG®, which is produced in low yield, is used only for children. This research focuses on developing three anti-BoNT monoclonal antibodies (mAbs), 1B18, C25, and M2, which will be produced using Nicotiana benthamiana as a …

Despite the extensive research successes and continuous developments in modern medicine in terms of diagnosis, prevention, and treatment, the lack of clinically useful disease-modifying drugs or immunotherapeutic agents that can successfully treat or prevent neurodegenerative diseases is an ongoing challenge. To date, only one of the 244 drugs in clinical trials for the treatment of neurodegenerative diseases has been approved in the past decade, indicating a failure rate of 99.6%. In corollary, the approved monoclonal antibody did not demonstrate significant cognitive benefits. Thus, the prevalence of neurodegenerative diseases is increasing rapidly. Therefore, there is an urgent need for creative approaches …

Impact Of A Culturally-Grounded And Interprofessional Approach To Managing Latino/Hispanic Patients At Risk For Or Diagnosed With Type 2 Diabetes Mellitus: A Pilot Study, Jahnavi Yalamanchili
University of the Pacific Theses and Dissertations
Background: Despite significant progress in diabetes research, there remain significant health disparities between Hispanic/Latino patients in the United States, particularly those with lower socioeconomic status, compared to non-Hispanic white patients. Previous studies have identified potential reasons behind these disparities, including the need for a multifaceted approach that addresses social determinants of health (SDOH) faced by patients. Social cognitive theory (SCT) stipulates that interventions that promote self-management by improving knowledge, self-efficacy, and problem-solving skills can be successful in improving health outcomes. However, there is a paucity of studies that have evaluated an interprofessional approach to providing culturally relevant care, in particular …

4d Epr Oximetry Imaging For 4d Bioprinting And Tissue Engineering, Sajad Sarvari
Graduate Theses, Dissertations, and Problem Reports (ETD)
Bioprinting, akin to 3D printing, employs cell-laden hydrogels such as GelMA (Gelatine Methacrylate) and Alginate instead of plastic or resin to create biological structures. The printing process followed by subsequent tissue maturation is known as 4D bioprinting, where the fourth dimension is time. Ensuring adequate and consistent oxygen, O2, supply within 3D bioprint volume over the entire maturation period process is crucial, as even transient hypoxia can impact cellular behavior and phenotypes. However, printing of fully developed vasculature remains an unresolved technical challenge. Several approaches to chemically (peroxides) or biologically (photosynthesis) generate oxygen during bioprint maturation have been proposed. However, …

Lysosomes and the endoplasmic reticulum (ER) are Ca2+ stores mobilized by the second messengers NAADP and IP3, respectively. Here, we establish Ca2+ signals between the two sources as fundamental building blocks that couple local release to global changes in Ca2+. Cell-wide Ca2+ signals evoked by activation of endogenous NAADP-sensitive channels on lysosomes comprise both local and global components and exhibit a major dependence on ER Ca2+ despite their lysosomal origin. Knockout of ER IP3 receptor channels delays these signals, whereas expression of lysosomal TPC2 channels accelerates them. High-resolution Ca2+ imaging reveals elementary events upon TPC2 opening and signals coupled to …

Respiratory syncytial virus (RSV) is a virus that causes outbreaks of acute lower respiratory tract infections (LRTI) worldwide. The infections are associated with high rates of hospitalizations, which may lead to death, especially in older adults and young children. Due to the morbidity and mortality associated with RSV, pharmaceutical companies have dedicated research to create a vaccine since the 1960s. Despite setbacks after the failure of the first RSV vaccine, new developments in research have led to the approval of two new vaccines. Ensuring healthcare professionals and patients are well-informed about these vaccines is crucial for facilitating informed, collaborative clinical …

Hydrocarbon releases might result in serious consequences in various aspects. In addition to the contribution to environmental pollution, repetitive leakages need high repair costs. This study aim is to minimize potential repetitive leakage for other typical 3-phase piping systems. We conducted the risk assessment by adopting Risk Based Inspection (RBI) API 581 to identify risk level, calculating piping lifetime, recommended inspection plan and mitigations. The most relevant root causes can be obtained through quantitative Fault Tree Analysis (FTA). Observation and investigation was taken from eight 3-phase piping systems that experienced repetitive leakages. It has been found that the risk level …
