Insulin-Like Growth Factor Ii Acts Through An Endogenous Growth Pathway Regulated By Imprinting In Early Mouse Embryos, Daniel A. Rappolee, Karin S. Sturm, Ole Behrendtsen, Gilbert A. Schultz, Roger A. Pedersen, Zena Werb

Department of Obstetrics and Gynecology Research Publications

We present evidence that insulin-like growth factor II (IGF-II) mediates growth in early mouse embryos and forms a pathway in which imprinted genes influence development during preimplantation stages, mRNA and protein for IGF-II were expressed in preimplantation mouse embryos, but the related factors IGF-I and insulin were not. IGF-I and insulin receptors and the IGF-II/mannose-6-phosphate receptor were expressed. Exogenous IGF-II or IGF-I increased the cell number in cultured blastocysts, but a mutant form of IGF-II that strongly binds only the IGF-II receptor did not. Reduction of IGF-II expression by antisense IGF-II oligonucleotides decreased the rate of progression to the blastocyst …

Platelets And Leucocyte Counts In Pregnancy, Saadiya Aziz Karim, Javaid H. Rizvi, Iffat Rizwana, Mohammad Khurshid, Sadiqua N. Jafarey

Department of Obstetrics & Gynaecology

A study involving the obstetric clinic population of 3 major teaching hospitals of Karachi has been conducted to define changes in leucocyte and platelet counts as pregnancy progresses. 573 “normal” pregnant women - 183 in the first trimester, 195 in the second trimester and 194 in the third trimester were included in this analysis. We have found leucocytosis to be a feature of normal pregnancy; the change is subsequent to a progressive increase in granulocytes. Platelet counts were found to decrease slightly as pregnancy progresses

Role Of Thymectomy In Myasthenia Gravis, Sohaila Mohsin Ali, Aziz Sonawalla, Irfan Altafullah, Hizbullah Sheikh

Section of Urology

Thirteen patients with myasthenia gravis underwent total thymectomy between January, 1988 and December, 1991. The duration of symptoms prior to surgery varied from 2 months to 20 years. In a follow-up ranging from 2 months to 4 years, 11 patients showed a significant improvement with either complete discontinuation of medication of a marked reduction in doses. One patient with a small benign thymoma showed some improvement but subsequently required stepping up of anticholinesterase medication and addition of steroids and immune suppressants; another patient with atrophic thymic tissue had complete remission after thymectomy but developed myasthenic symptoms six months later requiring …

The Use Of Self Blood Glucose Monitoring And Its Relationship To Effective Management For People With Insulin Dependent Diabetes Mellitus, Maureen Unsworth Jan 1992

The Use Of Self Blood Glucose Monitoring And Its Relationship To Effective Management For People With Insulin Dependent Diabetes Mellitus, Maureen Unsworth

Theses : Honours

It is believed that people who effectively self manage their own diabetes will achieve higher levels of health and well being (Pender, 1987). Although diabetes education has increased rapidly in the last 10 years, the emphasis has been placed on increasing people's knowledge of diabetes as a medical condition, which does not necessarily ensure effective self management of diabetes. Previous research suggests that self blood glucose monitoring (SBOM) is an effective tool for promoting the use of blood glucose levels as a basis for initiating behaviour changes related to eating, exercise and insulin dosage. Yet, while most people with diabetes …

Heterogeneous Macromolecular Composition Of Renal Basement Membranes In Health, Diabetes And Glomerulonephritis, Douglas K. Brees Jan 1992

Heterogeneous Macromolecular Composition Of Renal Basement Membranes In Health, Diabetes And Glomerulonephritis, Douglas K. Brees

MUSC Theses and Dissertations

Although basement membranes are ubiquitous structures throughout the body, basement membranes have distinct compositions that are specific to their location. This basement membrane heterogeneity may, in part, reflect functional differences among various basement membranes. We examined basement membrane heterogeneity in normal, healthy mouse kidneys to assess the similarities and differences between glomerular and tubular basement membrane composition. It was demonstrated that mouse glomerular and tubular basement membrane share similar compositions but differ with respect to specific amounts of some components. In diabetes mellitus and passive Heymann nephritis (PHN), damage to the glomerular barrier occurs and is accompanied by an increase …

The Measurement Of Knowledge, Attitudes And Knowledge-Seeking Behaviours Of Women In Relation To The Menopause And Health Related Issues, Amanda Devine

Theses : Honours

Research suggests that women are not well informed about the health related issues which are associated with the menopause. The purpose of this study was to determine women's: current knowledge of the menopause and of the disease osteoporosis; attitudes to estrogen replacement therapy (ERT); frequency of menopausal symptoms; knowledge-seeking behaviours In relation to the menopause; and, open-ended responses to menopausal issues. This study surveyed 70 women who responded to an advertisement in Community Newspapers. Data were collected using a "Knowledge and attitudes to the menopause and health related issues" (KAMHRI) questionnaire. The KAMHRI questionnaire was developed by the researcher and …

Altered Expression Of Adenovirus 12 Dna-Binding Protein But Not Dna Polymerase During Abortive Infection Of Hamster Cells, Lynne A. Lucher, Benjawan Khuntirat, Jiansheng Zhao, Peter C. Angeletti Jan 1992

Altered Expression Of Adenovirus 12 Dna-Binding Protein But Not Dna Polymerase During Abortive Infection Of Hamster Cells, Lynne A. Lucher, Benjawan Khuntirat, Jiansheng Zhao, Peter C. Angeletti

Nebraska Center for Virology: Faculty Publications

Replication of human adenovirus type 12 DNA is blocked in abortively infected baby hamster kidney cells. The activity and accumulation of adenovirus 12 DNA polymerase is equivalent in infected hamster and human cell extracts. However, the accumulation of adenovirus type 12 DNA-binding protein is approximately 120-fold lower in extracts from infected hamster cells when compared to infected permissive human cells. This difference in accumulation is not because of replication of viral DNA during productive infection, since this difference is observed in the presence of hydroxyurea. The DNA-binding protein from infected hamster cells retains the ability to bind denatured DNA-cellulose. An …

Prevalence Of Iron Deficiency Anaemia In Children Of The Urban Slums Of Karachi, Ayesha Molla, Mohammad Khurshid, A. Majid Molla Jan 1992

Prevalence Of Iron Deficiency Anaemia In Children Of The Urban Slums Of Karachi, Ayesha Molla, Mohammad Khurshid, A. Majid Molla

Department of Pathology and Laboratory Medicine

A preliminary baseline survey was conducted to estimate the prevalence of anaemia in a group of 391 children aged 6-60 months, randomly selected from three urban slums of Karachi. Haemoglobin and the red cell indices including haematocrit, MCV, MCH, MCHC, ABC and red cell distribution width (RDW) were estimated for each of the selected children. Ferritin estimation was done on 354 (9 1%) children to assess the iron storage status. According to WHO criteria, the accepted cut-off point for anaemia screening in children is set at 11 gm/dl, 70 fI and 20 pg for haemoglobin, MCV and MCH respectively. Following …

Radiation Induced Tumor Lysis Syndrome In Patients With Leukemia, Imtiaz A. Malik, Paul Vellozo, Mohammad Khurshid, M. Ata Khan Jan 1992

Radiation Induced Tumor Lysis Syndrome In Patients With Leukemia, Imtiaz A. Malik, Paul Vellozo, Mohammad Khurshid, M. Ata Khan

Department of Medicine

Tumour lysis syndrome is a catastrophic complicalion of treatment of certain neoplasms. It is usually seen with the tumours that have high growth fractions, increased bulk and extreme sensitivity to cytotoxic therapy. Most commonly it occurs in association with hematologic malignancies such as lymphomas and leukaemias. Rarely it has been observed with solid tumours like small cell lung cancer, seminoma and carcinoma of the breast. The syndrome develops due to massive cell lysis occurring within a few hours to a few days of starting the cytotoxic therapy. It is characterized by hyperuricemia, hyperkalemia, hyperphosphatemia and hypocalcaemia. Acute renal failure, cardiac …

Supervised Training Of Practical Procedures In The Internal Medicine Residency, Z Abbas, S Abid, J A. Khan, A Jabbar

Section of Gastroenterology

In order to get feedback whether the medical residents are getting adequate supervised training to perform practical procedures, a questionnaire was distributed among the residents asking their involvement in sixteen frequently performed procedures in the medical unit of the Aga Khan University Medical Centre. Though the residents were satisfied about their training in doing lumbar punctures, pleural and peritoneal aspirations, bone marrow aspiration and trephine and chest tubes placement, they showed their concern about training in putting central lines, venous cut downs, cardiac pacing and biopsies. A better schedule training programme, organized subspecialty rotation, continuous evaluation of resident's skill, credentialing …

Regulation Of Transforming Growth Factor-Beta 1 Gene Expression By Glucocorticoids In Normal Human T Lymphocytes., O. Ayanlarbatuman, A. P. Ferrero, Arturo Diaz, Sergio A. Jimenez Nov 1991

Regulation Of Transforming Growth Factor-Beta 1 Gene Expression By Glucocorticoids In Normal Human T Lymphocytes., O. Ayanlarbatuman, A. P. Ferrero, Arturo Diaz, Sergio A. Jimenez

Department of Medicine Faculty Papers

Glucocorticoids (GC) modulate immune function in a number of ways, including suppression of T cell proliferation and other IL-2-mediated T cell functions. These inhibitory effects are similar to those induced by transforming growth factor-beta 1 (TGF-beta 1), a cytokine with potent T cell inhibiting activities. We examined the hypothesis that GC effects may be at least partially achieved through modulation of the expression of the TGF-beta 1 gene in activated T cells. Normal T cells were cultured with or without purified phytohemagglutinin (PHA-p) and 4 beta-phorbol 12-myristate 13-acetate (PMA) in the presence or absence of the synthetic GC, dexamethasone (100-200 …

Dartmouth Scholarship

Fc gamma Rs (Fc gamma RI, Fc gamma RII, and Fc gamma RIII) are highly expressed on human mononuclear phagocytes and function in the clearance of immune complexes and opsonized pathogens. We have examined the role of Fc gamma R in mediating antibody-dependent clearance of HIV-1 by human monocytes and monocyte-derived macrophages by using bispecific antibodies (BsAbs) to independently target the virus to Fc gamma RI, Fc gamma RII, or Fc gamma RIII. Virus production was markedly reduced in monocytes cultured with strain HIV-1IIIB opsonized with BsAbs that target the virus to either Fc gamma RI or Fc gamma RII …

Section of Gastroenterology

The clinical features, course and histology of liver in 20 patients; mostly middle aged to elderly females, closely resembling chronic Non A Non B hepatitis is presented. They presented quite late in their disease and therefore, complications such as variceal bleeds, ascites and encephalopathy were frequent. Our patients were negative for hepatitis B and C virus serology. Metabolic and immune causes of chronic liver disease were also ruled out. To the best of our knowledge, this is the first study of its kind elaborating the clinical features, course and histology of liver in chronic Non B Non C hepatitis and …

The Role Of Gaba In Reproductive Neuroendocrine Systems, Robert Carter Mcree Oct 1991

The Role Of Gaba In Reproductive Neuroendocrine Systems, Robert Carter Mcree

Theses and Dissertations in Biomedical Sciences

Local gamma-amino butyric acid (GABA) neurons in the hypothalamus can modulate the luteinizing hormone releasing hormone (LHRH) pulse generating system.

Two animal models (intact and ovariectomized rats) were used to determine the nature of modulation of LHRH release by GABA. The experiment determined the release of LHRH, serotonin (5-HT), and 5-hydroxyindole acetic acid (5-HIAA) during two hormonal states with each model. In the intact rat the proestrus and estrus states were examined, and in the ovariectomized rat in-vitro release was determined with (OVX2) and without (OVX) estradiol treatment.

In-vivo experiments studied the effect of stimulation of GABA receptors …
